    Response of selected microbial strains and their consortia to the presence of automobile paints: Biofilm growth, matrix protein content and hydrolytic enzyme activity

    The goal of the current study was to examine the effects of pollutants (White color – CP; Metallic red color – FM; Thinner – CN; Thinner for rinsing paint – MF; Basic color (primer) – FH) originating from the automotive industry on the biofilm growth, matrix protein content, and activity of the hydrolytic enzymes of selected microbial strains in laboratory conditions that mimic the bioreactor conditions. The chosen microorganisms (bacteria, yeasts, and fungi) were isolated from automotive industry wastewater. Pure microbe cultures and their consortia were injected into AMB Media carriers and developed into biofilms. The use of AMB media carriers has been linked to an increase in the active surface area colonized by microorganisms. Afterwards, the carriers were transferred to Erlenmeyer flasks with nutrient media and pollutants at a concentration of 200 μL/mL. The current study found that, depending on the microbial strain, development phase, and chemical structure, the assessed pollutants had an inhibitory or stimulatory influence on the growth of single cultures and their consortia. Statistical analysis found positive correlations between the protein content in the matrix and the biofilm biomass of Rhodotorula mucilaginosa and consortia in CP and FH media, respectively. The proteolytic activity of Candida utilis was very pronounced in media with MF and CN. The best alkaline phosphatase activity (ALP) was achieved in the CN medium of R. mucilaginosa. Acid invertase activity was the highest in the FM and CP media of Escherichia coli and consortia, respectively, whereas the highest alkaline invertase activity was measured in the MF medium of E. coli. A positive correlation was confirmed between ALP and the biofilm biomass of R. mucilaginosa in CP and CN media, as well as between ALP and the biofilm biomass of Penicillium expansum in FM medium. The findings provide novel insights into the extracellular hydrolytic activity of the investigated microbial strains in the presence of auto paints, as well as a good platform for subsequent research into comprehensive biofilm profiling using modern methodologies

It is important to establish a connection between CAD design of products and complex limitations of assembly operations in CAM so that the design process is provided with the conditions for development and modification in a virtual environment before the beginning of production. This integration can be seen on the example of tank waggons.     COMPARISON OF TWO DIFFERENT METHODS (PATIENT QUESTIONNAIRE AND MEDICATION POSSESSION RATIO - MPR) FOR MEASURING THE CHRONIC PATIENT’S BEHAVIOR

    Background: Medication adherence is the extent to which patients take medications as prescribed by their health care providers. There are a number of approaches to study medication-taking behavior. The aim was to compare two most common methods for measuring adherence: Patient Adherence Questionnaire and Medication Possession Ratio (MPR). They belong to the indirect methods. Methods: In this article four adherence studies were analysed and the results were compared, two wherein the patient questionnaire was applied and other two with medication possession ratio applied. Results: The obtained results reveal that more than half of respondents (58.9%) experienced constant nonadherence behavior according to the prescribed therapy. The main reason of nonadherence is oblivion, suggesting that it is necessary to pay more attention to this problem. Conclusions: Nonadherence with therapy has negative consequences on the health of the individual, and an adverse impact on the community health and wealth. Patients should be informed of the importance of regularly taking prescribed therapy. The main problem of long-term therapy is significantly decreased of adherence to medication in a very short time. It is important to stress that almost all the interventions effective for improving patient adherence in long-term care are complex and should be repeated after a while

    Effects of Combined Allogenic Adipose Stem Cells and Hyperbaric Oxygenation Treatment on Pathogenesis of Osteoarthritis in Knee Joint Induced by Monoiodoacetate

    The beneficial effects of HBO in inflammatory processes make it an attractive type of treatment for chronic arthritis. In addition, the effects of combination therapy based on adipose stem cells and HBO on OA progression have not been fully investigated. The current study explored the efficacy of intra-articular injection of allogeneic adipose-derived mesenchymal stem cells (ADMSCs) combined with hyperbaric oxygenation treatment (HBO) in a rat osteoarthritis (OA) model. The rat OA model was induced by intra-articular injection of monoiodoacetate (MIA) and 7 days after application of MIA rats were divided into five groups: healthy control (CTRL), osteoarthritis (OA), ADMSCs (ADS), the HBO+ADS21day and HBO+ADS28day groups. A single dose of 1 × 106 allogeneic ADMSCs suspended in sterile saline was injected into the knee joint alone or in combination with HBO treatment. Rats were sacrificed at 3 or 4 weeks after MIA injection. Treatment outcomes were evaluated by radiographic, morphological and histological analysis and by specific staining of articular cartilage. We also measured the level of inflammatory and pro/antioxidative markers. We confirmed that combined treatment of ADMSCs and HBO significantly improved the regeneration of cartilage in the knee joint. Rtg score of knee joint damage was significantly decreased in the HBO+ADS21day and HBO+ADS28day groups compared to the OA. However, the positive effect in the HBO+ADS28day group was greater than the HBO+ADS21day group. The articular cartilage was relatively normal in the HBO+ADS28day group, but moderate degeneration was observed in the HBO+ADS21day compared to the OA group. These findings are in line with the histopathological results. A significantly lower level of O2−. was observed in the HBO+ADS28day group but a higher NO level compared to the HBO+ADS21day group. Moreover, in the HBO+ADS28day group significantly higher concentrations of IL-10 were observed but there was no significant difference in proinflammatory cytokine in serum samples. These results indicate that a single intra-articular injection of allogeneic ADMSCs combined with HBO efficiently attenuated OA progression after 28 days with greater therapeutic effect compared to alone ADMSCs or after 3 weeks of combined treatment. Combined treatment might be an effective treatment for OA in humans

    A bibliometric analysis of the top 100 most-cited case reports and case series in Endodontic journals

    Aim To identify the top 100 most-cited case reports and case series published in Endodontic journals and to analyse their bibliometric characteristics. Methodology The Clarivate Analytics’ Web of Science (WoS), Scopus and PubMed databases were used to identify the top 100 most-cited case reports and case series in Endodontic journals. Complete bibliographic records of the selected case reports and case series were exported in plain text or BibTeX format and imported into the R environment for statistical computing and graphics. The following parameters were then analysed: names and affiliations of the authors, title, year of publication, journal of publication, first author, corresponding author, literature cited within reports, language, citation counts, impact factor of the journal, keywords, Keywords Plus and research topic. Results In total, 88 case reports and 12 case series published in English between 1977 and 2016 were identified as the most-cited reports in the field of Endodontics. The terms “case report(s)” or “case series” were not included in the title of 57 articles. The number of authors per report ranged from one to seven, with the average number of co-authors per report being 3.14. The most-cited author was M Trope (University of Pennsylvania, USA). The University of Washington and Private Practice, Cetraro, Italy, were the most productive institutions. The country whose case reports received the largest total number of citations was the United States. The largest number of the most-cited reports appeared in 2002, 2004 and 2007 (n = 7, respectively). According to the WoS database, the total number of citations ranged from 42 to 453, with the average number of citations per report being 79.97. The majority of the top 100 most-cited articles were published in the Journal of Endodontics and the International Endodontic Journal. The most frequently used author keywords were revascularization and mineral trioxide aggregate. The majority of the case reports and case series dealt with topics related to pulp regeneration. Conclusion This bibliometric study provides a comprehensive overview on the progress, trends and current directions in clinical practice within the field of Endodontics

    Reporting quality of scoping reviews in endodontics: A meta‐research study

    Objectives: To evaluate the reporting quality of Scoping Reviews (ScRs) in endodontics according to the PRISMA Extension Checklist for Scoping Reviews (PRISMA‐ScR) and to analyse their association with a range of publication and methodological/reporting characteristics. Methods: Pubmed, Scopus, and Web of Science databases were searched up to 31 January 2024 to identify scoping reviews in the field of endodontics. An additional search was performed in three leading endodontic journals. Study selection and appraising the quality of the studies was carried out independently by two reviewers. Each of the 20 PRISMA‐ScR items were allocated a score of either 0, 0.5 or 1 to reflect the completeness of the reporting. An item‐specific and overall percentage reporting quality score was calculated and reported through descriptive statistics across a range of publication, as well as methodological/reporting characteristics. A univariable and multivariable quantile regression was performed to identify the effect of publication and methodological/reporting characteristics (year of publication, journal, inclusion of an appropriate reporting guideline, and study registration) on the overall percentage reporting quality score. Association of reporting quality score with publication characteristics was then investigated. Results: A total of 40 ScRs were identified and included for appraisal. Most of the studies were published from 2021 onwards. The overall median reporting quality score was 86%. The most frequent items not included in the studies were: a priori protocol registration (22/40 compliant; 55%), and reporting of funding (16/40 compliant; 40%). Other key elements that were inadequately reported were the abstract (7/40 compliant; 18%), the rationale and justification of the ScR (21/40 compliant; 52%) and the objectives of the study (18/40 compliant; 45%). Studies that adhered to appropriate reporting guidelines were associated with greater reporting quality scores (β‐coefficient: 10; 95%CI: 1.1, 18.9; p = .03). ScRs with protocols registered a priori had significantly greater reporting quality scores (β‐coefficient: 12.5; 95%CI: 6.1, 18.9; p < .001), compared with non‐registered reviews. Conclusions: The reporting quality of the ScRs in endodontics varied and was greater when the ScR protocols were registered a priori and when the authors adhered to reporting guidelines

    Phytochemical Analysis, Antioxidant, Antimicrobial, and Cytotoxic Activity of Different Extracts of Xanthoparmelia stenophylla Lichen from Stara Planina, Serbia

    The aim of this study was to identify some of the secondary metabolites present in acetonic, methanolic, and hexanic extracts of lichen Xanthoparmelia stenophylla and to examine their antioxidant, antimicrobial, and cytotoxic activity. Compounds of the depsid structure of lecanoric acid, obtusic acid, and atranorin as well as usnic acid with a dibenzofuran structure were identified in the extracts by HPLC. The acetone extract was shown to have the highest total phenolic (167.03 ± 1.12 mg GAE/g) and total flavonoid content (178.84 ± 0.93 mg QE/g) as well as the best antioxidant activity (DPPH IC50 = 81.22 ± 0.54). However, the antimicrobial and antibiofilm tests showed the best activity of hexanic extract, especially against strains of B. cereus, B. subtilis, and S. aureus (MIC < 0.08, and 0.3125 mg/mL, respectively). Additionally, by using the MTT method, the acetonic extract was reported to exhibit a strong cytotoxic effect on the HeLa and HCT-116 cell lines, especially after 72 h (IC50 = 21.17 ± 1.85 and IC50 = 21.48 ± 3.55, respectively). The promising antioxidant, antimicrobial, and cytotoxic effects of Xanthoparmelia stenophylla extracts shown in the current study should be further investigated in vivo and under clinical conditions
