    A transcriptomal analysis of bovine oviductal epithelial cells collected during the follicular phase versus the luteal phase of the estrous cycle

    BACKGROUND: Reproductive success depends on a functional oviduct for gamete storage, maturation, fertilization, and early embryonic development. The ovarian-derived steroids estrogen and progesterone are key regulators of oviductal function. The objective of this study was to investigate luteal and follicular phase-specific oviductal epithelial cell function by using microarray-based transcriptional profiling, to increase our understanding of mRNAs regulating epithelial cell processes, and to identify novel genes and biochemical pathways that may be found to affect fertility in the future. METHODS: Six normally cycling Angus heifers were assigned to either luteal phase (LP, n = 3) or follicular phase (FP, n = 3) treatment groups. Heifers in the LP group were killed between day 11 and 12 after estrus. Heifers in the FP group were treated with 25 mg PGF(2α) (Lutalyse, Pfizer, NY) at 8 pm on day 6 after estrus and killed 36 h later. Transcriptional profiling by microarray and confirmation of selected mRNAs by real-time RT-PCR analyses was performed using total RNA from epithelial cells isolated from sections of the ampulla and isthmus collected from LP and FP treatment groups. Differentially expressed genes were subjected to gene ontology classification and bioinformatic pathway analyses. RESULTS: Statistical one-way ANOVA using Benjamini-hochberg multiple testing correction for false discovery rate (FDR) and pairwise comparison of epithelial cells in the ampulla of FP versus LP groups revealed 972 and 597 transcripts up- and down-regulated, respectively (P < 0.05). Within epithelial cells of the isthmus in FP versus LP groups, 946 and 817 transcripts were up- and down-regulated, respectively (P < 0.05). Up-regulated genes from both ampulla and isthmus were found to be largely involved in cholesterol biosynthesis and cell cycle pathways, while down-regulated genes were found in numerous inflammatory response pathways. CONCLUSIONS: Microarray-based transcriptional profiling revealed phase of the cycle-dependent changes in the expression of mRNA within the epithelium of the oviducts’ ampulla and isthmus.     Climate factors affecting conception rate of high producing dairy cows in northeastern Spain.

    Abstract Summer heat stress is a main factor related to low conception rate in high producing dairy herds in warm areas worldwide. We assessed the impact of several climate variables on conception rate in high producing dairy cows in northeastern Spain by examining 10,964 inseminations. The temperature-humidity index (THI) was compared with maximum temperature in terms of its efficiency at predicting conception rate. The following data were recorded for each animal: herd, lactation number, insemination number, insemination date, inseminating bull, and AI technician along with climate variables such as mean and maximum temperatures, rainfall, mean and maximum THI for individual time points Days 7 to 1 before insemination, the day of insemination and 1, 2 and 3 days after insemination. Averages were also established for the following periods: from 7 days before insemination to the insemination day, from 3 days before insemination to the insemination day and from the insemination day to 3 days postinsemination. Based on the odds ratios, the likelihood of conception rate increased significantly by factors of 1.48, 1.47, 1.5, and 1.1 for the respective maximum THI classes <70, 71-75, 76-80, and 81-85 only on Day 3 before AI, while on the insemination day, it increased by factors of 1.73, 1.53, 1.11, and 1.3 for the respective maximum THI classes <70, 71-75, 76-80, and 81-85. In a subsequent logistic regression excluding mean and maximum THI, the effectiveness of temperature at predicting conception rate was evaluated. Although high, the fit of the second logistic regression model was slightly lower than that of the full model (P=0.88 versus P=0.98, respectively) and the information provided by the THI model. The likelihood of conception rate increased significantly by factors of 1.5, 1.2, 1.0, 1.0 for the respective maximum temperature classes <20, 21-25, 26-30, 31-35 degrees C on Day 1 after AI. The choice of the THI or temperature to monitor the farm environment would have to depend on the particular farm and situation. In our study conditions, the use of maximum temperature alone gives a new point of view regarding the information provided by the THI variables

    Considerations of viscosity in the preliminaries to mammalian fertilisation

    Migration of spermatozoa in the female genital tract will be strongly influenced by the viscosity of the fluids encountered, yet little systematic analysis has been given to such a consideration. This essay reviews the series of milieux confronting a fertilising sperm during its progression to the oviduct ampulla. Two groups are discussed, first those in which ejaculation is into the vagina, second those in which semen enters the uterus during a protracted mating. Viscous glycoprotein secretions that accumulate in the oviduct isthmus of both groups before ovulation are highlighted, as is the environment generated in the ampulla by the post-ovulatory suspension of oocyte(s), cumulus cells and spermatozoa; follicular and peritoneal fluids may also be present. The viscosity of all female tract fluids responds to cyclical variations in temperature, and these exist within the oviduct near the time of ovulation. Gradations in viscosity influence the pattern and strength of sperm flagellar activity and the rate of forward movement. Measurements of sperm motility are currently made in a physiological medium of constant viscosity and temperature, thereby overlooking changes in the female genital tract. A more sophisticated approach might reveal an adequate fertilising potential in a proportion of putatively poor semen samples

    Reproductive and Immune System Interactions in the Context of Life History and Sexual Selection Theory

    Host variation in parasite load abounds, both within and across natural populations. The forces that shape and maintain this variation, however, are much less obvious. Over the past two decades, the emerging field of ecoimmunology has begun to address the underlying sources of this variation and its evolutionary consequences. Clearly, spatial and temporal heterogeneity in the environment contribute to variation in host parasite load by varying parasite distribution and abundance. However, host variation in the ability to acquire resources, and differences in how these resources are allocated, also play an integral role in parasite susceptibility. That is to say, not all individuals within a population are capable of managing the cost of immune defense, or may employ different cost-managing strategies. This realization has spurred a surge in studies focused on how immune pathways compete with other costly physiological pathways, such as those associated with reproduction. Interest in this relationship has no doubt been driven by the reproductive system\u27s high energetic cost and its direct association with host fitness. In this chapter, I examine the interactions between reproduction and immunity to highlight the simultaneous role both play in the evolution of immunological and reproductive adaptations. I begin by placing this interaction in the context of life history theory and discuss how competition for limited resources may constrain the evolution and expression of both systems. I then discuss the role that parasites play in directly shaping reproductive adaptations through sexual selection. In this discussion, I attempt to shed light on the lingering controversy that has overshadowed genetic benefit models and provide concrete predictions for future directions
