Quercetin 7-rhamnoside reduces porcine epidemic diarrhea virus replication via independent pathway of viral induced reactive oxygen species
<p>Abstract</p> <p>Background</p> <p>On the base of our previous study we were observed relevant studies on the hypothesis that the antiviral activity of quercetin 7-rhamnoside (Q7R), a flavonoid, won't relate ability of its antioxidant.</p> <p>Methods</p> <p>We were investigated the effects of Q7R on the cytopathic effects (CPE) by CPE reduction assay. Production of DNA fragment and reactive oxygen species (ROS) induced by PEDV infection were studied using DNA fragmentation assay and flow cytometry.</p> <p>Results</p> <p>In the course of this study it was discovered that Q7R is an extremely potent compound against PEDV. The addition of Q7R to PEDV-infected Vero cells directly reduced the formation of a visible cytopathic effect (CPE). Also, Q7R did not induce DNA fragmentation. Furthermore, ROS increased the infection of PEDV, which was strongly decreased by N-acetyl-L-cysteins (NAC). However, the increased ROS was not decreased by Q7R. Antiviral activity of antioxidants such as NAC, pyrrolidine dithiocarbamate (PDTC), and the vitamin E derivative, trolox, were hardly noticed.</p> <p>Conclusions</p> <p>We concluded that the inhibition of PEDV production by Q7R is not simply due to a general action as an antioxidants and is highly specific, as several other antioxidants (NAC, PDTC, trolox) are inactive against PEDV infection.</p
Density functional calculations of the electronic structure and magnetic properties of the hydrocarbon K3picene superconductor near the metal-insulator transition
We have investigated the electronic structures and magnetic properties of of
K3picene, which is a first hydrocarbon superconductor with high transition
temperature T_c=18K. We have shown that the metal-insulator transition (MIT) is
driven in K3picene by 5% volume enhancement with a formation of local magnetic
moment. Active bands for superconductivity near the Fermi level E_F are found
to have hybridized character of LUMO and LUMO+1 picene molecular orbitals.
Fermi surfaces of K3picene manifest neither prominent nesting feature nor
marked two-dimensional behavior. By estimating the ratio of the Coulomb
interaction U and the band width W of the active bands near E_F, U/W, we have
demonstrated that K3picene is located in the vicinity of the Mott transition.Comment: 5 pages, 5 figure
KINEMATIC SYNTHESIS OF A MEDICAL BED FOR DECUBITUS ULCER PATIENTS
ABSTRACT A decubitus ulcer or bedsore is a pressure-induced ulceration of the skin occurring in persons confined to bed for long periods of time. Reduction of pressure over bony prominences is of primary importance to prevent and cure bedsores. For this purpose, specially designed mattresses can be used and/or the patient should be turned frequently to avoid ischemia of soft tissue. In addition to pressure, other principal factors causing bedsore are friction and shear forces. In this paper, we designed a new 5 degree of freedom bed mechanism that can be used to change the posture of pressure ulcer patients, which generates 7 motions including backrest elevation, kneerest elevation, lounge position, left and right rotation, trendelenberg and reverse trendelenberg motion, and straight elevation. Particularly, we focused on the synthesis of a backrest and seatrest assembly that can reduce sliding between the bed and the patient
Development of a Method to Predict the Bending Strength of Lumber Without Regard to Species Using X-Ray Images
Several models have been developed for predicting bending strength of lumber using X-rays, but most require species-specific classifications. However, the classification is very difficult because logs or cants can arrive without leaves or bark. This study was carried out to develop an alternative bending strength prediction model that does not lose precision when the species is unknown. The study proposes an Equivalent Density Model (EDM), in which a cross-section is quantified as equivalent density. Because the relationship between density and strength of small clear specimens is not affected by species, the EDM was expected to correlate to strength regardless of species. This model predicted the modulus of rupture in two species with R2 = 0.73, although the two were mixed. Therefore, it may be possible to predict bending strength using X-rays without classifying lumber by species
A Compact Vertical Scanner for Atomic Force Microscopes
A compact vertical scanner for an atomic force microscope (AFM) is developed. The vertical scanner is designed to have no interference with the optical microscope for viewing the cantilever. The theoretical stiffness and resonance of the scanner are derived and verified via finite element analysis. An optimal design process that maximizes the resonance frequency is performed. To evaluate the scanner’s performance, experiments are performed to evaluate the travel range, resonance frequency, and feedback noise level. The role of Qa-2, the functional homolog of HLA-G, in a Behcet's disease-like mouse model induced by the herpes virus simplex
<p>Abstract</p> <p>Background</p> <p>It has been suggested that the HLA-G molecule is a genetic risk factor for Behcet's disease (BD). In this study, we evaluated the level of Qa-2, a murine nonclassical class I MHC molecule and possible functional homolog of HLA-G, to determine if it was associated with various symptoms of BD-like mice. In addition, we investigated siRNA (small interfering RNA) treatment to determine if it inhibited Qa-2 expression, thereby changing the symptoms of mice.</p> <p>Methods</p> <p>RNA interference (RNAi) and vector transfection were employed to manipulate gene expression <it>in vivo </it>in mice. siRNA (small interfering RNA) or Qa-2 expression vector was applied to inhibit or up-regulate Qa-2 expression, respectively.</p> <p>Results</p> <p>The Qa-2 levels in granulocytes were lower in BD-like mice than in normal controls. The silencing of Qa-2 by intravenous injection of siRNA (500 nmol/mouse, 4 times at 3-day intervals) specifically reduced the Qa-2 levels and worsened the BD-like symptoms.</p> <p>Conclusions</p> <p>Silencing Qa-2 by injecting siRNA into mice resulted in deterioration of symptoms in BD-like mice.</p
Validation and Reliability of a Smartphone Application for the International Prostate Symptom Score Questionnaire: A Randomized Repeated Measures Crossover Study
Background: Smartphone-based assessment may be a useful diagnostic and monitoring tool for patients. There have been many attempts to create a smartphone diagnostic tool for clinical use in various medical fields but few have demonstrated scientific validity. Objective: The purpose of this study was to develop a smartphone application of the International Prostate Symptom Score (IPSS) and to demonstrate its validity and reliability. Methods: From June 2012 to May 2013, a total of 1581 male participants (>= 40 years old), with or without lower urinary tract symptoms (LUTS), visited our urology clinic via the health improvement center at Soonchunhyang University Hospital (Republic of Korea) and were enrolled in this study. A randomized repeated measures crossover design was employed using a smartphone application of the IPSS and the conventional paper form of the IPSS. Paired t test under a hypothesis of non-inferior trial was conducted. For the reliability test, the intraclass correlation coefficient (ICC) was measured. Results: The total score of the IPSS (P=.289) and each item of the IPSS (P=.157-1.000) showed no differences between the paper version and the smartphone version of the IPSS. The mild, moderate, and severe LUTS groups showed no differences between the two versions of the IPSS. A significant correlation was noted in the total group (ICC=.935, P<.001). The mild, moderate, and severe LUTS groups also showed significant correlations (ICC=.616,.549, and .548 respectively, all P<.001). There was selection bias in this study, as only participants who had smartphones could participate. Conclusions: The validity and reliability of the smartphone application version were comparable to the conventional paper version of the IPSS. Sensory-to-Motor Overflow: Cooling Foot Soles Impedes Squat Jump Performance
Partial funding for Open Access provided by the UMD Libraries' Open Access Publishing Fund.Evidence from recent studies on animals and humans suggest that neural overflow from the primary sensory cortex (S1) to the primary motor cortex (M1) may play a critical role in motor control. However, it is unclear if whole-body maximal motor tasks are also governed by this mechanism. Maximum vertical squat jumps were performed by 15 young adults before cooling, then immediately following a 15-min cooling period using an ice-water bath for the foot soles, and finally immediately following a 15-min period of natural recovery from cooling. Jump heights were, on average, 3.1 cm lower immediately following cooling compared to before cooling (p = 3.39 × 10−8) and 1.9 cm lower following natural recovery from cooling (p = 0.00124). The average vertical ground reaction force (vGRF) was also lower by 78.2 N in the condition immediately following cooling compared to before cooling (p = 8.1 × 10−5) and 56.7N lower following natural recovery from cooling (p = 0.0043). Decreased DBC1 Expression Is Associated With Poor Prognosis in Patients With Non-Muscle-Invasive Bladder Cancer
Purpose The deleted in bladder cancer 1 (DBC1) gene is located within chromosome 9 (9q32-33), a chromosomal region that frequently shows loss of heterozygosity in bladder cancer (BC). It is suspected that it acts as a tumor suppressor gene, but its prognostic value remains unclear. The aim of the present study was to investigate the value of DBC1 as a prognostic marker in BC. Materials and Methods The expression of DBC1 was determined by real-time polymerase chain reaction analysis in 344 patients with BC (220 non-muscle-invasive BC [NMIBC] and 124 muscle-invasive BC [MIBC]) and in 34 patients with normal bladder mucosa. The results were compared with clinicopathologic parameters, and the prognostic value of DBC1 was evaluated by Kaplan-Meier analysis and a multivariate Cox regression model. Results: DBC1 expression was significantly decreased in patients with MIBC compared with those diagnosed with NMIBC (p=0.010). Patients with aggressive tumor characteristics had lower DBC1 expression levels in NMIBC (each, p<0.05). By multivariate Cox regression analysis, low DBC1 expression was a predictor of progression to MIBC (hazard ratio, 7.104; p=0.013). Kaplan-Meier estimates revealed a significant difference in tumor recurrence, progression to MIBC, and cancer-specific survival depending on the level of DBC1 expression in NMIBC (log-rank test, each, p<0.05). Conclusions: The expression of DBC1 was associated with tumor aggressiveness, progression to MIBC, and survival in NMIBC.
