    Resonant diaphragm pressure measurement system with ZnO on Si excitation

    The principle of measuring pressure by means of a resonant diaphragm has been studied. An oscillator consisting of an integrated amplifier with a piezoelectrically driven diaphragm in its feedback loop has been built. The oscillator frequency is accurately proportional to the square of the pressure in the range of 60 to 130 Torr.\ud The frequency range is 1324 to 1336 Hz (this range being limited by a spurious mode which could be suppressed by better processing) for a 25 mm diameter diaphragm made of a silicon wafer and with PZT ceramics as driver and receptor. We have made an integrated version (1 × 1 mm2) of a square resonant diaphragm pressure guage by selective etching of (1 0 0) planes with ethylenediamine. The piezoelectric driving materials was sputtered zinc oxide. A driver was deposited midway between the bending point and the point of greatest curvature.\ud A receptor was located at a symmetrical position to give a optimum transfer condition.\ud The integrated current amplifier had a low impedance differential input stage, two gain cells and a high impedance output stage. These electrical conditions ensured maximum elastic freedom of the diaphragm. A digital circuit in I2L technology has been designed and made with eight-bit parallel read out of the frequency. This circuit may be directly connected to a microprocessor. The whole system contains the sensor chip, the analog amplifier chip and the digital chip, all in compatible technology.\ud \u

    Malic Acid Distribution and Degradation in Grape Must During Skin Contact: The Influence of Recombinant Malo-Ethanolic Wine Yeast Strains

    Wine acidity plays an important role in determining wine quality and ensuring physiochemical and microbiological stability. In high-acid wines, the L-malic acid concentration is usually reduced through bacterial malolactic fermentation, while acidulation in low-acidity wines is usually done during final blending of the wine before bottling.  This study showed that skin contact did not influence the relative concentration of L-malic acid in the pulp and juice fractions from Colombard, Ruby Cabernet and Cabernet Sauvignon grape musts, with 32%-44% of the L-malic acid present in the pulp fraction. Four recombinant malo-ethanolic (ME) Saccharomyces wine yeast strains containing the malic enzyme (mae2) and malate transporter (mael) genes of Schizasaccharomyces pombe, effectively degraded the L-malic acid in both the juice and pulp fractions of all three cultivars, with a complete degradation of malic acid in the juice fraction within 2 days

    Genetic Engineering of an Industrial Strain of Saccharomyces cerevisiae for L-Malic Acid Degradation via an Efficient Malo-Ethanolic Pathway

    The optimal ratio of L-malic and L-tartaric acid in relation to other wine components is one of the most important aspects that ultimately determine wine quality during winemaking. Winemakers routinely rely on the judicious use of malolactic fermentation (MLF) after alcoholic fermentation to deacidify and stabilise their wines. However, due to theunreliability of the process and unsuitable sensory modifications in some grape cultivars, especially for fruity-floral wines, MLF is often regarded as problematic and undesirable. Alternative methods for reducing the amounts of L-malic acid in wine will contribute to improving the production of quality wines in the future, especially in coolclimate regions. Most wine yeast strains of Saccharomyces are unable to effectively degrade L-malic acid, whereas the fission yeast Schizosaccharomyces pombe efficiently degrades high concentrations of L-malic acid by means of malo-ethanolic fermentation. However, strains of S. pombe are not suitable for vinification due to the production of undesirable off-flavours. Previously, the 5. pombe malate permease (mael) and malic enzyme (mae2) genes were  successfully expressed under the 3-phosphoglycerate kinase (PGK1) regulatory elements in 5. cerevisiae, resulting in a recombinant laboratory strain of S. cerevisiae with an efficient malo-ethanolic pathway. Stable integration of the S. pombe malo-ethanolic pathway genes has now been obtained through the construction of a unique integration strategy in a commercial wine yeast strain. Co-transformation of the linear integration cassette containing the mael and mae2 genes and PGK1 regulatory elements and a multi-copy plasmid containing the phleomycin-resistance marker into a commercial Saccharomyces cerevisiae strain resulted in the successful transformation and integration of the malo-ethanolic genes. The recombinant 5. cerevisiae strain was successfully cured of phleomycin-resistance plasmid DNA in order to obtain malo-ethanolic yeast containing only yeast-derived DNA. The integrated malo-ethanolic genes were stable in 5. cerevisiae and during synthetic and grape must fermentation, L-malic acid was completely fermented to ethanol without any negative effect on fermentation kinetics and wine quality

    Strong time dependence of ocean acidification mitigation by atmospheric carbon dioxide removal

    In Paris in 2015, the global community agreed to limit global warming to well below 2 ∘C, aiming at even 1.5 ∘C. It is still uncertain whether these targets are sufficient to preserve marine ecosystems and prevent a severe alteration of marine biogeochemical cycles. Here, we show that stringent mitigation strategies consistent with the 1.5 ∘C scenario could, indeed, provoke a critical difference for the ocean’s carbon cycle and calcium carbonate saturation states. Favorable conditions for calcifying organisms like tropical corals and polar pteropods, both of major importance for large ecosystems, can only be maintained if CO2 emissions fall rapidly between 2025 and 2050, potentially requiring an early deployment of CO2 removal techniques in addition to drastic emissions reduction. Furthermore, this outcome can only be achieved if the terrestrial biosphere remains a carbon sink during the entire 21st century

    In The Netherlands a research programme has been initiated by the Ministry of Agriculture, Nature Management and Fisheries to increase the proportion of high-quality home-grown forage in rations for dairy cows. This should result in a more sustainable dairy husbandry by improving the efficiency of utilization of nitrogen and phosphorus from home-grown forages (mainly grass and maize) on farm level.     Abstract A subset X of the vertex set of a graph G is a secure dominating set of G if each vertex of G which is not in X is adjacent to some vertex in X and if, for each vertex u not in X, there is a neighbouring vertex v of u in X such that the swap set (X − {v}) ∪ {u} is again a dominating set of G. The secure domination number of G is the cardinality of a smallest secure dominating set of G. The notion of secure graph domination finds applications in the generic setting where the vertex set of G represents distributed locations in some spatial domain and the edges of G represent links between these locations. Patrolling guards, each stationed at one of these locations, may move along the links in order to protect the graph. A minimum secure dominating set of G then represents a smallest collection of locations at which guards may be stationed so that the entire location complex modelled by G is protected in the sense that if a security concern arises at location u, there will either be a guard stationed at that location who can deal with the problem, or else a guard dealing with the problem from an adjacent location v will still leave the location complex protected after moving from location v to location u in order to deal with the problem. A graph G is q-critical if the smallest arbitrary subset of edges whose removal from G necessarily increases the secure domination number, has cardinality q. The notion of qcriticality is important in applications such as the one mentioned above, because it provides threshold information as to the number of edge failures (perhaps caused by an adversary) that will necessitate the hiring of additional guards to secure the location complex. Denote by Ωn the largest value of q for which q-critical graphs of order n exist. It has previously been established that Ω2 = 1, Ω3 = 2, Ω4 = 4, Ω5 = 6 and Ω6 = 9. In this paper we present a repository of all q-critical graphs of orders 2, 3, 4, 5 and 6 for all admissible values of q and we also establish the previously unknown values Ω7 = 12, Ω8 = 17 and Ω9 = 23. These values support an existing conjecture that Ωn = n 2 − 2n + 5 for all n ≥ 7

    Evaluating the use of biomass energy with carbon capture and storage in low emission scenarios

    Biomass Energy with Carbon Capture and Storage (BECCS) is heavily relied upon in scenarios of future emissions that are consistent with limiting global mean temperature increase to 1.5 °C or 2 °C above pre-industrial. These temperature limits are defined in the Paris Agreement in order to reduce the risks and impacts of climate change. Here, we explore the use of BECCS technologies in a reference scenario and three low emission scenarios generated by an integrated assessment model (IMAGE). Using these scenarios we investigate the feasibility of key implicit and explicit assumptions about these BECCS technologies, including biomass resource, land use, CO2 storage capacity and carbon capture and storage (CCS) deployment rate. In these scenarios, we find that half of all global CO2 storage required by 2100 occurs in USA, Western Europe, China and India, which is compatible with current estimates of regional CO2 storage capacity. CCS deployment rates in the scenarios are very challenging compared to historical rates of fossil, renewable or nuclear technologies and are entirely dependent on stringent policy action to incentivise CCS. In the scenarios, half of the biomass resource is derived from agricultural and forestry residues and half from dedicated bioenergy crops grown on abandoned agricultural land and expansion into grasslands (i.e. land for forests and food production is protected). Poor governance of the sustainability of bioenergy crop production can significantly limit the amount of CO2 removed by BECCS, through soil carbon loss from direct and indirect land use change. Only one-third of the bioenergy crops are grown in regions associated with more developed governance frameworks. Overall, the scenarios in IMAGE are ambitious but consistent with current relevant literature with respect to assumed biomass resource, land use and CO2 storage capacity
