    Even stressed cells are individuals: second messengers of free radicals in pathophysiology of cancer

    Abstract Pathophysiological processes associated with disturbances in cell and tissue oxidative homeostasis, are associated with self-catalyzed process of lipid peroxidation. The end products of lipid peroxidation are reactive aldehydes such as 4-hydroxy-2-nonenal (HNE), acting as ā€œsecond messengers of free radicals.ā€ Although reactive aldehydes were first recognized only as cytotoxic, new evidence has come to light, related to their cell growth regulatory functions achieved through cell signaling. The variable appearance of HNE in several organs indicates that its mode of action might be related to an individual cell stress adaptation. The underlying mechanism could be that specific mutations and epigenetic changes on one hand interfere with hormesis on the other. The precise role of oxidative stress and lipid peroxidation in these processes still needs more clarification at molecular level. Finally, an individual approach to each patient, based on the individual cell response to stress, opens a new possibility of integrative medicine in cancer treatment and strongly supports modern concepts of personalized medicine

    Aflatoxin, fumonisin, ochratoxin, zearalenone and deoxynivalenol biomarkers in human biological fluids:A systematic literature review, 2001-2018

    Human exposure to mycotoxins occurs mostly through dietary intake, although exposure through dermal and inhalation routes has also been shown. Depending on the type of mycotoxins, the applied dose and duration of exposure, a particular toxin can cause either chronic or acute illnesses such as kidney failure and cancer. Thus, understanding the biotransformation of mycotoxins and identification of reliable biomarkers in the human body is important for accurate risk assessment of mycotoxin exposure. This review provides a comprehensive overview of worldwide aflatoxins, fumonisins, ochratoxin, zearalenone and deoxynivalenol mycotoxin biomonitoring studies reported in the last 18 years. The studies performed in Africa, Europe, Asia and America are based on the measurement of a limited number of mycotoxin biomarkers and do not provide a comprehensive risk assessment of the mycotoxin exposure. Although the findings represent a small segment of a much larger health risk of mycotoxins exposure, it is acknowledged that a multianalyte approach covering bioconjugated and other metabolites of most often occurring mycotoxins would better reflect the extent of the global exposure problems with these highly toxic compounds.</p

    Short Overview of Some Assays for the Measurement of Antioxidant Activity of Natural Products and Their Relevance in Dermatology

    Impaired systemic redox homeostasis is implicated in the onset and development of various diseases, including skin diseases. Therefore, continuous search for natural products with antiox-idant bioactivities applicable in biomedicine is attractive topic of general interest. Research ef-forts aiming to validate antioxidant potentials of natural products has led to the development of several assays based on various test principles. Hence, understanding the advantages and limita-tions of various assays is important for selection of assays useful to study antioxidant and relat-ed bioactivities of natural products of biomedical interest. This review paper gives a short over- view on some chemical and cellular bioassays used to estimate the antioxidant activity of chosen natural products together with a brief overview on the use of natural products with antioxidant activities as adjuvant medicinal remedies in dermatology

    The NRF2, Thioredoxin, and Glutathione System in Tumorigenesis and Anticancer Therapies

    Cancer remains an elusive, highly complex disease and a global burden. Constant change by acquired mutations and metabolic reprogramming contribute to the high inter- and intratumor heterogeneity of malignant cells, their selective growth advantage, and their resistance to anticancer therapies. In the modern era of integrative biomedicine, realizing that a personalized approach could benefit therapy treatments and patientsā€™ prognosis, we should focus on cancer-driving advantageous modifications. Namely, reactive oxygen species (ROS), known to act as regulators of cellular metabolism and growth, exhibit both negative and positive activities, as do antioxidants with potential anticancer effects. Such complexity of oxidative homeostasis is sometimes overseen in the case of studies evaluating the effects of potential anticancer antioxidants. While cancer cells often produce more ROS due to their increased growth-favoring demands, numerous conventional anticancer therapies exploit this feature to ensure selective cancer cell death triggered by excessive ROS levels, also causing serious side effects. The activation of the cellular NRF2 (nuclear factor erythroid 2 like 2) pathway and induction of cytoprotective genes accompanies an increase in ROS levels. A plethora of specific targets, including those involved in thioredoxin (TRX) and glutathione (GSH) systems, are activated by NRF2. In this paper, we briefly review preclinical research findings on the interrelated roles of the NRF2 pathway and TRX and GSH systems, with focus given to clinical findings and their relevance in carcinogenesis and anticancer treatments

    Dietary exposure of the Qatari population to food mycotoxins and reflections on the regulation limits

    The article estimates the exposure of the population of Qatar to aflatoxins, fumonisins and ochratoxins, measured in food samples collected from the markets of Doha. The mycotoxin extractions were performed using the ā€œQuick Easy Cheap Effective Rugged Safeā€ Method (QuEChERS) and the extract measured with LCā€“MS/MS. High contamination with aflatoxins was detected in Nuts and Spices (e.g. 534.15ng/g and 371.6ng/g respectively). The estimated daily intake (EDI) level was estimated using statistical data on average Qatari population. Additionally, the probability to exceed TDI was calculated for fumonisins and ochratoxins. The results indicate high exposure to aflatoxins (with alarming values of margin of exposure, i.e. MoE). The article points to the necessity of a regular assessment and reevaluation of the concentration limits allowed in food products accounting for statistics on the population (i.e. food consumption), economic growth of the country, and monitoring of mycotoxin contamination of food as much as technological, financial and human resources of a country allows
