    ICAM-1 Targeting of Doxorubicin-Loaded PLGA Nanoparticles to Lung Epithelial Cells

    Interaction of leukocyte function associated antigen-1 (LFA-1) on T-lymphoctytes and intercellular adhesion molecule-1 (ICAM-1) on epithelial cells controls leukocyte adhesion, spreading, and extravasation. This process plays an important role in leukocyte recruitment to a specific site of inflammation and has been indentified as a biomarker for certain types of carcinomas. Cyclo-(1,12)-PenITDGEATDSGC (cLABL) has been shown to inhibit LFA-1 and ICAM-1 interaction via binding to ICAM-1. In addition, cLABL has been shown to internalize after binding ICAM-1. The possibility of using cLABL conjugated nanoparticles (cLABL-NP) as a targeted and controlled release drug delivery system has been investigated in this study. The cLABL peptide was conjugated to a modified PluronicÂź surfactant on poly (DL-lactic-co-glycolic acid) (PLGA) nanoparticles. The cLABL-NP showed more rapid cellular uptake by A549 lung epithelial cells compared to nanoparticles without peptide. The specificity of ICAM-1 mediated internalization was confirmed by blocking the uptake of cLABL-NP to ICAM-1 using free cLABL peptide to block the binding of cLABL-NP to ICAM-1 on the cell surface. Cell studies suggested that cLABL-NPs targeted encapsulated doxorubicin to ICAM-1 expressing cells. Cytotoxicity assay confirmed the activity of the drug incorporated in nanoparticles. Sustained release of doxorubicin afforded by PLGA nanoparticles may enable cLABL-NP as a targeted, controlled release drug delivery system

    The Stellar Content of the Polar Rings in the Galaxies NGC 2685 and NGC 4650A

    We present the results of stellar photometry of polar-ring galaxies NGC 2685 and NGC 4650A, using the archival data obtained with the Hubble Space Telescope's Wide Field Planetary Camera 2. Polar rings of these galaxies were resolved into ~800 and ~430 stellar objects in the B, V and Ic bands, considerable part of which are blue supergiants located in the young stellar complexes. The stellar features in the CM-diagrams are best represented by isochrones with metallicity Z = 0.008. The process of star formation in the polar rings of both galaxies was continuous and the age of the youngest detected stars is about 9 Myr for NGC 2685 and 6.5 Myr for NGC 4650A.Comment: 21 pages, 9 figures, AJ 2004 February, accepte

    Morphine Exacerbates HIV-1 Tat-Induced Cytokine Production in Astrocytes through Convergent Effects on [Ca2+]i, NF-ÎșB Trafficking and Transcription

    Astroglia are key cellular sites where opiate drug signals converge with the proinflammatory effects of HIV-1 Tat signals to exacerbate HIV encephalitis. Despite this understanding, the molecular sites of convergence driving opiate-accelerated neuropathogenesis have not been deciphered. We therefore explored potential points of interaction between the signaling pathways initiated by HIV-1 Tat and opioids in striatal astrocytes. Profiling studies screening 152 transcription factors indicated that the nuclear factor-kappa B (NF-ÎșB) subunit, c-Rel, was a likely candidate for Tat or Tat plus opiate-induced increases in cytokine and chemokine production by astrocytes. Pretreatment with the NF-ÎșB inhibitor parthenolide provided evidence that Tat±morphine-induced release of MCP-1, IL-6 and TNF-α by astrocytes is NF-ÎșB dependent. The nuclear export inhibitor, leptomycin B, blocked the nucleocytoplasmic shuttling of NF-ÎșB; causing p65 (RelA) accumulation in the nucleus, and significantly attenuated cytokine production in Tat±morphine exposed astrocytes. Similarly, chelating intracellular calcium ([Ca2+]i) blocked Tat±morphine-evoked MCP-1 and IL-6 release, while artificially increasing the concentration of extracellular Ca2+ reversed this effect. Taken together, these results demonstrate that: 1) exposure to Tat±morphine is sufficient to activate NF-ÎșB and cytokine production, 2) the release of MCP-1 and IL-6 by Tat±morphine are highly Ca2+-dependent, while TNF-α appears to be less affected by the changes in [Ca2+]i, and 3) in the presence of Tat, exposure to opiates augments Tat-induced NF-ÎșB activation and cytokine release through a Ca2+-dependent pathway

    The Right Tool for the Job: Detection of Soil-Transmitted Helminths in Areas Co-endemic for Other Helminths

    Due to the recent increased use of the McMaster (MM) fecal egg counting method for assessing benzimidazole drug efficacy for treating soil-transmitted helminth (STH) infections, the aim of the current study was to determine the operational value of including the MM method alongside the Kato-Katz (KK) fecal thick smear to increase the diagnostic sensitivity when STHs are co-endemic with trematode helminths (e.g., Schistosoma mansoni). Methods A cross-sectional study was conducted in school-aged children aged 4-18 years in the northeastern region of the State of Minas Gerais (Brazil), where Necator americanus, Ascaris lumbricoides, Trichuris trichiura, and S. mansoni are co-endemic. One fecal sample from each participant was collected and transported to the field laboratory for analysis. Coprological diagnosis was performed on each fecal sample by three different methods: Formalin-Ether Sedimentation (FES), KK and the MM technique. The diagnostic sensitivity and negative predictive value (NPV) of each technique was calculated using the combination of all three techniques as the composite standard. In order to determine the agreement between the three techniques FleissÂŽ kappa was used. Both the Cure Rate (CR) and the Fecal Egg Count Reduction (FECR) were calculated using the two quantification techniques (i.e., the MM and KK). Results Fecal samples from 1260 children were analyzed. The KK had higher diagnostic sensitivity than the MM for the detection of both A. lumbricoides (KK 97.3%, MM 69.5%) and hookworm (KK 95.1%, MM 80.8%). The CR of a single dose of mebendazole varied significantly between the KK and MM for both A. lumbricoides (p = 0.016) and hookworm (p = 0.000), with lower rates obtained with the KK. On the other hand, the FECR was very similar between both techniques for both A. lumbricoides and hookworm. Conclusion The MM did not add any diagnostic value over the KK in areas where both STHs and trematodes were co-endemic. The lower sensitivity of the MM would have an important impact on the administration of selective school-based treatment in this area since if only the MM were used, 36 (13.9%) children diagnosed with A. lumbricoides would have gone untreated. Author Summary Diagnosis of intestinal helminths and Schistosoma mansoni infections is based on the detection of eggs in feces. There are many techniques available for both detection and quantification of infection. For the quantification of helminth infections, the methods traditionally used are the Kato-Katz (KK) fecal think smear in humans, and the McMaster (MM) counting method in animals. Recently, the MM has been used for assessing the efficacy of benzimidazole drugs for treating soil-transmitted helminth (STH) infections in humans. In most parts of the world, however, STHs occur simultaneously with other helminth species, and the MM does not detect other helminth eggs. Therefore, in this study we sought to determine if the use of the MM in an area of Brazil were both STHs and S. mansoni are co-endemic, added any value to the current standard of diagnosis using the KK

    Study of the structure and kinematics of the NGC 7465/64/63 triplet galaxies

    This paper is devoted to the analysis of new observational data for the group of galaxies NGC 7465/64/63, which were obtained at the 6-m telescope of the Special Astrophysical Observatory of the Russian Academy of Sciences (SAO RAS) with the multimode instrument SCORPIO and the Multi Pupil Fiber Spectrograph. For one of group members (NGC 7465) the presence of a polar ring was suspected. Large-scale brightness distributions, velocity and velocity dispersion fields of the ionized gas for all three galaxies as well as line-of-sight velocity curves on the basis of emission and absorption lines and a stellar velocity field in the central region for NGC 7465 were constructed. As a result of the analysis of the obtained information, we revealed an inner stellar disk (r ~ 0.5 kpc) and a warped gaseous disk in addition to the main stellar disk, in NGC 7465. On the basis of the joint study of photometric and spectral data it was ascertained that NGC 7464 is the irregular galaxy of the IrrI type, whose structural and kinematic peculiarities resulted most likely from the gravitational interaction with NGC 7465. The velocity field of the ionized gas of NGC 7463 turned out typical for spiral galaxies with a bar, and the bending of outer parts of its disk could arise owing to the close encounter with one of galaxies of the environment.Comment: 20 pages, 6 figure

    NGC 7468: a galaxy with an inner polar disk

    We present our spectroscopic observations of the galaxy NGC 7468 performed at the 6-m Special Astrophysical Observatory telescope using the UAGS long-slit spectrograph, the multipupil fiber spectrograph MPFS, and the scanning Fabry-Perot interferometer (IFP). We found no significant deviations from the circular rotation of the galactic disk in the velocity field in the regions of brightness excess along the major axis of the galaxy (the putative polar ring). Thus, these features are either tidal structures or weakly developed spiral arms. However, we detected a gaseous disk at the center of the galaxy whose rotation plane is almost perpendicular to the plane of the galactic disk. The central collision of NGC 7468 with a gas-rich dwarf galaxy and their subsequent merging seem to be responsible for the formation of this disk.Comment: 8 pages, 6 figures, accepted in Astronomy Letters, 2004, vol 30., N 9, p. 58
