    Scale of Auditory Behaviors e testes auditivos comportamentais para avaliação do processamento auditivo em crianças falantes do português europeu

    PURPOSE: The objective of this research was to assess the auditory abilities of Portuguese children and compare such abilities to the score of the Scale of Auditory Behaviors (SAB). METHODS: Fifty-one children were evaluated with audiometry, speech audiometry, acoustic immittance measures, and eight behavioral tests involving dichotic listening, monotic listening, temporal processing, and sound localization. Their parents filled in the SAB questionnaire adapted to European A. SAB scores and auditory tests scores were submitted to Pearson's correlation coefficient. RESULTS: There is significant correlation between the score on SAB questionnaire and the auditory processing tests. The greatest coefficient was observed in temporal processing test (p=0.000). CONCLUSION: There was correlation between the score of SAB and the performance in auditory processing tests, suggesting that the SAB may be used for auditory processing screening.OBJETIVO: Investigar as habilidades auditivas de crianças portuguesas e verificar se há correlação entre aquelas e o escore do Scale of Auditory Behaviors (SAB). MÉTODOS: Todas as crianças foram submetidas a audiometria tonal, logoaudiometria, medidas de imitância acústica e oito testes comportamentais do processamento auditivo, envolvendo tarefas de escuta dicótica, escuta monótica, processamento temporal e localização sonora. Os pais das 51 crianças portuguesas avaliadas preencheram o questionário SAB adaptado ao português europeu. Foram calculados os valores do coeficiente de correlação de Pearson entre os escores obtidos no questionário e os dos testes do processamento auditivo. RESULTADOS: Observou-se correlação significativa entre o escore do questionário e o dos testes comportamentais, tendo a maior sido observada nos testes relacionados ao processamento temporal (p=0,000). CONCLUSÃO: Houve correlação entre o escore da SAB e os resultados obtidos nos testes auditivos comportamentais em crianças portuguesas, sugerindo que este questionário pode ser utilizado em triagem do processamento auditivo.CIEC – Research Centre on Child Studies, UM (FCT R&D 317

    Perceived neighborhood safety and incident mobility disability among elders: the hazards of poverty

    <p>Abstract</p> <p>Background</p> <p>We investigated whether lack of perceived neighborhood safety due to crime, or living in high crime neighborhoods was associated with incident mobility disability in elderly populations. We hypothesized that low-income elders and elders at retirement age (65 – 74) would be at greatest risk of mobility disability onset in the face of perceived or measured crime-related safety hazards.</p> <p>Methods</p> <p>We conducted the study in the New Haven Established Populations for Epidemiologic Studies of the Elderly (EPESE), a longitudinal cohort study of community-dwelling elders aged 65 and older who were residents of New Haven, Connecticut in 1982. Elders were interviewed beginning in 1982 to assess mobility (ability to climb stairs and walk a half mile), perceptions of their neighborhood safety due to crime, annual household income, lifestyle characteristics (smoking, alcohol use, physical activity), and the presence of chronic co-morbid conditions. Additionally, we collected baseline data on neighborhood crime events from the New Haven Register newspaper in 1982 to measure local area crime rates at the census tract level.</p> <p>Results</p> <p>At baseline in 1982, 1,884 elders were without mobility disability. After 8 years of follow-up, perceiving safety hazards was associated with increased risk of mobility disability among elders at retirement age whose incomes were below the federal poverty line (HR 1.56, 95% CI 1.02 – 2.37). No effect of perceived safety hazards was found among elders at retirement age whose incomes were above the poverty line. No effect of living in neighborhoods with high crime rates (measured by newspaper reports) was found in any sub-group.</p> <p>Conclusion</p> <p>Perceiving a safety hazard due to neighborhood crime was associated with increased risk of incident mobility disability among impoverished elders near retirement age. Consistent with prior literature, retirement age appears to be a vulnerable period with respect to the effect of neighborhood conditions on elder health. Community violence prevention activities should address perceived safety among vulnerable populations, such as low-income elders at retirement age, to reduce future risks of mobility disability.</p

    Barrier-to-autointegration factor 1 (Banf1) regulates poly [ADP-ribose] polymerase 1 (PARP1) activity following oxidative DNA damage

    The DNA repair capacity of human cells declines with age, in a process that is not clearly understood. Mutation of the nuclear envelope protein barrier-to-autointegration factor 1 (Banf1) has previously been shown to cause a human progeroid disorder, Néstor–Guillermo progeria syndrome (NGPS). The underlying links between Banf1, DNA repair and the ageing process are unknown. Here, we report that Banf1 controls the DNA damage response to oxidative stress via regulation of poly [ADP-ribose] polymerase 1 (PARP1). Specifically, oxidative lesions promote direct binding of Banf1 to PARP1, a critical NAD-dependent DNA repair protein, leading to inhibition of PARP1 auto-ADP-ribosylation and defective repair of oxidative lesions, in cells with increased Banf1. Consistent with this, cells from patients with NGPS have defective PARP1 activity and impaired repair of oxidative lesions. These data support a model whereby Banf1 is crucial to reset oxidative-stress-induced PARP1 activity. Together, these data offer insight into Banf1-regulated, PARP1-directed repair of oxidative lesions

    Distinguishing crystallographic from biological interfaces in protein complexes: role of intermolecular contacts and energetics for classification

    BACKGROUND: Study of macromolecular assemblies is fundamental to understand functions in cells. X-ray crystallography is the most common technique to solve their 3D structure at atomic resolution. In a crystal, however, both biologically-relevant interfaces and non-specific interfaces resulting from crystallographic packing are observed. Due to the complexity of the biological assemblies currently tackled, classifying those interfaces, i.e. distinguishing biological from crystal lattice interfaces, is not trivial and often prone to errors. In this context, analyzing the physico-chemical characteristics of biological/crystal interfaces can help researchers identify possible features that distinguish them and gain a better understanding of the systems. RESULTS: In this work, we are providing new insights into the differences between biological and crystallographic complexes by focusing on "pair-properties" of interfaces that have not yet been fully investigated. We investigated properties such intermolecular residue-residue contacts (already successfully applied to the prediction of binding affinities) and interaction energies (electrostatic, Van der Waals and desolvation). By using the XtalMany and BioMany interface datasets, we show that interfacial residue contacts, classified as a function of their physico-chemical properties, can distinguish between biological and crystallographic interfaces. The energetic terms show, on average, higher values for crystal interfaces, reflecting a less stable interface due to crystal packing compared to biological interfaces. By using a variety of machine learning approaches, we trained a new interface classification predictor based on contacts and interaction energetic features. Our predictor reaches an accuracy in classifying biological vs crystal interfaces of 0.92, compared to 0.88 for EPPIC (one of the main state-of-the-art classifiers reporting same performance as PISA). CONCLUSION: In this work we have gained insights into the nature of intermolecular contacts and energetics terms distinguishing biological from crystallographic interfaces. Our findings might have a broader applicability in structural biology, for example for the identification of near native poses in docking.     O processamento auditivo é fundamental para a cognição e pode ocasionar deficiência no aprendizado. Os portadores de sua deficiência podem ser habilitados, melhorando o desempenho escolar. É fundamental identificá-los. Dentre os que apresentam baixo custo e facilidade operacional está o exame de emissões otoacústicas. TIPO DE ESTUDO: Clínico e experimental. OBJETIVO: Estudar a relação do aproveitamento escolar com a inibição da emissão otoacústica transiente por estímulo auditivo contralateral. Material e Métodos: Foram avaliados 39 alunos, de sete a doze anos, sendo 19 (48,7%) com bom aproveitamento escolar e 20 (51,3%) com aproveitamento inadequado. Os exames emissão otoacústica com inibição contralateral foram comparados aos resultados de aproveitamento escolar. RESULTADOS: A falha da supressão da otoemissão transiente por estímulo acústico contralateral foi mais encontrada no grupo de crianças com mau aproveitamento escolar. Foi estabelecido um valor de corte de 1.6 dB SPL de redução da otoemissão que caracteriza a criança como pertencente ao grupo com mau aproveitamento com sensibilidade de 65,0%, especificidade de 72,2%, acurácia de 68,4% e valor preditivo positivo de 72,2%. CONCLUSÃO: O teste da falha da inibição contralateral da emissão otoacústica por estímulo auditivo contralateral é preditivo de transtorno do aproveitamento escolar em indivíduos de seis a doze anos de idade.<br>School learning can be hampered if there are defects on the central auditory process. Since those with auditory deficiency can be rehabilitated, it is fundamental that we identify them. Otoacoustic emissions test has low cost and operational ease. Study design: clinical and experimental. AIM: to study the relationship between school learning and transient otoacoustic emission suppression by contralateral stimuli. MATERIAL AND METHODS: 39 individuals, from 7 to 12 years of age were evaluated, 19 (48.7%) with good school performance and 20 (51.3%) poor performers. RESULTS: A transient otoacoustic emission suppression failure for contralateral acoustic stimuli was more frequently found among children with poor school performance. We established a value of 1.6 dB SPL for emission reduction that characterized those children as belonging to the poor learning performance group: sensitivity 65%, specificity 72,2%, accuracy of 68.4%, positive predictive value of 72.2%. CONCLUSION: The contralateral emission suppression test of the right ear can be predictive of school difficulties in individuals from six to twelve years of age
