    A cryptic promoter in potato virus X vector interrupted plasmid construction

    BACKGROUND: Potato virus X has been developed into an expression vector for plants. It is widely used to express foreign genes. In molecular manipulation, the foreign genes need to be sub-cloned into the vector. The constructed plasmid needs to be amplified. Usually, during amplification stage, the foreign genes are not expressed. However, if the foreign gene is expressed, the construction work could be interrupted. Two different viral genes were sub-cloned into the vector, but only one foreign gene was successfully sub-cloned. The other foreign gene, canine parvovirus type 2 (CPV-2) VP1 could not be sub-cloned into the vector and amplified without mutation (frame shift mutation). RESULTS: A cryptic promoter in the PVX vector was discovered with RT-PCR. The promoter activity was studied with Northern blots and Real-time RT-PCR. CONCLUSION: It is important to recognize the homologous promoter sequences in the vector when a virus is developed as an expression vector. During the plasmid amplification stage, an unexpected expression of the CPV-2 VP1 gene (not in the target plants, but in E. coli) can interrupt the downstream work

Il capitolo riguardante l'alta quota appenninica è il più corposo e propone il nuovo syntaxon Leontopodio-Elynion a livello di alleanza. La grande novità sta nel fatto che questa alleanza è da considerarsi anfi-Adriatica e unsice Appennino e balcani, differenziandosi dall'alleanza già definita da altri per le Alpi. i Pirenei e i Carpazi.During preparation of the European checklist of vegetation units (EuroVegChecklist), it became clear that some earlier described syntaxa need to be typified in order to stabilize nomenclature and some new syntaxa need to be described. Here we propose nomenclature adjustments and formal description of four new alliances for the Arctic, alpine and oro-Mediterranean vegetation of Europe, Greenland and Anatolia. First, we typify the class Juncetea trifidi. Second, we describe four new alliances, such as the Puccinellion nuttallianae (Low-Arctic salt steppes of Greenland; class Saxifrago tricuspidatae-Calamagrostietea purpurascentis), Dryado octopetalae- Caricion arctisibiricae (Arctic tundra vegetation of north-eastern European Russia; class Carici rupestris-Kobresietea bellardii), Leontopodio nivalis-Elynion myosuroidis (southern European alpine tundra vegetation; class Carici rupestris-Kobresietea bellardii) and Lagotido uralensis-Caricion ensifoliae (alpine tundra vegetation of the Southern Ural Mountains; class Juncetea trifidi). Two new associations are described within the first two of these alliances. Finally, we present an interpretation of the alliance Muscario-Scillion nivalis

    Motor preparation of spatially and temporally defined movements: Evidence from startle

Copyright © 2011 the American Physiological Society.Previous research has shown that the preparation of a spatially targeted movement performed at maximal speed is different from that of a temporally constrained movement (Gottlieb et al. 1989b). In the current study, we directly examined preparation differences in temporally vs. spatially defined movements through the use of a startling stimulus and manipulation of the task goals. Participants performed arm extension movements to one of three spatial targets (20°, 40°, 60°) and an arm extension movement of 20° at three movement speeds (slow, moderate, fast). All movements were performed in a blocked, simple reaction time paradigm, with trials involving a startling stimulus (124 dB) interspersed randomly with control trials. As predicted, spatial movements were modulated by agonist duration and timed movements were modulated by agonist rise time. The startling stimulus triggered all movements at short latencies with a compression of the kinematic and electromyogram (EMG) profile such that they were performed faster than control trials. However, temporally constrained movements showed a differential effect of movement compression on startle trials such that the slowest movement showed the greatest temporal compression. The startling stimulus also decreased the relative timing between EMG bursts more for the 20° movement when it was defined by a temporal rather than spatial goal, which we attributed to the disruption of an internal timekeeper for the timed movements. These results confirm that temporally defined movements were prepared in a different manner from spatially defined movements and provide new information pertaining to these preparation differences

    We present a systematic derivation of the constraints that the relativity principle imposes between coefficients of a deformed (but rotational invariant) momentum composition law, dispersion relation, and momentum transformation laws, at first order in a power expansion of an ultraviolet energy scale. This work generalizes previous results to the case of particle-dependent relativistic kinematics, which can have interesting phenomenological applications that we explore in the second part of the manuscript.Comment: 13 pages, no figure

    The effect of boundary conditions on the vacuum structure of quantum field theories is analysed from a quantum information viewpoint. In particular, we analyse the role of boundary conditions on boundary entropy and entanglement entropy. The analysis of boundary effects on massless free field theories points out the relevance of boundary conditions as a new rich source of information about the vacuum structure. In all cases the entropy does not increase along the flow from the ultraviolet to the infrared.Comment: 10 page

    Local and Global Casimir Energies for a Semitransparent Cylindrical Shell

    The local Casimir energy density and the global Casimir energy for a massless scalar field associated with a λδ\lambda\delta-function potential in a 3+1 dimensional circular cylindrical geometry are considered. The global energy is examined for both weak and strong coupling, the latter being the well-studied Dirichlet cylinder case. For weak-coupling,through O(λ2)\mathcal{O}(\lambda^2), the total energy is shown to vanish by both analytic and numerical arguments, based both on Green's-function and zeta-function techniques. Divergences occurring in the calculation are shown to be absorbable by renormalization of physical parameters of the model. The global energy may be obtained by integrating the local energy density only when the latter is supplemented by an energy term residing precisely on the surface of the cylinder. The latter is identified as the integrated local energy density of the cylindrical shell when the latter is physically expanded to have finite thickness. Inside and outside the delta-function shell, the local energy density diverges as the surface of the shell is approached; the divergence is weakest when the conformal stress tensor is used to define the energy density. A real global divergence first occurs in O(λ3)\mathcal{O}(\lambda^3), as anticipated, but the proof is supplied here for the first time; this divergence is entirely associated with the surface energy, and does {\em not} reflect divergences in the local energy density as the surface is approached.Comment: 28 pages, REVTeX, no figures.
