    Comparison of acoustic travel-time measurement of solar meridional circulation from SDO/HMI and SOHO/MDI

    Time-distance helioseismology is one of the primary tools for studying the solar meridional circulation. However, travel-time measurements of the subsurface meridional flow suffer from a variety of systematic errors, such as a center-to-limb variation and an offset due to the P-angle uncertainty of solar images. Here we apply the time-distance technique to contemporaneous medium-degree Dopplergrams produced by SOHO/MDI and SDO/HMI to obtain the travel-time difference caused by meridional circulation throughout the solar convection zone. The P-angle offset in MDI images is measured by cross-correlating MDI and HMI images. The travel-time measurements in the south-north and east-west directions are averaged over the same observation period for the two data sets and then compared to examine the consistency of MDI and HMI travel times after correcting the systematic errors. The offsets in the south-north travel-time difference from MDI data induced by the P-angle error gradually diminish with increasing travel distance. However, these offsets become noisy for travel distances corresponding to waves that reach the base of the convection zone. This suggests that a careful treatment of the P-angle problem is required when studying a deep meridional flow. After correcting the P-angle and the removal of the center-to-limb effect, the travel-time measurements from MDI and HMI are consistent within the error bars for meridional circulation covering the entire convection zone. The fluctuations observed in both data sets are highly correlated and thus indicate their solar origin rather than an instrumental origin. Although our results demonstrate that the ad hoc correction is capable of reducing the wide discrepancy in the travel-time measurements from MDI and HMI, we cannot exclude the possibility that there exist other systematic effects acting on the two data sets in the same way.Comment: accepted for publication in A&

    Solar meridional circulation from twenty-one years of SOHO/MDI and SDO/HMI observations: Helioseismic travel times and forward modeling in the ray approximation

    The south-north travel-time differences are measured by applying time-distance helioseismology to the MDI and HMI medium-degree Dopplergrams covering May 1996-April 2017. Our data analysis corrects for several sources of systematic effects: P-angle error, surface magnetic field effects, and center-to-limb variations. An interpretation of the travel-time measurements is obtained using a forward-modeling approach in the ray approximation. The travel-time differences are similar in the southern hemisphere for cycles 23 and 24. However, they differ in the northern hemisphere between cycles 23 and 24. Except for cycle 24's northern hemisphere, the measurements favor a single-cell meridional circulation model where the poleward flows persist down to ∼\sim0.8 R⊙R_\odot, accompanied by local inflows toward the activity belts in the near-surface layers. Cycle 24's northern hemisphere is anomalous: travel-time differences are significantly smaller when travel distances are greater than 20∘^\circ. This asymmetry between northern and southern hemispheres during cycle 24 was not present in previous measurements (e.g., Rajaguru & Antia 2015), which assumed a different P-angle error correction where south-north travel-time differences are shifted to zero at the equator for all travel distances. In our measurements, the travel-time differences at the equator are zero for travel distances less than ∼\sim30∘^\circ, but they do not vanish for larger travel distances. This equatorial offset for large travel distances need not be interpreted as a deep cross-equator flow; it could be due to the presence of asymmetrical local flows at the surface near the end points of the acoustic ray paths.Comment: accepted for publication in A&

    Time-distance helioseismology of solar Rossby waves

    Context. Solar Rossby waves (r modes) have recently been discovered in the near-surface horizontal flow field using the techniques of granulation-tracking and ring-diagram analysis applied to six years of SDO/HMI data. Aims. Here we apply time-distance helioseismology to the combined SOHO/MDI and SDO/HMI data sets, which cover 21 years of observations from May 1996 to April 2017. The goal of this study is to provide an independent confirmation over two solar cycles and in deeper layers of the Sun. Methods. We have measured south-north helioseismic travel times along the equator, which are sensitive to subsurface north-south flows. To reduce noise, the travel times were averaged over travel distances from 6∘^\circ to 30∘^\circ; the mean distance corresponds to a p-mode lower turning point of 0.91 R⊙R_\odot. The 21-year time series of travel-time measurements was split into three seven-year subsets and transformed to obtain power spectra in a corotating frame. Results. The power spectra all show peaks near the frequencies of the classical sectoral Rossby waves for azimuthal wavenumbers in the range 3≤m≤153 \leq m \leq 15. The mode frequencies and linewidths of the modes with m≤9m \leq 9 are consistent with a previous study whereas modes with m≥10m \geq 10 are shifted toward less negative frequencies by 10--20 nHz. While most of these modes have e-folding lifetimes on the order of a few months, the longest lived mode, m=3m=3, has an e-folding lifetime of more than one year. For each mode, the rms velocity at the equator is in the range of 1--3 m s−1^{-1} , with the largest values for m∼10m\sim10. No evidence for the m=2m=2 sectoral mode is found in the power spectrum, implying that the rms velocity of this mode is below ∼\sim0.5 m s−1^{-1}.Comment: accepted for publication in A&

    Inhibitors of DNA polymerase III as novel antimicrobial agents against gram-positive eubacteria

    6-Anilinouracils are selective inhibitors of DNA polymerase III, the enzyme required for the replication of chromosomal DNA in gram-positive bacteria (N. C. Brown, L. W. Dudycz, and G. E. Wright, Drugs Exp. Clin. Res. 12:555-564, 1986). A new class of 6-anilinouracils based on N-3 alkyl substitution of the uracil ring was synthesized and analyzed for activity as inhibitors of the gram-positive bacterial DNA polymerase III and the growth of gram-positive bacterial pathogens. Favorable in vitro properties of N-3-alkyl derivatives prompted the synthesis of derivatives in which the R group at N-3 was replaced with more-hydrophilic methoxyalkyl and hydroxyalkyl groups. These hydroxyalkyl and methoxyalkyl derivatives displayed K(i) values in the range from 0.4 to 2.8 microM against relevant gram-positive bacterial DNA polymerase IIIs and antimicrobial activity with MICs in the range from 0.5 to 15 microg/ml against a broad spectrum of gram-positive bacteria, including methicillin-resistant staphylococci and vancomycin-resistant enterococci. The BspA protein from Bacteroides forsythus mediates bacterial binding to fibronectin and contains leucine rich repeats of the Treponema pallidum (TpLRRP) family.     HIV-1 Integrase-Targeted Short Peptides Derived from a Viral Protein R Sequence

    HIV-1 integrase (IN) inhibitors represent a new class of highly effective anti-AIDS therapeutics. Current FDA-approved IN strand transfer inhibitors (INSTIs) share a common mechanism of action that involves chelation of catalytic divalent metal ions. However, the emergence of IN mutants having reduced sensitivity to these inhibitors underlies efforts to derive agents that antagonize IN function by alternate mechanisms. Integrase along with the 96-residue multifunctional accessory protein, viral protein R (Vpr), are both components of the HIV-1 pre-integration complex (PIC). Coordinated interactions within the PIC are important for viral replication. Herein, we report a 7-mer peptide based on the shortened Vpr (69–75) sequence containing a biotin group and a photo-reactive benzoylphenylalanyl residue, and which exhibits low micromolar IN inhibitory potency. Photo-crosslinking experiments have indicated that the peptide directly binds IN. The peptide does not interfere with IN-DNA interactions or induce higher-order, aberrant IN multimerization, suggesting a mode of action for the peptide that is distinct from clinically used INSTIs and developmental allosteric IN inhibitors. This compact Vpr-derived peptide may serve as a valuable pharmacological tool to identify a potential new pharmacologic site

    Abstract: Analysis of acute skin rejection in non-human primate models of face and hand allotransplantation

    Introduction: The incidence of acute rejection (AR) of the skin within the first year after hand or face transplantation is approximately 85% and up to 56% of patients experience multiple episodes1. Significant immunosuppression is required to prevent allograft loss, and recent studies suggest that repeated AR episodes can lead to VCA dysfunction and loss2. The mechanisms underlying variability in AR presentation remain poorly defined however. Materials and Methods: 8 cynomolgus monkeys received either an orthotopic hand (n=2) or heterotopic face VCA (n=6) from MHC-mismatched donors following induction with anti-thymocyte globulin. Post-operatively, triple immunosuppression – tacrolimus, mycophenolate mofetil, methylprednisolone – was maintained for up to 120 days before bone marrow transplantation (BMT) was performed. Protocol biopsies of VCA skin were performed at 30-day intervals for histopathology and flow cytometric analysis of resident skin leukocyte populations; VCA-resident cells were differentiated by H38 status (mouse antihuman HLA class I monoclonal antibody that cross reacts with cynomolgus monkeys) for donor or recipient derivation. Clinical AR was treated with steroids and further biopsies were taken for histologic confirmation; corresponding anti-donor responses were evaluated by mixed lymphocyte reaction (MLR) and allo-antibody formation. Results: Up to three episodes of AR (from POD 14, Banff I to II) developed while recipient animals were maintained on triple immunosuppression. Corresponding flow cytometric analyses demonstrate > 80% of skin-resident T lymphocytes (CD4+, CD8+) within VCA dermis were of recipient origin, suggesting rapid immigration of various lineages into the VCA. These observations coincided with the first episode of AR in fully mismatched recipients but haplomatched animals remained rejection-free. All but one episode of AR were successfully treated. No allo-antibodies were detected and anti-donor responses by MLR were comparable to that against third-party. Following BMT, mixed chimerism was detected and enabled immunosuppression withdrawal. However, this was transient and once lost, clinical AR developed and nearly 100% of both dermal and epidermal lymphocytes were recipient-derived. Conclusion: We report a clinically-relevant model for studying AR in VCA. Our results suggest that further understanding of the relative importance of MHC differences in transplant pairs may lead to differences in outcomes for VCA recipients maintained under standard immunosuppression. Immunosuppression-free tolerance of non-hematopoietic antigens in composite tissues can be achieved, but require additional strategies to achieve stable, rather than transient mixed chimerism following BMT

We aimed to compare the effects of beta-alanine on Traditional Resistance Training (TRAD) or Blood Flow Restriction Training (BFR). Methods: 19 subjects were randomly allocated to a Placebo (n = 10) or beta-alanine (n = 9) group. Subjects from both groups were trained unilaterally (unilateral arm curl) for six weeks, and each arm was trained using a different paradigm (BFR or TRAD). One repetition maximum (1RM) test measurements were performed before and after the strength training program. Work output was accessed as the total weight lifted (repetitions × weight lifted × sets) for the entire strength training program. Results: 1RM or total weight lifted was not increased by beta-alanine supplementation. However, the TRAD-trained arm showed a significantly increased 1RM and total weight lifted compared to the BFR arm (p \u3c 0.05). Conclusion: We conclude that in the short-term (6 weeks) and following the current experimental conditions, beta-alanine does not benefit BFR or TRAD in terms of total weight lifted (volume of training) or maximal strength (1RM)
