Glucose or Sucrose Intakes and Plasma Levels of Essential and Nonessential Amino Acids
It is not known whether the administration of glucose or sucrose influences plasma levels of amino acids. We want to know whether plasma levels of amino acids and if the administration of glucose or sucrose are different in young and old men and are influenced by the administration of glucose or sucrose. We found that the levels of most amino acids in plasma are lower in old men than young men. When sucrose was administered to old men, levels of total amino acids decreased significantly in old men. In both old and young men plasma levels of total nonessential amino acids significantly decreased at 120 min. after the administration of glucose but not sucrose. On the other hand, total essential and branched amino acids levels decreased significantly after the administration of both glucose and sucrose in young and old men. From these results, responses to the administration of glucose were different from the response to sucrose between young and old men. Also glucose was more effective in decreasing plasma levels of various amino acids. These results seem to suggest that glucose was more effective in stimulating insulin release and young men were more responsive to sugar than old men in stimulating insulin release
Development of Methods to Evaluate Several Levels of Uranium Concentrations in Drainage Water Using Total Reflection X-Ray Fluorescence Technique
As a country's law stipulates the effluent standard uranium concentration in drainage water, the uranium concentration must be determined when drainage water is released from a uranium handling facility, such as the Fukushima Daiichi nuclear power plant. The maximum allowable limit for uranium release at each facility is defined taking into consideration the situation of the facility, such as 1/10 to 1/100 of this effluent standard value. Currently, the uranium concentration of drainage water is commonly determined by α-particle spectrometry, in which several liters of drainage water must be evaporated, requiring about half of a day followed by 2–3 h of measurements, due to the low specific radioactivity of uranium. This work proposes a new methodology for the rapid and simple measurement of several levels of uranium in drainage water by a total reflection X-ray fluorescence (TXRF) analysis. Using a portable device for TXRF measurements was found to enable measurements with 1/10 the sensitivity of the effluent standard value by 10 times condensation of the uranium-containing sample solution; a benchtop device is useful to measure uranium concentrations <1/100 of the effluent standard value. Therefore, the selective usage of methods by a portable and benchtop devices allows for screening and precise evaluation of uranium concentrations in drainage water
Roles of Glucose and Sucrose Intakes on the Brain Functions Measured by the Working Ability and Morris Maze
Sugars such as glucose or sucrose are considered hazardous foods because their intakes lead to obesity, further causing diabetes mellitus (DM), or cardiovascular diseases. However, glucose is needed for many brain functions such as memory and emotion among others. Glucose induces the secretion of insulin, which is needed for transportation of tryptophan from the blood to the brain. Serotonin, which is converted from tryptophan, is important for mood stability, control of emotion, and feeding is inhibited by serotonin in the hypothalamus. We discuss transportation of glucose from the blood to the glia cells. After glycolysis of glucose in the glia lactic acid is transported to cells such as glutaminergic neurons. After the release from neurons glutamic acid is taken up into glia cells and further to neurons again. Sucrose is degraded into glucose and fructose in the intestine thus intake of sucrose increases plasma levels of glucose. We show that intake of sucrose enhanced memory measured by Morris maze in rats and improved the working ability in humans. Roles of glucose and sucrose intakes are discussed together with the function of serotonin in feeding
Food Intakes and Correlations between Food Intakes and Body Mass Index (BMI) in Japanese Old Men, Women, and Male Medical Doctors
Objective; Obesity is an important health problem, leading to many metabolic diseases such as type2 diabetes mellitus, cardiovascular diseases, cancer. The are many diet proposals to combat obesity. Since obesity is relatively rare in Japan, we wante to know what kind of foods influence body mass index (BMI) in old Japanese people. METHODS; Healthy participants, old men and women and male medical doctors (MD) were given self-administered diet history questionnaires and described answers on each item by recollection of diets they took (7 days dietary recall). We used a brief-type self-administered diet history questionnaire (BDHQ) by using which the Japanese Ministry of Health, Labour and Welfare reports national Nutrition Surveys. From these questionnaires, we calculated the intakes of energy, carbohydrate, fat, protein or other foods. RESULTS; Me take more alcohol, salt fruit, beans than women. Intakes of major foods such as carbohydrate, lipid, and protein did not influence BMI in men and women. MD with higher BMI tend to take vegetables and fruits. MD may be more health concerned than lay people. CONCLUSION; within the range of foods intakes in Japan, no restriction of any food such as carbohydrate is not necessary for staying lean. Medical doctors seem to be very health concerned compared to lay people
Impact of CRAB symptoms in survival of patients with symptomatic myeloma in novel agent era
The acronym CRAB summarizes the most typical clinical manifestations of multiple myeloma, these being hypercalcemia, renal failure, anemia, and bone disease. CRAB can be used to distinguish between active, symptomatic multiple myeloma and monoclonal gammopathy of undermined significance or smoldering myeloma. The distinction is relevant not only for classification and diagnosis but also for therapy. CRAB factors influence the prognosis of multiple myeloma. However, it is unclear whether the presence of CRAB factors has an influence on the prognosis of myeloma treated with novel agents. In the current study, patients with hypercalcemia and bone disease showed a significantly worse prognosis, whereas anemia and renal failure showed no difference in survival. Novel agents used for treatment of patients with renal failure suggested a favorable outcome compared with conventional therapy. Bone disease was the most common factor and may have the strongest prognostic value in symptomatic myeloma patients using novel agents
Clinical significance of dasatinib-induced pleural effusion in patients with de novo chronic myeloid leukemia
Dasatinib is currently approved for clinical use as a first-line treatment agent for newly diagnosed chronic myeloid leukemia (CML). However, only a few clinical trials have been performed to evaluate dasatinibinduced PE following first-line therapy. We investigated the incidence and clinical features of dasatinib-induced PE following first-line therapy in Japanese CML patients of real world clinical practice settings. Among 22 patients, the median age of PEpositive patients was higher than that of PEnegative patients. Major molecular response was achieved in 75% of PE-positive patients and 50% of PE-negative patients. Most patients developed PE more than 1 year after treatment. Appearance of PE is associated with better clinical response during dasatinib treatment, however it is developed at any time. Elderly and high-risk patients tend to develop PE. The clinical features of dasatinib-induced PE following first-line therapy might be late onset and might not immediately follow the increasing of large granular lymphocyte
SOX9 is a novel cancer stem cell marker surrogated by osteopontin in human hepatocellular carcinoma
The current lack of cancer stem cell (CSC) markers that are easily evaluated by blood samples prevents the establishment of new therapeutic strategies in hepatocellular carcinoma (HCC). Herein, we examined whether sex determining region Y-box 9 (SOX9) represents a new CSC marker, and whether osteopontin (OPN) can be used as a surrogate marker of SOX9 in HCC. In HCC cell lines transfected with a SOX9 promoter-driven enhanced green fluorescence protein gene, FACS-isolated SOX9+ cells were capable of self-renewal and differentiation into SOX9-cells, and displayed high proliferation capacity in vitro. Xenotransplantation experiments revealed that SOX9+ cells reproduced, differentiated into SOX9-cells, and generated tumors at a high frequency in vivo. Moreover, SOX9+ cells were found to be involved in epithelial-mesenchymal transition (EMT) and activation of TGFb/Smad signaling. Gain/loss of function experiments showed that SOX9 regulates Wnt/beta-catenin signaling, including cyclin D1 and OPN. Immunohistochemistry of 166 HCC surgical specimens and serum OPN measurements showed that compared to SOX9-patients, SOX9+ patients had significantly poorer recurrence-free survival, stronger venous invasion, and higher serum OPN levels. In conclusion, SOX9 is a novel HCCCSC marker regulating the Wnt/beta-catenin pathway and its downstream target, OPN. OPN is a useful surrogate marker of SOX9 in HCC
