    Therapy of Canine Hyperlipidemia with Bezafibrate.

    BackgroundBezafibrate (BZF) is effective in the treatment of hypertriglyceridemia in human patients, but there are no data on its use in dogs.ObjectiveTo assess the safety of BZF in hyperlipidemic dogs and its efficacy in decreasing serum triglyceride (TG) and cholesterol (CHO) concentrations.AnimalsForty-six dogs, 26 females and 20 males, mean (±SD) age of 9 (±3) years, with TG ≄150 mg/dL (33 dogs also were hypercholesterolemic [>300 mg/dL]).MethodsProspective, uncontrolled clinical trial. Dogs were treated with bezafibrate once daily, using 200 mg tablets at a dosage of 4-10 mg/kg (depending on body weight). Serum TG and CHO concentrations and alanine aminotransferase (ALT) and creatine kinase (CK) activity before and after 30 days of treatment were compared.ResultsSixteen dogs (34.8%) had primary hyperlipidemia, and 30 dogs (65.2%) had secondary hyperlipidemia (including spontaneous hyperadrenocorticism [41.3%, n = 19/46], chronic treatment with glucocorticoids [10.8%, n = 5/46], and hypothyroidism [15.2%, n = 7/46]). After 30 days, serum TG concentration normalized (<150 mg/dL) in 42 dogs (91.3%) and CHO concentration normalized (<270 mg/dL) in 22 of 33 dogs (66.7%). There was no difference in baseline TG concentration between the primary and secondary hyperlipidemia subgroups, but the decrease in TG concentration after treatment was greater in the primary hyperlipidemia subgroup. No adverse effects were observed, but ALT activity decreased significantly after 30 days of treatment.Conclusions and clinical importanceOver 30 days, BZF was safe and effective in treatment of primary and secondary hyperlipidemia in dogs

    Blue-green infrastructure in view of Integrated Urban Water Management: A novel assessment of an effectiveness index

    Addressing urban water management challenges requires a holistic view. Sustainable approaches such as blue-green infrastructure (BGI) provide several benefits, but assessing their effectiveness demands a systemic approach. Challenges are magnified in informal areas, leading to the combination of integrated urban water management (IUWM) with BGI as a proposed solution by this research. We employed the Urban Water Use (UWU) model to assess the effectiveness index (EI) of BGI measures in view of IUWM after stakeholder consultation. The procedure in this novel assessment includes expert meetings for scenario building and resident interviews to capture the community's vision. To assess the impact of IUWM on the effectiveness of BGI measures, we proposed a simulation with BGI only and then three simulations with improvements to the water and sewage systems. The results of the EI analysis reveal a substantial improvement in the effectiveness of BGI measures through IUWM combination. Moreover, we offer insights into developing strategies for UWU model application in informal settlements, transferrable to diverse urban areas. The findings hold relevance for policymakers and urban planners, aiding informed decisions in urban water management

    A first report on the efficacy of a single intra-articular administration of blood cell secretome, triamcinolone acetonide, and the combination of both in dogs with osteoarthritis

    Research Areas: Veterinary SciencesBackground: Osteoarthritis represents a signifcant welfare problem for many dogs, with limited therapeutic options other than palliative pain control. To evaluate the efect of the intra-articular administration of blood cell secretome and triamcinolone, 15 dogs with bilateral hip osteoarthritis were randomly assigned to a blood cell secretome (BCSG, n=5), triamcinolone (TG) or their combination group (BCS+TG, n=5). BCSG received a single intra-articular administration of 3 ml of blood cell secretome, TG 0.5 ml of triamcinolone acetonide 40 mg/ml, and BCS+TG received the combined products. The volume to administrate was corrected to 3.5 ml with saline. On days 0, 8, 15, 30, 60, 90, 120, 150, and 180, a copy of the Canine Brief Pain Inventory (divided into pain interference score—PIS and Pain Severity Score—PSS), Liverpool Osteoarthritis in Dogs (LOAD), Hudson Visual Analogue Scale (HVAS), and Canine Orthopedic Index (COI, divided into function, gait, stifness, and quality of life) was completed. Results were analyzed with the Kruskal–Wallis test and the Kaplan–Meier estimators were conducted and compared with the Log Rank test, p<0.05. Results: Animals in the sample had a mean age of 9.0±2.9 years and a bodyweight of 28.8±4.1 kg. Hips were classifed as moderate (8) and severe (7) osteoarthritis. No diferences were found between groups at T0 regarding considered evaluations. Signifcant diferences were observed between groups in pain scores from+8d-+150d, with BCS+TG exhibiting better results. The same was observed for HVAS and LOAD, from+8d-+120d. Improvements were also observed in several dimensions of the COI. Kaplan–Meier estimators showed that BCS+TG produced longer periods with better results, followed by BCSG and TG. Conclusion: The intra-articular administration of blood cell secretome improved the clinical signs and scores of several clinical metrology instruments in dogs with hip OA, particularly when combined with triamcinolone.     Olanzapine-Induced Hyperprolactinemia: Two Case Reports

    Background: Hyperprolactinemia is a common consequence of treatment with antipsychotics. It is usually defined by a sustained prolactin level above the laboratory upper level of normal in conditions other than that where physiologic hyperprolactinemia is expected. Normal prolactin levels vary significantly among different laboratories and studies. Several studies indicate that olanzapine does not significantly affect serum prolactin levels in the long term, although this statement has been challenged. Aims: Our aim is to report two olanzapine-induced hyperprolactinemia cases observed in psychiatric consultations. Methods: Medical records of the patients who developed this clinical situation observed in psychiatric consultations in the Psychiatry Department of the Prof. Dr. Fernando Fonseca Hospital during the year of 2017 were analyzed, complemented with a non-systematic review of the literature. Results: The case reports consider two women who developed prolactin-related symptoms after the initiation of olanzapine. No baseline prolactinemia was obtained, and prolactin serum levels were only evaluated after prolactin-related symptoms developed: at the time of its measurement, both patients had been taking olanzapine for more than 24 weeks. Hyperprolactinemia was found to be present in Case 2, whereas Case 1 (a 49-year-old woman) had "normal" serum prolactin levels for premenopausal and prolactin levels slightly above the maximum levels for postmenopausal women. Both patients underwent similar pharmacological adjustments, which comprised switches from olanzapine to aripiprazole. After all pharmacological changes, prolactin serum levels decreased to normal range values and prolactin-related symptoms disappeared. Discussion/Conclusions: Laboratorial and literature prolactinemia values variability and discrepancies may make the management of borderline hyperprolactinemia clinical situations difficult. Baseline prolactin levels should have been obtained, as they help in the management of patients who develop neuroleptic-induced hyperprolactinemia. Prolactin-related symptoms can occur with borderline or normal standardized prolactinemia values. Olanzapine-induced hyperprolactinemia is a rare but possible event.     A simple ZVI-Fenton pre-oxidation using steel-nails for NOM degradation in water treatment

    The feasibility of a heterogeneous Fenton Process (ZVI/H2O2) using commercial low-carbon-steel nails as the Zero-Valent Iron (ZVI) source was evaluated for the first time for the removal of natural organic matter (NOM) from natural surface waters with distinct physico-chemical characteristics. The synergistic effect of ZVI nails and H2O2 on the process was confirmed. Results showed similar removal efficiencies of NOM in water samples from Thames river and Regent's Park lake (both in London, UK) (under initial pH 3.5 and 100% excess of H2O2 dosage), reaching dissolved organic carbon (DOC) removals of 61.6% ± 3.0 and 59.6% ± 4.7, and UV254 removals of 79.9% ± 0.6 and 77.3 ± 6.2, respectively with 60 min of batch reaction time. ZVI nail surface characterization by scanning electron microscopy (SEM), X-ray energy-dispersive spectroscopy (EDS), and X-ray photoemission spectroscopy (XPS) revealed the formation of a passivating oxide-hydroxide layer on the nail during the reaction, which reduces its surface activity with 20% in continuous use. Results indicate that ZVI/H2O2 process using commercial iron nails is a promising pre-oxidation step for drinking water treatment. The low cost of commercial nails together with the facility of separating them from the water are the main advantages for the application of this process in remote regions with limitations in infrastructure and/or finance

    Impacts of behavioural factors on the household water consumption in urban areas

    Gaps in understanding what influences household water consumption has led water providers failing to convince their customers to report sustainable practices. To this end, the present study aimed to answer the question, “How do social and cultural factors influence water consumption in urban areas”? The response to this issue has been identified through an investigation that involved a group of selected socio-cultural factors, whose analysis was based on collected survey data from participants in Lagos-Nigeria, Salvador-Brazil, Sao Paulo-Brazil, London-UK and Los Angeles-USA. The Capability-Opportunity-Motivation-Behaviour (COM-B) model was used as a data analysis framework to identify influences. The investigation revealed that Motivation is the most reported driver of water consumption. In a scale from 0 (lowest) to 5 (highest), this component presented the most significant scores in Lagos (3.93), Salvador (4.13), Sao Paulo (3.88), London (4.13) and Los Angeles (3.59). The Capability dimension had the second-highest weight in Lagos, Salvador, Sao Paulo, and Los Angeles, with scores of 2.80, 3.60, 3.60 and 3.20, respectively. Participants from London have Opportunity (score= 2.88) as the second influential pillar in water consumption. These findings are aimed at helping to best drive water saving practices by gaining insight into factors underpinning water consumption in a structured manner

    Characterization of weight-bearing compensation in dogs with bilateral hip osteoarthritis

    Área de pesquisa: Veterinary SciencesTo describe the weight-bearing compensation in working dogs with bilateral hip osteoarthritis (OA), 50 police working dogs were evaluated with a weight distribution platform at the initial evaluation and after intra-articular treatment (a negative control 0.9% sodium chloride (NaCl), a platelet concentrate, Hylan G-F 20, triamcinolone hexacetonide or stanozolol). Six evaluation sessions were performed, over a 180-day period. Results were compared by breed, age, sex, weight and Orthopedic Foundation for Animals hip grade scores with the Independent Samples T-Test, repeated samples Analysis of variance and Pearson correlation coefficient, P < .05. Animals had a mean age of 6.5 § 2.4 years and a bodyweight of 26.7 § 5.2kg. No significant differences were observed when comparing weight-bearing for different breeds, sex, hip grades or weight during the initial evaluation. Significant differences were observed in deviation (P < .01) and symmetry index (P < .01) between the control and treatment groups during the follow-up period. A weight shift from pelvic to thoracic limbs was observed, with a weak, although a significant, correlation between a pelvic limb and the opposing contralateral thoracic limb. Labrador Retrievers showed higher symmetry index and deviation from normal values during the follow-up period than German Shepherd Dogs and Dutch Shepherd Dogs. Male dogs also showed higher symmetry index and deviation compared with females. At this period, the symmetry index showed a weak, although significant, correlation with body weight. Weight-bearing of all limbs correlated with the remaining limbs, reflecting a more balanced weight distribution than the initial evaluation.
