    Neuroimaging of Habit-based vs. Goal-directed behavior in Instrumental Learning

    Addiction has been proposed to result from an overreliance on the habit-based and goal-directed controllers of behavior; however, few data exist to simultaneously support both behavioral and neuoranatomical aspects of this theory in humans. Here, we clarify the locations of the homologous structures controlling behavior in the human brain to those studied in animal models. The study included two parts. 1) The first part established in a behavioral experiment that the devaluation video in the present paradigm was able to influence instrumental behavior. Using a 3-session instrumental learning task to examine behavior, we examined 78 participants, aged 18-35. A significant difference in the change in response rate immediately before and after devaluation was found between the 2 groups viewing worms in devaluation compared to the group not viewing worms. There was a significant difference in change in liking immediately before and after devaluation between the three conditions, as well as in the change in liking, hunger, and response rate between the paired and empty bowl unpaired conditions. There was a significant correlation between snack liking pre-session 3 and response rate in session 3, as well as between pre-extinction snack liking and response rate in the start of extinction. 2) The second part of the study used the same 3-session training paradigm over 3-days, with fMRI on the third day to measure neural activity during this same instrumental learning task. Although the results are preliminary (N=10), these show that the comparable regions of the human brain are involved in goal-directed and habit-based control of behavior, with a perfect negative Spearman correlation of mean vmPFC activity at the end of training and the change in responding from immediately before to immediately after devaluation. Three of the 10 subjects were addicted smokers, which is insufficient data to determine whether they were less sensitive to reward devaluation and whether they relied more heavily on brain structures associated with habit-based controllers of behavior. However, understanding the relationship between habit-based and goal-directed controllers of behavior and their role in addiction and clarifying the human brain structures responsible for these systems can lead to the development of therapies for addiction

    Non-universality of commonly used correlation-energy density functionals

    The correlation energies of the helium isoelectronic sequence and of Hooke's atom isoelectronic sequence have been evaluated using an assortment of local, gradient and meta-gradient density functionals. The results are compared with the exact correlation energies, showing that while several of the more recent density functionals reproduce the exact correlation energies of the helium isoelectronic sequence rather closely, none is satisfactory for Hooke's atom isoelectronic sequence. It is argued that the uniformly acceptable results for the helium sequence can be explained through simple scaling arguments that do not hold for Hooke's atom sequence, so that the latter system provides a more sensitive testing ground for approximate density functionals. This state of affairs calls for further effort towards formulating correlation-energy density functionals that would be truly universal at least for spherically-symmetric two-fermion systems.Comment: To appear in J.     Evidence for the impact of Reactive oxygen species on the control of branching in Neurospora crassa

    In preliminary screens, several functions, most notably, genes involved in the control of reactive oxygen species (ROS), were identified as playing a role in the process of growth rate compensation of branch density. The maintenance of branch density under growth at various temperatures was examined in a selection of mutants in genes known to be important in the control of ROS. In all ROS control mutants tested, growth was shown to branch tighter when grown at higher temperatures (which result in faster growth rates) and looser when grown at lower temperatures (which results in slower growth rates). This can be contrasted with wild-type Neurospora which branches at the same density under both conditions. We also tested the impact of environmental agents which lower the concentration of ROS on branching. In tests, various water soluble anti-oxidants (ie reducing agents); Ascorbic Acid (aka Vitamin C) and Glutathione produced unusual bra nching patterns in wild-type Neurospora strains exposed

    Urinary porphyrin excretion in hepatitis C infection

    A high prevalence of hepatitis C virus infection in porphyria cutanea tarda in some populations suggests a close link between viral hepatitis and alteration of porphyrin metabolism. Moreover, there is evidence of a role of porphyrinopathies in hepatocarcinogenesis. The aim of our study was to obtain data on the prevalence and patterns of heme metabolism alterations in patients with chronic hepatitis C virus infection. Urinary porphyrin excretion was prospectively studied in 100 consecutive outpatients with chronic hepatitis C infection without signs of photosensitivity, using an ion-pair high performance liquid chromatography method. Increased total porphyrin excretion was found in 41 patients, with predominant excretion of coproporphyrins (whole study group: mean 146 mu g/g creatinine, interquartile range 76-186; normal <150), in 10 patients excretion exceeded 300 mu g/g creatinine. In the majority of all patients studied (75/100) an increased ratio of the relatively hydrophobic coproporphyrin isomer I to isomer III was found. In just one case, urinary porphyrin pattern characteristic for chronic hepatic porphyria was present (uroporphyrin > coproporphyrin, heptacarboxyporphyrin III increased) but the total porphyrin excretion was only slightly elevated in this case. In the whole group, total urinary porphyrin excretion correlated well with serum bilirubin and was inversely correlated with albumin and thrombin time. In conclusion, secondary coproporphyrinuria occurs frequently in heptatitis C infection, whereas in Germany, preclinical porphyria cutanea tarda seems to be rare in these patients

    Determination of serum cortisol by isotope-dilution liquid-chromatography electrospray ionization tandem mass spectrometry with on-line extraction

    A liquid chromatographic-mass spectrometric method for the determination of cortisol in serum using atmospheric pressure electrospray ionization and tandem mass spectrometry is described. During sample preparation, 150 mul of serum were deproteinized with methanol/zinc sulfate followed by on-line solid phase extraction employing column switching. Tri-deuterated cortisol was used as the internal standard. The following transitions were monitored: cortisol, 363 > 309 m/z; d3-cortisol, 366 > 312 m/z. The total run-time was 5 minutes. The method proved linear (0-500 mug/l; r=0.999), precise (total coefficient of variation between 5.0% and 3.2% at a mean cortisol concentration of 15.1 mug/l and 269 mug/l, respectively; n=16) and specific with regard to relevant endogenous and exogenous steroids
