    What burned the forest? Wildfires, climate change and human activity in the Mesolithic – Neolithic transition in SE Iberian Peninsula

    Climate variability such as higher or lower temperatures, changing precipitation patterns, shifts in plant communities and other climate-related changes have particularly affected areas with Mediterranean-type climates. A multi-proxy analysis including pollen, sedimentary charcoal, mineralogy and Summed Probability Distributions (SPD) of archaeological 14C dates, allowed the reconstruction of landscape change, geomorphological evolution and fire history at the Laguna de Villena, in the southeastern Iberian Peninsula, during the Mesolithic-Neolithic transition. The Villena paleolake was sensitive to Early Holocene and Middle Holocene regional climatic variability that included several arid phases (around 8.2 ka cal BP, 6.8 ka cal BP and 5.9 ka cal BP) according to geochemical and pollen data. During this period, landscape dynamics show the degradation of oak forests and expansion of pyrophytic pine forests and shrublands, as well as open spaces predominated by grasses. The charcoal record shows a decreasing trend of biomass burned from 8.5 ka cal BP onwards, although fire peaks occurred recurrently during the Early and Middle Holocene. The most intense phase of fire activity was experienced in the last millennia of the Early Holocene, with five fire episodes from 9.1 to 8.4 ka cal BP, coinciding with a phase of higher archaeological evidence in the area. A decrease in archaeological evidence coincides with a gap in fire episodes during the Early Holocene-Middle Holocene transition, suggesting an effect of the abrupt 8.2 ka cal BP event on human activity and on landscape dynamics. After 8.0 ka cal BP, lowerer biomass burned is explained by the configuration of a more open landscape due to the combination of climate (increasing aridity) and increasing human activities in the region. The mineralogical and palynological data highlighted the interaction between human activities, climate and fire dynamics. The sedimentary charcoal record evidenced how most of the fire peaks did not occur in the context of dry episodes, as often assumed, deriving on an anthropogenic explanation related to Early and Late Mesolithic burning practices during a phase of higher archaeological evidence in the Villena paleolake surroundings. Afterwards, combined agropastoral activities from the Early Neolithic onwards and increasing aridity during the Middle Holocene maintained the forest clearances, in the context of fire episodes characterised by decreased biomass burned. This study shows how Middle Holocene palaeoecological records reflect complex histories blending climate and anthropogenic processes that derived in major landscape changes explaining the origin of current landscapes.This research has received funding from the European Research Council (ERC) under the Horizon 2020 research and innovation programme (Grant agreement No. 683018) to JFLdP.     The polygenic basis of relapse after a first episode of schizophrenia

    Little is known about genetic predisposition to relapse. Previous studies have linked cognitive and psychopathological (mainly schizophrenia and bipolar disorder) polygenic risk scores (PRS) with clinical manifestations of the disease. This study aims to explore the potential role of PRS from major mental disorders and cognition on schizophrenia relapse. 114 patients recruited in the 2EPs Project were included (56 patients who had not experienced relapse after 3 years of enrollment and 58 patients who relapsed during the 3-year follow-up). PRS for schizophrenia (PRS-SZ), bipolar disorder (PRS-BD), education attainment (PRS-EA) and cognitive performance (PRS-CP) were used to assess the genetic risk of schizophrenia relapse.Patients with higher PRS-EA, showed both a lower risk (OR=0.29, 95% CI [0.11–0.73]) and a later onset of relapse (30.96± 1.74 vs. 23.12± 1.14 months, p=0.007. Our study provides evidence that the genetic burden of neurocognitive function is a potentially predictors of relapse that could be incorporated into future risk prediction models. Moreover, appropriate treatments for cognitive symptoms appear to be important for improving the long-term clinical outcome of relapse

    Involvement of NRN1 gene in schizophrenia-spectrum and bipolar disorders and its impact on age at onset and cognitive functioning

    Objectives Neuritin 1 gene (NRN1) is involved in neurodevelopment processes and synaptic plasticity and its expression is regulated by brain-derived neurotrophic factor (BDNF). We aimed to investigate the association of NRN1 with schizophrenia-spectrum disorders (SSD) and bipolar disorders (BPD), to explore its role in age at onset and cognitive functioning, and to test the epistasis between NRN1 and BDNF. Methods The study was developed in a sample of 954 SSD/BPD patients and 668 healthy subjects. Genotyping analyses included 11 SNPs in NRN1 and one functional SNP in BDNF. Results The frequency of the haplotype C-C (rs645649-rs582262) was significantly increased in patients compared to controls (P = 0.0043), while the haplotype T-C-C-T-C-A (rs3763180-rs10484320-rs4960155-rs9379002-rs9405890-rs1475157) was more frequent in controls (P = 3.1 × 10-5). The variability at NRN1 was nominally related to changes in age at onset and to differences in intelligence quotient, in SSD patients. Epistasis between NRN1 and BDNF was significantly associated with the risk for SSD/BPD (P = 0.005). Conclusions Results suggest that: (i) NRN1 variability is a shared risk factor for both SSD and BPD, (ii) NRN1 may have a selective impact on age at onset and intelligence in SSD, and (iii) the role of NRN1 seems to be not independent of BDNF

Of 1045 adults collected, 16% proved positive for viral polyhedrin gene transcripts by RT-PCR. The prevalence of covert infection varied significantly according to sex and sample date. Of 1660 progeny of field-collected insects, lethal disease was observed in 10-33% of offspring of transcript-positive females and 9-49% of offspring of transcript-negative females. Isolates associated with vertically-transmitted infections were characterized by restriction endonuclease analysis using BglII and compared with isolates believed to be horizontally-transmitted. Insects from a sublethally-infected Almerian colony were ~3.4-fold more susceptible to infection than healthy insects from a Swiss colony. Horizontally-transmitted isolates were significantly more pathogenic than vertically-transmitted isolates in insects from both colonies. Mean speed of kill varied between isolates by >20 h, whereas mean occlusion body (OB) production varied by 3.8-fold among isolates. Intriguingly, all three horizontally transmitted isolates were very similar in speed of kill and OB production, whereas all three vertically transmitted isolates differed significantly from one another in both variables, and also differed significantly from the group of horizontally-transmitted isolates in one or both variables. We conclude that key pathogenicity and virulence traits of SeMNPV isolates vary according to their principal transmission strategy.Peer Reviewe

    Gene co-expression architecture in peripheral blood in a cohort of remitted first-episode schizophrenia patients

    A better understanding of schizophrenia subtypes is necessary to stratify the patients according to clinical attributes. To explore the genomic architecture of schizophrenia symptomatology, we analyzed blood co-expression modules and their association with clinical data from patients in remission after a first episode of schizophrenia. In total, 91 participants of the 2EPS project were included. Gene expression was assessed using the Clariom S Human Array. Weighted-gene co-expression network analysis (WGCNA) was applied to identify modules of co-expressed genes and to test its correlation with global functioning, clinical symptomatology, and premorbid adjustment. Among the 25 modules identified, six modules were significantly correlated with clinical data. These modules could be clustered in two groups according to their correlation with clinical data. Hub genes in each group showing overlap with risk genes for schizophrenia were enriched in biological processes related to metabolic processes, regulation of gene expression, cellular localization and protein transport, immune processes, and neurotrophin pathways. Our results indicate that modules with significant associations with clinical data showed overlap with gene sets previously identified in differential gene-expression analysis in brain, indicating that peripheral tissues could reveal pathogenic mechanisms. Hub genes involved in these modules revealed multiple signaling pathways previously related to schizophrenia, which may represent the complex interplay in the pathological mechanisms behind the disease. These genes could represent potential targets for the development of peripheral biomarkers underlying illness traits in clinical remission stages after a first episode of schizophrenia
