Are Asian elephants afraid of honeybees? Experimental studies in northern Thailand
This research article published Springer Nature Switzerland AG., 2020In many parts of South and Southeast Asia, rural farmers living at the borders of protected areas frequently encounter Asian elephants (Elephas maximus) raiding their crops and threatening farmers lives and livelihoods. Traditional deterrent methods often have limited success as elephants become habituated or alternate their movement and behavior. While African bees (Apis mellifera scutellate) have been shown to effectively and sustainably deter African elephants (Loxodonta africana) little is known about their Asian counterparts. We conducted two experiments to estimate the effectiveness of bees as an Asian elephant deterrent method. We analyzed the behavioral reaction of seven captive Asian elephants when confronted with a fence of A. mellifera hives blocking their way to a desired source of food. In addition, we explored the defensive reaction of five A. cerana hives and six A. mellifera hives to an artificial disturbance during both day and night time. The elephants crossed the beehive fence in 51% of the cases, the probability of crossing increased over time and the number of exposures had a significant effect on an elephantâs crossing probability, indicating that elephants became habituated to the presence of the beehive fence. In the bee experiment, only one out of five A. cerana hives and one out of six A. mellifera hives reacted to the disturbance during the daytime, while during nighttime, none of them reacted defensively after being disturbed. We, therefore, conclude that neither A. mellifera nor A. cerana bees are likely to be effective in deterring wild Asian elephants from entering crop fields

particle tracking techniques. From these data, the velocity profiles can be
obtained and the validity of some long-standing approaches can be assessed.

Prefrontal Cortex Glutamate Correlates with Mental Perspective-Taking
Background: Dysfunctions in theory of mind and empathic abilities have been suggested as core symptoms in major psychiatric disorders including schizophrenia and autism. Since self monitoring, perspective taking and empathy have been linked to prefrontal (PFC) and anterior cingulate cortex (ACC) function, neurotransmitter variations in these areas may account for normal and pathological variations of these functions. Converging evidence indicates an essential role of glutamatergic neurotransmission in psychiatric diseases with pronounced deficits in empathy. However, the role of the glutamate system for different dimensions of empathy has not been investigated so far. Methodology/Principal Findings: Absolute concentrations of cerebral glutamate in the ACC, left dorsolateral PFC and left hippocampus were determined by 3-tesla proton magnetic resonance spectroscopy (1H-MRS) in 17 healthy individuals. Three dimensions of empathy were estimated by a self-rating questionnaire, the Interpersonal Reactivity Index (IRI). Linear regression analysis showed that dorsolateral PFC glutamate concentration was predicted by IRI factor ââperspective takingââ (T = 22.710, p = 0.018; adjusted alpha-level of 0.017, Bonferroni) but not by ââempathic concernâ â or ââpersonal distressââ. No significant relationship between IRI subscores and the glutamate levels in the ACC or left hippocampus was detected. Conclusions/Significance: This is the first study to investigate the role of the glutamate system for dimensions of theory of mind and empathy. UV photochemistry of the L-cystine disulfide bridge in aqueous solution investigated by femtosecond X-ray absorption spectroscopy
The photolysis of disulfide bonds is implicated in denaturation of proteins exposed to ultraviolet light. Despite this biological relevance in stabilizing the structure of many proteins, the mechanisms of disulfide photolysis are still contested after decades of research. Herein, we report new insight into the photochemistry of L-cystine in aqueous solution by femtosecond X-ray absorption spectroscopy at the sulfur K-edge. We observe homolytic bond cleavage upon ultraviolet irradiation and the formation of thiyl radicals as the single primary photoproduct. Ultrafast thiyl decay due to geminate recombination proceeds at a quantum yield of >80 % within 20âps. These dynamics coincide with the emergence of a secondary product, attributed to the generation of perthiyl radicals. From these findings, we suggest a mechanism of perthiyl radical generation from a vibrationally excited parent molecule that asymmetrically fragments along a carbon-sulfur bond. From L-Dopa to Dihydroxyphenylacetaldehyde: A Toxic Biochemical Pathway Plays a Vital Physiological Function in Insects
One protein in Aedes aegypti, classified into the aromatic amino acid decarboxylase (AAAD) family based on extremely high sequence homology (âŒ70%) with dopa decarboxylase (Ddc), was biochemically investigated. Our data revealed that this predicted AAAD protein use L-dopa as a substrate, as does Ddc, but it catalyzes the production of 3,4-dihydroxylphenylacetaldehyde (DHPAA) directly from L-dopa and apparently has nothing to do with the production of any aromatic amine. The protein is therefore named DHPAA synthase. This subsequently led to the identification of the same enzyme in Drosophila melanogaster, Anopheles gambiae and Culex quinquefasciatus by an initial prediction of putative DHPAA synthase based on sequence homology and subsequent verification of DHPAA synthase identity through protein expression and activity assays. DHPAA is highly toxic because its aldehyde group readily reacts with the primary amino groups of proteins, leading to protein crosslinking and inactivation. It has previously been demonstrated by several research groups that Drosophila DHPAA synthase was expressed in tissues that produce cuticle materials and apparent defects in regions of colorless, flexible cuticular structures have been observed in its gene mutants. The presence of free amino groups in proteins, the high reactivity of DHPAA with the free amino groups, and the genetically ascertained function of the Drosophila DHPAA synthase in the formation of colorless, flexible cuticle, when taken together, suggest that mosquito and Drosophila DHPAA synthases are involved in the formation of flexible cuticle through their reactive DHPAA-mediated protein crosslinking reactions. Our data illustrate how a seemingly highly toxic pathway can serve for an important physiological function in insects
