PATHOGEN IMPACT ON THE ACTIVITY DYNAMICS OF POTATO SUSPENSION CELLS EXTRA-CELLULAR PEROXIDASE

    Get PDF
Changes in the activity of extracellular peroxidases were measured in cell suspension cultures of potato infected by Clavibacter michiganensis subsp. sepedonicus (Spieck. et Kotth.) Skapt et Burkh. The total extracellular peroxidases activity of the resistant potato variety was higher than that of the sensitive variety both before and after infection. The enzyme of the resistant variety had a рН optimum of 6.2, while that of the sensitive variety was 5.4. Extracellular peroxidases of the sensitive potato variety were activated 10 minutes after infection, and displayed highest activity 1.5-2 hours later. In the resistant variety, peroxidase activity rose sharply in the first minutes of infection, and second peak of activity occurred 1.5-2 hours later. The increase of extracellular peroxidases activity of the sensitive potato variety under pathogenesis is connected with the change of genome expression and synthesis of proteins. The increase of enzyme activity of resistant potato variety in the first moments of infection is not related to proteins synthesis and is apparently conditioned by the change of kinetic parameters

Water and carbohydrate content at leafs of plants used in medicine during vegetation

    Get PDF
Increase of carbohydrate content with cryoprotective function to the end of vegetation period was shown. The accumulation of carbohydrates in plants on Lake Baikal shores region was greater than it in Irkutsk region

Activity and isoenzyme spectrum of peroxidases and dehydrins of some plant species, growing on the shores of lake Baikal, under abiotic stress

Termostability and optimal pH of weak-associated with plant cell wall and soluble peroxidases was shown to change in relation to natural conditions and season of year. Also the activity of peroxidase was variable during vegetation period. Dehydrine expression was followed by spike of peroxidase activity (and, a priori, an increase of hydrogen peroxide concentration)

THE CHANGE OF KINETIK PARAMETERS OF THE WEAK-ASSOCIATED WITH WALL CELL PEROXIDASE IN THE SUSPENSION CULTURE OF POTATO CELLS IN THE BEGINNING OF INFECTION

The change in kinetic parameters of extracellular peroxidase of suspension cells of resistant potato variety (Lugovskoi) and sensitive variety (Luk,ynovskii) in the initial period of infection by 5369 Clavibacter michiganensis subsp. sepedonicus (Spieck. et Kotth.) Skapt et Burkh. pathogen was examined. Extracellular peroxidases of resistant and sensitive potato variety without pathogens were shown to be concurrently inhibited. At the beginning of infection enzyme activity was extremely increased due to enhanced affinity to substrate as a result of reducing of competitive inhibiting. In increasing enzyme activity is sensitive potato variant evidently caused by other mechanism

EFFECT OF CONIFEROUS EXTRACT ON POTATO PLANTS

Low doses of extracts from conifer trees, such as Novosil and Lariksin, increased the complex resistance of potato plants to diseases in vegetation period. Also they act as growth stimulation factors and promote the growth yield

For the first time, the guaiacol-dependent peroxidase activities have been studied by different pH in lamina of Plantago major L

The Composition of Fatty Acids of Pinus sylvestris L. of Olkha Village Surrounding Forests

Palmitic, linoleic and α-linolenic acids are the major fatty acids during the study. Analyzing seasonal dynamics derived fatty acids revealed that for acids with 18 carbon atoms are characterized by virtually similar dynamics during the year in all years. Investigation of composition of fatty acids in the needles of different ages throughout the growing season revealed that in different periods of vegetation dominated by those or other fatty acids

Seasonal Dynamics of Stress Proteins in Leaves of Medicinal Plants in a Natural Environment of Irkutsk and on the Shores of the Lake Baikal

We study leafs of five plant species, growing in Irkutsk city and on the southeastern shore of Lake Baikal. These species are Achillea asiatica Serg., Taraxacum officinale Wigg., Plantago major L., Veronica chamaedrys L. and Alchemilla subcrenata Buser. In its leafs we identify some types of stress-induced proteins. In autumn, the accumulation of stress proteins in leafs of plants both from shores of Lake Baikal and from Irkutsk have been registered

Synthesis on the basis of natural matrices in order to acquire products with the desired properties is one of the promising trends of modern science. Using polysaccharides as a matrix allowed to generate derivatives with diverse structures and new properties. Growing interest towards anti-microbe effect of selenium-containing nano-composites is induced by the phenomenon of antibiotic-resistance of contemporary pathogenic microorganisms.Clavibacter genus bacteria are the most significant and widely spread among gram-positive bacteria. Bacteria cells are static pleimorphous rods, normally singular, sometimes coupled or joined in short chains, strict anaerobes in need of certain growth factors, non-sporogenous. Clavibacter michiganensis subsp. sepedonicus cause potato ring rot. At the tuber slice the damage is shaped as a ring; growing bacteria are accumulated in the conducting vessels causing their occlusion and therefore gradual withering of leaves and stem. This disease is distributed at all the continents including Australia. Harvest loss through ring rot damage may reach 10-45%.Our work was aimed at the study of complex interaction between microbe cultivar and selenium-based nanocomposites. Bacterial strain Аs1405 was acquired from the All-Russia collection of microorganisms, IMBP RAS. This genus is not included in the classification of pathogenic microorganisms by pathogenic groups of Sanitary-Epidemiological Rules SP 1.3.2322-08. The present study was focused on characteristics of the acquired strain.Fluorescent and electronic-scanning microscope was used to acquire photographs of bacterial cells. Pathogen was identified by PCR-analysis, which confirmed the presence of DNA of desired size. The extracted DNA was sequenced with the sequenced sequence added to Gen Bank under the number HQ394204. Cellulolytic and phytotoxic activity of this strain was determined.Chemistry Institute named A.E. Favorsky provided water-soluble nano-composites containing selenium stabilized by various polymers. Nano-composites anti-microbe activity was studied on the investigated strain of potato ring rot. Nano-composites of elementary selenium (3.4% Se) and Se with arabinogalactan acquired from SeO2 (1.23% Se) were found to demonstrate anti-microbe effect increasing with the rise of selenium content. The work enumerates various conditions and time periods of cultivation and determination of the influence of the given water-soluble nano-composites on bacterial cells survivability

The objects of investigation were leaves and inflorescences of Alchemilla subcrenata and Veronica chamaedry, growing on the left bank at 700 m from the edge of Lake Baikal. Some small differences in the quantity and quality of phenolic compounds in the leaves of one species even selected at one time were found. At the same time, differences in the quantity and quality of phenolic compounds in the different times of the day were even more pronounced. In the inflorescences of Alchemilla subcrenata the dynamics of flavonoids has been characterized by more stable composition than in its leaves. For Veronica's tissues and organs more variety of seasonal cocktail of phenolic compounds than in the Alchemilla subcrenata was detected. Expect a variety of phenolic compounds in inflorescences of Veronica seasonal cocktail was higher than in the leaves
