    Masked millennial-scale climate variations in South West Africa during the last glaciation

    To address the connection between tropical African vegetation development and high-latitude climate change we present a high-resolution pollen record from ODP Site 1078 (off Angola) covering the period 50–10 ka BP. Although several tropical African vegetation and climate reconstructions indicate an impact of Heinrich Stadials (HSs) in Southern Hemisphere Africa, our vegetation record shows no response. Model simulations conducted with an Earth System Model of Intermediate Complexity including a dynamical vegetation component provide one possible explanation. Because both precipitation and evaporation increased during HSs and their effects nearly cancelled each other, there was a negligible change in moisture supply. Consequently, the resulting climatic response to HSs might have been too weak to noticeably affect the vegetation composition in the study area. Our results also show that the response to HSs in southern tropical Africa neither equals nor mirrors the response to abrupt climate change in northern Africa

    CACHE (Critical Assessment of Computational Hit-finding Experiments): A public–private partnership benchmarking initiative to enable the development of computational methods for hit-finding

    One aspirational goal of computational chemistry is to predict potent and drug-like binders for any protein, such that only those that bind are synthesized. In this Roadmap, we describe the launch of Critical Assessment of Computational Hit-finding Experiments (CACHE), a public benchmarking project to compare and improve small-molecule hit-finding algorithms through cycles of prediction and experimental testing. Participants will predict small-molecule binders for new and biologically relevant protein targets representing different prediction scenarios. Predicted compounds will be tested rigorously in an experimental hub, and all predicted binders as well as all experimental screening data, including the chemical structures of experimentally tested compounds, will be made publicly available and not subject to any intellectual property restrictions. The ability of a range of computational approaches to find novel binders will be evaluated, compared and openly published. CACHE will launch three new benchmarking exercises every year. The outcomes will be better prediction methods, new small-molecule binders for target proteins of importance for fundamental biology or drug discovery and a major technological step towards achieving the goal of Target 2035, a global initiative to identify pharmacological probes for all human proteins.     Spatio-temporal, optogenetic control of gene expression in organoids

    Organoids derived from stem cells become increasingly important to study human development and to model disease. However, methods are needed to control and study spatio-temporal patterns of gene expression in organoids. To this aim, we combined optogenetics and gene perturbation technologies to activate or knock-down RNA of target genes, at single-cell resolution and in programmable spatio-temporal patterns. To illustrate the usefulness of our approach, we locally activated Sonic Hedgehog (SHH) signaling in an organoid model for human neurodevelopment. High-resolution spatial transcriptomic and single-cell analyses showed that this local induction was sufficient to generate stereotypically patterned organoids in three dimensions and revealed new insights into SHH's contribution to gene regulation in neurodevelopment. With this study, we propose optogenetic perturbations in combination with spatial transcriptomics as a powerful technology to reprogram and study cell fates and tissue patterning in organoids

    Seit dem Jahr 2000 haben sich die Strompreise für private Haushalte praktisch verdoppelt. Von steigenden Stromkosten sind nicht zuletzt Millionen von armutsgefährdeten Haushalten betroffen. Vor diesem Hintergrund untersucht der vorliegende Beitrag exemplarisch für einige Haushaltstypen mit geringem Einkommen, wie stark ihre Stromkostenbelastung in den vergangenen Jahren relativ zum Einkommen zugenommen hat und in welchem Maße diese Belastung künftig weiter steigen könnte. Nach unseren Abschätzungen auf Basis stilisierter Fakten musste beispielsweise der von uns betrachtete armutsgefährdete alleinstehende Rentner im Jahr 2013 etwa gleich viel für Strom ausgeben wie zur Deckung seines Energiebedarfs zum Heizen und zur Warmwassererzeugung. Es muss davon ausgegangen werden, dass aufgrund des Ausbaus der erneuerbaren Energien die Stromkostenbelastung auch in den nächsten Jahren weiter steigen wird. Damit stellt sich immer drängender die Frage nach Maßnahmen zur Abschwächung der Entwicklung steigender Strompreise und zur sozialen Abfederung ihrer regressiven Wirkungen

    We analyse the strength of CP violation in an extension of the standard model with an extra Q=1/3Q=-1/3 vector-like singlet quark, in the framework of the hypothesis of universality of strength of Yukawa couplings connecting standard quarks.     Bilateral Multi-Electrode Neurophysiological Recordings Coupled to Local Pharmacology in Awake Songbirds

    Here we describe a protocol for bilateral multielectrode neurophysiological recordings during intracerebral pharmacological manipulations in awake songbirds. This protocol encompasses fitting adult animals with head-posts and recording chambers, and acclimating them to periods of restraint. The adaptation period is followed by bilateral penetrations of multiple electrodes to obtain acute, sensory-driven neurophysiological responses before versus during the application of pharmacological agents of interest. These local manipulations are achieved by simultaneous and restricted drug infusions carried out independently for each hemisphere. We have used this protocol to elucidate how neurotransmitter and neuroendocrine systems shape the auditory and perceptual processing of natural, learned communication signals. However, this protocol can be used to explore the neurochemical basis of sensory processing in other small vertebrates.     Differential impact of chronic stress along the hippocampal dorsal–ventral axis

    First published online 06 February 2014Stress impacts differently in distinct brain regions. However, so far few studies have focused on the differential responses triggered by stressful stimuli on the intrinsic functional heterogeneity of the hippocampal axis. In this study, we assessed the functional and structural alterations caused by exposure to a chronic unpredictable stress (CUS) paradigm on the dorsal-ventral axis of the hippocampus. The morphological analysis demonstrated that CUS had opposite outcomes in the structure of the dorsal (DH) and ventral hippocampus (VH): whereas in the DH, stress triggered a volumetric reduction as a result of atrophy of CA3 and CA1 apical dendrites, in the VH there was an increase in hippocampal volume concurrent with the increase of CA3 apical dendrites. In parallel, electrophysiological data revealed that stress led to a decrease in VH LTD.
