Immobilization of cells by electrostatic droplet generation: a model system for potential application in medicine
The process of electrostatic extrusion as a method for cell immobilization was investigated that could be used for potential applications in medicine. An attempt was made to assess the effects of cell addition and polymer concentration on the overall entrapment procedure, ie, on each stage of immobilization: polymer-cell suspension rheological characteristics, electrostatic extrusion process, and the process of gelation. The findings should contribute to a better understanding of polymer–cell interactions, which could be crucial in possible medical treatments. Alginate–yeast was used as a model system for carrier-cells. The electrostatic extrusion was considered as a complex two-phase flow system and the effects of cell and alginate concentrations on the resulting microbead size and uniformity were assessed. Under investigated conditions, microbeads 50–600 μm in diameter were produced and the increase in both alginate and cell concentrations resulted in larger microbeads with higher standard deviations in size. We attempted to rationalize the findings by rheological characterization of the cell–alginate suspensions. Rheological characterization revealed non-Newtonian, pseudoplastic behavior of cell-alginate suspensions with higher viscosities at higher alginate concentrations. However, the presence of cells even at high concentrations (5×108 and 1×109 cells/mL) did not significantly affect the rheological properties of Na-alginate solution. Lastly, we investigated the kinetics of alginate gelation with respect to the quantity of Ca2+ ions and cell presence. The gelation kinetics were examined under conditions of limited supply with Ca2+ ions, which can be essential for immobilization of highly sensitive mammalian cells that require minimal exposure to CaCl2 solution. The molar ratio of G units to Ca2+ ions of 3.8:1 provided complete crosslinking, while the increase in alginate concentration resulted in prolonged gelation times but higher strength of the resulting gel. The cell presence decreased the rate of network formation as well as the strength of the obtained Ca-alginate hydrogel
Cellulose Fibre-Reinforced Biofoam for Structural Applications
Traditionally, polymers and macromolecular components used in the foam industry are mostly derived from petroleum. The current transition to a bio-economy creates demand for the use of more renewable feedstocks. Soybean oil is a vegetable oil, composed mainly of triglycerides, that is suitable material for foam production. In this study, acrylated epoxidized soybean oil and variable amounts of cellulose fibres were used in the production of bio-based foam. The developed macroporous bio-based architectures were characterised by several techniques, including porosity measurements, nanoindentation testing, scanning electron microscopy, and thermogravimetric analysis. It was found that the introduction of cellulose fibres during the foaming process was necessary to create the three-dimensional polymer foams. Using cellulose fibres has potential as a foam stabiliser because it obstructs the drainage of liquid from the film region in these gas-oil interfaces while simultaneously acting as a reinforcing agent in the polymer foam. The resulting foams possessed a porosity of approximately 56%, and the incorporation of cellulose fibres did not affect thermal behaviour. Scanning electron micrographs showed randomly oriented pores with irregular shapes and non-uniform pore size throughout the samples
Protein disorder in the human diseasome: unfoldomics of human genetic diseases
Background
Intrinsically disordered proteins lack stable structure under physiological conditions, yet carry out many crucial biological functions, especially functions associated with regulation, recognition, signaling and control. Recently, human genetic diseases and related genes were organized into a bipartite graph (Goh KI, Cusick ME, Valle D, Childs B, Vidal M, et al. (2007) The human disease network. Proc Natl Acad Sci U S A 104: 8685–8690). This diseasome network revealed several significant features such as the common genetic origin of many diseases.
Methods and findings
We analyzed the abundance of intrinsic disorder in these diseasome network proteins by means of several prediction algorithms, and we analyzed the functional repertoires of these proteins based on prior studies relating disorder to function. Our analyses revealed that (i) Intrinsic disorder is common in proteins associated with many human genetic diseases; (ii) Different disease classes vary in the IDP contents of their associated proteins; (iii) Molecular recognition features, which are relatively short loosely structured protein regions within mostly disordered sequences and which gain structure upon binding to partners, are common in the diseasome, and their abundance correlates with the intrinsic disorder level; (iv) Some disease classes have a significant fraction of genes affected by alternative splicing, and the alternatively spliced regions in the corresponding proteins are predicted to be highly disordered; and (v) Correlations were found among the various diseasome graph-related properties and intrinsic disorder.
Conclusion
These observations provide the basis for the construction of the human-genetic-disease-associated unfoldome
Detection and prediction of epileptic seizures: a patient's case study
Human epilepsy is a disease characterized by sudden, unprovoked, recurrent seizures accompanied by pathological electrical activity in the brain, and is frequently resistant to drug treatment. The ability to anticipate the onset of these incapacitating episodes would -hopefully- permit clinical interventions and avoid the serious consequences they may provoque. In this work we first consider the problem of detection of the onset of an epileptic seizure, comparing linear and non-linear techniques of time series analysis applied to electro-encephalogram recordings against onset times determined clinically. Automatic detection would be useful for fast seizure recognition which is of importance for further diagnostic procedures. The second, more ambitious goal is to foresee the ocurrence of an upcoming seizure, exploiting the widely conjectured "decrease in complexity" associated with ictal episodes. Roughly speaking, we monitor changes in time-varying windowed estimates of different magnitudes characterizing the brain's intrinsic dynamics. We face these problems for five seizures belonging to a single patient, using two strategies of brain activity reconstruction: single and multiple-channel delay embedding of the dynamics. We have found that the studied approaches successfully reflect the non-stationary character of ictal episodes, and seizure onsets were clearly accussed. For prediction, the criteria employed in the determination of clinical onset times appeared crucial.I Workshop de Agentes y Sistemas Inteligentes (WASI)Red de Universidades con Carreras en Informática (RedUNCI
Graphene Nano-Ribbon Electronics
We have fabricated graphene nano-ribbon field-effect transistor devices and
investigated their electrical properties as a function of ribbon width. Our
experiments show that the resistivity of a ribbon increases as its width
decreases, indicating the impact of edge states. Analysis of temperature
dependent measurements suggests a finite quantum confinement gap opening in
narrow ribbons. The electrical current noise of the graphene ribbon devices at
low frequency is found to be dominated by the 1/f noise.Comment: 6 pages, 7 figure
Polyphenol Content and Antioxidant Activity of Sour Cherries From Serbia
The aim of this study was to evaluate the content of phenolics: the total phenols (TP), flavonoids (TF), anthocyanins (TA), as well as the total antioxidant\ud
capacity (TAC) in three sour cherry cultivars (Prunus cerasus L.) introduced to the southeast Serbia climate conditions. Among the investigated sour cherries,\ud
„Oblačinska“ cultivar contained the highest amounts of all groups of phenolics, followed by „Cigančica“ > „Marela“. A significant difference were observed in the phenolic content among different cultivars and growing seasons (p 0.05), and the phenolic compounds were significantly higher in the growing season 2009. The examined cultivars possess a high antioxidant capacity, and all phenolics of highy correlation with TAC. The following compounds were identified and quantified using HPLC-DAD: 4 anthocyanins, the most abundant of which was cyanidin-3-glucoside in “Marela” and “Oblačinska”, and cyanidin-3-glucosylrutinoside in „Cigančica“, and 4 hydroxycinnamic acids, the most abundant of which was neochlorogenic acid in all sour cherry cultivars. The growing and ripening process on the tree of sour cherry cv. „Oblačinska“ was evaluated also. The results showed significant increases in total phenols during the ripening, the total anthocyanins and total antioxidant capacity and 4 quantified anthocyanins, however the neochlorogenic acid decreased during the ripening. The study indicated that the growing and climate conditions in southeast Serbia are convenient for introducing sour cherry cultivars.\u
Cosmic string parameter constraints and model analysis using small scale Cosmic Microwave Background data
We present a significant update of the constraints on the Abelian Higgs
cosmic string tension by cosmic microwave background (CMB) data, enabled both
by the use of new high-resolution CMB data from suborbital experiments as well
as the latest results of the WMAP satellite, and by improved predictions for
the impact of Abelian Higgs cosmic strings on the CMB power spectra. The new
cosmic string spectra (presented in a previous work) were improved especially
for small angular scales, through the use of larger Abelian Higgs string
simulations and careful extrapolation. If Abelian Higgs strings are present
then we find improved bounds on their contribution to the CMB anisotropies,
f10< 0.095, and on their tension, G\mu< 0.57 x 10^-6, both at 95% confidence
level using WMAP7 data; and f10 < 0.048 and G\mu < 0.42 x 10^-6 using all the
CMB data. We also find that using all the CMB data, a scale invariant initial
perturbation spectrum, ns=1, is now disfavoured at 2.4\sigma\ even if strings
are present. A Bayesian model selection analysis no longer indicates a
preference for strings.Comment: 8 pages, 3 figures; Minor corrections, matches published versio
Characterization of MgAl₂O₄ Sintered Ceramics
Single phase MgAl2O4 was made from a one-to-one molar ratio of MgO and Al2O3 powders mixed using ball-milling. Mixtures of MgO and Al2O3 were subsequently treated in planetary ball mill for 30, 60, 90 and 120 minutes in air. The aim of this study was to examine phase composition, microstructure, and densification behavior of sintered specimens. After sintering in dilatometer at 1500 °C, the powder was converted to single phase MgAl2O4. The results show that mechanical activation improved the densification behavior of MgAl2O4 sintered specimens, and it reduced the onset temperature for sintering by approx. 100°C. Based on dilatometer data, powders were subsequently densified at 1450°C by hot pressing. Almost all specimens exhibited full density, while sample activated for 30 minutes showed the fastest densification rate
