    Association of Single Nucleotide Polymorphisms in the IL-18 Gene with Production of IL-18 Protein by Mononuclear Cells from Healthy Donors

    IL-18 has proinflammatory effects and participates in both innate and adaptive cellular and humoral immunity. A number of SNPs that influence IL-18 production are found in the gene promoter region. We investigated the association of SNPs in the IL-18 promoter at āˆ’607 and āˆ’137 with the level of IL-18 protein production by PBMC from healthy donors from Southwestern Siberia. The genetic distribution of these SNPs in the promoter site was established by PCR. IL-18 protein production was determined by ELISA. Our results showed that PBMC from donors carrying allele 137C have lower levels of both spontaneous and LPS-stimulated IL-18 production. In contrast, PBMC from donors carrying allele 607A showed significant increases in spontaneous and stimulated IL-18 production compared to wild type. Our study suggests that the SNPs āˆ’607 and āˆ’137 in the promoter region of the IL-18 gene influence the level of IL-18 protein production by PBMC from healthy donors in Southwestern Siberia

    THE FREQUENCY OF ALLELIC VARIANTS OF GENES TNFRI AT POSITIONS -609 AND -1207 AND TNFRII AT POSITIONS -1709 AND -3609 IN HEALTHY DONORS AND PATIENTS WITH RHEUMATOID ARTHRITIS

    In this paper we studied the frequency of allelic variants of genes TNFRI at positions -609 and -1207 and TNFRII at positions -1709 and -3609 in patients with rheumatoid arthritis and in population control group. It was shown that the resulting frequency distribution of alleles and genotypes of TNFRI gene promoter in a group of population control is characteristic for Asian populations, for TNFRII gene polymorphisms and the frequency distribution differed from the European and Asian populations. Comparative analysis of allele and genotype frequencies of TNFRI gene promoter at positions -609 and -1207 and TNFRII and -3609 and positions -1709 in patients with rheumatoid arthritis and in population control group showed no statistically significant differences. However in the analysis of combinations has been shown that in patients with rheumatoid arthritis compared with the control population was significantly less common combination of genotypes TNFRI-609GT + TNFRII-3609CC

    Abstract. The subpopulation structure of immunocompetent cells of peripheral blood, concentration of IgA, IgM, IgG, IgE and serum levels of IL-4 and IL-10 for the patients was studied. Reduced contents of CD8+, CD56+ lymphocytes and increased contents of CD4+, CD22+ lymphocytes, serum levels of IgA, IgM, IgG, IgE are revealed in Hkakass with asthma compared to healthy Hkakass. It was also revealed in Hkakass with asthma that contents of CD3+ and CD8+ lymphocytes, serum concentration of IgA, IgE increased compared to Caucasoids with asthma. The serum level of cytokines IL-4 were found to be considerably increased in Hkakass with asthma compared to healthy Hkakass and to Caucasoids with asthma. On the other hand, the comparison of concentration of IL-10 showed that serum level was higher in Caucasoids with asthma than Hkakass with asthma
