    Some procedures for computerized ability testing

    For computerized test systems to be operational, the use of item response theory is a prerequisite. As opposed to classical test theory, in item response models the abilities of the examinees and the properties of the items are parameterized separately. Hence, when measuring the abilities of examinees, the model implicitly corrects for the item properties, and measurement on an item-independent scale is possible. In addition, item response theory offers the use of test and item information as local reliability indices defined on the ability scale. In this chapter, it is shown how the main features of item response theory have given rise to the development of promising procedures for computerized testing. Among the topics discussed are procedures for item bank calibration, automated test construction, adaptive test administration, generating norm distributions, and diagnosing test scores

    Locative and Directional Prepositions in Conceptual Spaces: The Role of Polar Convexity

Focusing on purely spatial locative and directional prepositions, we analyze both types of prepositions in terms of polar coordinates instead of Cartesian coordinates. This makes it possible to demonstrate that the property of convexity holds quite generally in the domain of prepositions of location and direction, supporting the important role that this property plays in conceptual spaces

    The economic and ecological effects of water management choices in the upper Niger river: Development of decision support methods

    One million people in the Inner Niger Delta make a living from arable farming, fisheries and livestock. Upstream dams (one built for electricity generation and one for irrigation) affect this downstream multifunctional use of water. Additionally, the Inner Niger Delta, which is one of the largest Ramsar sites in the world, is a hotspot of biodiversity and accommodates two of the largest known breeding colonies of large wading birds in Africa and in addition, is a vital part of the eco-regional network, supporting up to 3 to 4 million staging waterbirds, residents and migrants from all over Europe and western Asia. The hydrological and related ecological conditions in the Inner Delta largely determine the population size of these waterbird species. The major aim of the three-year study was to develop a decision-support system for river management in the Upper Niger, in which ecological and socio-economical impacts and benefits of dams and irrigation systems can be analysed in relation to different water management scenarios. The study involves various components: hydrology, arable farming, livestock, fisheries, ecology and socio-economics. An economic analysis has been conducted to determine the role of dams in the economy of the Inner Niger Delta and the Upper Niger region. By innovatively combining the above information on hydrology, ecology, fisheries, and agriculture, the study shows that building new dams is not an efficient way to increase economic growth and reduce poverty in the region. In fact, such efforts are counter-effective. Instead, development efforts should be aimed at improving the efficiency of the existing infrastructure, as well as of current economic activities in the Inner Niger Delta itself. This approach will also provide greater certainty for the essential eco-regional network functioning of the Inner Delta. © 2006 Taylor & Francis

    SlgA, the homologue of the human schizophrenia associated PRODH gene, acts in clock neurons to regulate <i>Drosophila </i>aggression

    Mutations in the proline dehydrogenase gene PRODH are linked to behavioral alterations in schizophrenia and as part of DiGeorge and velo-cardio-facial syndromes, but the role of PRODH in their etiology remains unclear. Here, we establish a Drosophila model to study the role of PRODH in behavioral disorders. We determine the distribution of the Drosophila PRODH homolog slgA in the brain and show that knockdown and overexpression of human PRODH and slgA in the lateral neurons ventral (LNv) lead to altered aggressive behavior. SlgA acts in an isoform-specific manner and is regulated by casein kinase II (CkII). Our data suggest that these effects are, at least partially, due to effects on mitochondrial function. We thus show that precise regulation of proline metabolism is essential to drive normal behavior and we identify Drosophila aggression as a model behavior relevant for the study of the mechanisms that are impaired in neuropsychiatric disorders

    Population overlap and habitat segregation in wintering Black-tailed Godwits Limosa limosa

    Distinct breeding populations of migratory species may overlap both spatially and temporally, but differ in patterns of habitat use. This has important implications for population monitoring and conservation. To quantify the extent to which two distinct breeding populations of a migratory shorebird, the Black-tailed Godwit Limosa limosa, overlap spatially, temporally and in their use of different habitats during winter. We use mid-winter counts between 1990 and 2001 to identify the most important sites in Iberia for Black-tailed Godwits. Monthly surveys of estuarine mudflats and rice-fields at one major site, the Tejo estuary in Portugal in 2005-2007, together with detailed tracking of colour-ringed individuals, are used to explore patterns of habitat use and segregation of the Icelandic subspecies L. l. islandica and the nominate continental subspecies L. l. limosa. In the period 1990-2001, over 66 000 Black-tailed Godwits were counted on average in Iberia during mid-winter (January), of which 80% occurred at just four sites: Tejo and Sado lower basins in Portugal, and Coto Dontildeana and Ebro Delta in Spain. Icelandic Black-tailed Godwits are present throughout the winter and forage primarily in estuarine habitats. Continental Black-tailed Godwits are present from December to March and primarily use rice-fields. Iberia supports about 30% of the Icelandic population in winter and most of the continental population during spring passage. While the Icelandic population is currently increasing, the continental population is declining rapidly. Although the estuarine habitats used by Icelandic godwits are largely protected as Natura 2000 sites, the habitat segregation means that conservation actions for the decreasing numbers of continental godwits should focus on protection of rice-fields and re-establishment of freshwater wetlands

    PMP22 related neuropathies: Charcot-Marie-Tooth disease type 1A and Hereditary Neuropathy with liability to Pressure Palsies

    PMP22 related neuropathies comprise (1) PMP22 duplications leading to Charcot-Marie-Tooth disease type 1A (CMT1A), (2) PMP22 deletions, leading to Hereditary Neuropathy with liability to Pressure Palsies (HNPP), and (3) PMP22 point mutations, causing both phenotypes. Overall prevalence of CMT is usually reported as 1:2,500, epidemiological studies show that 20-64% of CMT patients carry the PMP22 duplication. The prevalence of HNPP is not well known. CMT1A usually presents in the first two decades with difficulty walking or running. Distal symmetrical muscle weakness and wasting and sensory loss is present, legs more frequently and more severely affected than arms. HNPP typically leads to episodic, painless, recurrent, focal motor and sensory peripheral neuropathy, preceded by minor compression on the affected nerve. Electrophysiological evaluation is needed to determine whether the polyneuropathy is demyelinating. Sonography of the nerves can be useful. Diagnosis is confirmed by finding respectively a PMP22 duplication, deletion or point mutation. Differential diagnosis includes other inherited neuropathies, and acquired polyneuropathies. The mode of inheritance is autosomal dominant and de novo mutations occur. Offspring of patients have a chance of 50% to inherit the mutation from their affected parent. Prenatal testing is possible; requests for prenatal testing are not common. Treatment is currently symptomatic and may include management by a rehabilitation physician, physiotherapist, occupational therapist and orthopaedic surgeon. Adult CMT1A patients show slow clinical progression of disease, which seems to reflect a process of normal ageing. Life expectancy is norma

    Identification of the fructose transporter GLUT5 (SLC2A5) as a novel target of nuclear receptor LXR

    Fructose has become a major constituent of our modern diet and is implicated as an underlying cause in the development of metabolic diseases. The fructose transporter GLUT5 (SLC2A5) is required for intestinal fructose absorption. GLUT5 expression is induced in the intestine and skeletal muscle of type 2 diabetes (T2D) patients and in certain cancers that are dependent on fructose metabolism, indicating that modulation of GLUT5 levels could have potential in the treatment of these diseases. Using an unbiased screen for transcriptional control of the human GLUT5 promoter we identified a strong and specific regulation by liver X receptor alpha (LXR alpha, NR1H3). Using promoter truncations and site-directed mutagenesis we identified a functional LXR response element (LXRE) in the human GLUT5 promoter, located at -385 bp relative to the transcriptional start site (TSS). Finally, mice treated with LXR agonist T0901317 showed an increase in Glut5 mRNA and protein levels in duodenum and adipose tissue, underscoring the in vivo relevance of its regulation by LXR. Together, our findings show that LXR alpha regulates GLUT5 in mice and humans. As a ligand-activated transcription factor, LXR alpha might provide novel pharmacologic strategies for the selective modulation of GLUT5 activity in the treatment of metabolic disease as well as cancer.</p
