Role of body condition score and adiponectin expression in the progression of canine mammary carcinomas
Obesity has been identified as a risk factor for developing breast cancer in post-menopausal period in humans and has been suspected to be associated with a worse prognosis also in the bitch. The aims of this study were to investigate the association between body condition score (BCS) and the prognosis of canine mammary carcinomas (CMCs) and the relationships between adiponectin expression and tumour behaviour. Seventy-three bitches with tubular, tubulopapillary, solid or complex carcinomas were included in the present study. For each dog, evaluation of BCS was conducted using a nine-point BCS system and the study population was divided into normal weight (4–5/9 points; n = 42), overweight (6–7/9 points; n = 19) and obese (8–9/9 points; n = 12). Type of diet (commercial, homemade or mixed) was recorded. After surgical excision, histological type, tumour size and nodal status were assessed and adiponectin expression was determined and quantified by immunohistochemistry and morphometric analysis. CMC histotype was not correlated with BCS, while a positive correlation between BCS and histological grade (p <.01) was observed. Overweight and obese bitches combined showed a shorter cancer-specific survival than normal weighted bitches (p <.01). Bitches fed with a homemade diet had a higher BCS than dogs fed with a commercial one, although no relationship was observed between diet and cancer-specific survival. Thirty-six CMCs scored positive for adiponectin expression (49%), but no correlation was found between the hormone expression and either CMC characteristics or prognosis. In conclusion, a higher BCS seems to be related with a higher prevalence of more aggressive CMCs and negatively affects the survival time in bitches with these mammary tumours
Crackling Noise, Power Spectra and Disorder Induced Critical Scaling
Crackling noise is observed in many disordered non-equilibrium systems in
response to slowly changing external conditions. Examples range from Barkhausen
noise in magnets to acoustic emission in martensites to earthquakes. Using the
non-equilibrium random field Ising model, we derive universal scaling
predictions for the dependence of the associated power spectra on the disorder
and field sweep rate, near an underlying disorder-induced non-equilibrium
critical point. Our theory applies to certain systems in which the crackling
noise results from avalanche-like response to a (slowly) increasing external
driving force, and is characterized by a broad power law scaling regime of the
power spectra. We compute the critical exponents and discuss the relevance of
the results to experiments.Comment: 27 Latex Pages, 14 eps figure
Redondovirus DNA in human respiratory samples
Background: Redondovirus (ReDoV) is a recently discovered circular, Rep-encoding single-stranded DNA (CRESS-DNA) virus in humans. Its pathogenesis and clinical associations are still completely unknown. Methods: The presence of ReDoV DNA was investigated in biological specimens of 543 Italian subjects by in-house developed PCR assays. Results: The overall ReDoV prevalence was about 4% (23 of 543 samples). The virus was detected in 22 of 209 (11 %) respiratory samples. One stool sample was also ReDoV positive. Viral DNA was not found in blood samples from immunocompetent and immunosuppressed subjects and cerebrospinal fluids from patients with neurological diseases. Genomic nucleotide differences were detected among the ReDoV isolates by sequencing a 582-nucleotide fragment of the capsid gene of the viral genome. Conclusions: The results demonstrate that ReDoV is mainly present in the respiratory tract of infected people. Further investigations are needed to reveal possible clinical implications of this new CRESS-DNA virus in humans
Developing food, water and energy nexus workflows
There is a growing recognition of the interdependencies among the supply systems that rely upon food, water and energy. Billions of people lack safe and sufficient access to these systems, coupled with a rapidly growing global demand and increasing resource constraints. Modeling frameworks are considered one of the few means available to understand the complex interrelationships among the sectors, however development of nexus related frameworks has been limited. We describe three open- source models well known in their respective domains (i.e. TerrSysMP, WOFOST and SWAT) where components of each if combined could help decision-makers address the nexus issue. We propose as a first step the development of simple workflows utilizing essential variables and addressing components of the above-mentioned models which can act as building-blocks to be used ultimately in a comprehensive nexus model framework. The outputs of the workflows and the model framework are designed to address the SDG
Phospho-HDAC6 Gathers Into Protein Aggregates in Parkinson’s Disease and Atypical Parkinsonisms
HDAC6 is a unique histone deacetylase that targets cytoplasmic non-histone proteins and has a specific ubiquitin-binding activity. Both of these activities are required for HDAC6-mediated formation of aggresomes, which contain misfolded proteins that will ultimately be degraded via autophagy. HDAC6 deacetylase activity is increased following phosphorylation on serine 22 (phospho-HDAC6). In human, HDAC6 localizes in neuronal Lewy bodies in Parkinson’s disease (PD) and in oligodendrocytic Papp–Lantos bodies in multiple system atrophy (MSA). However, the expression of phospho-HDAC6 in post-mortem human brains is currently unexplored. Here, we evaluate and compare the distribution of HDAC6 and its phosphorylated form in human brains obtained from patients affected by three forms of parkinsonism: two synucleinopathies (PD and MSA) and a tauopathy (progressive supranuclear palsy, PSP). We find that both HDAC6 and its phosphorylated form localize with pathological protein aggregates, including \u3b1-synuclein-positive Lewy bodies in PD and Papp–Lantos bodies in MSA, and phospho-tau-positive neurofibrillary tangles in PSP. We further find a direct interaction of HDAC6 with \u3b1-synuclein with proximity ligation assay (PLA) in neuronal cell of PD patients. Taken together, our findings suggest that both HDAC6 and phospho-HDAC6 regulate the homeostasis of intra-neuronal proteins in parkinsonism
Demagnetization via Nucleation of the Nonequilibrium Metastable Phase in a Model of Disorder
We study both analytically and numerically metastability and nucleation in a
two-dimensional nonequilibrium Ising ferromagnet. Canonical equilibrium is
dynamically impeded by a weak random perturbation which models homogeneous
disorder of undetermined source. We present a simple theoretical description,
in perfect agreement with Monte Carlo simulations, assuming that the decay of
the nonequilibrium metastable state is due, as in equilibrium, to the
competition between the surface and the bulk. This suggests one to accept a
nonequilibrium "free-energy" at a mesoscopic/cluster level, and it ensues a
nonequilibrium "surface tension" with some peculiar low-T behavior. We
illustrate the occurrence of intriguing nonequilibrium phenomena, including:
(i) Noise-enhanced stabilization of nonequilibrium metastable states; (ii)
reentrance of the limit of metastability under strong nonequilibrium
conditions; and (iii) resonant propagation of domain walls. The cooperative
behavior of our system may also be understood in terms of a Langevin equation
with additive and multiplicative noises. We also studied metastability in the
case of open boundaries as it may correspond to a magnetic nanoparticle. We
then observe burst-like relaxation at low T, triggered by the additional
surface randomness, with scale-free avalanches which closely resemble the type
of relaxation reported for many complex systems. We show that this results from
the superposition of many demagnetization events, each with a well- defined
scale which is determined by the curvature of the domain wall at which it
originates. This is an example of (apparent) scale invariance in a
nonequilibrium setting which is not to be associated with any familiar kind of
criticality.Comment: 26 pages, 22 figure
La scuola che funziona. Indagine sulle caratteristiche organizzative e didattiche in istituti con effetto-scuola positivo e negativo
Theoretical models suggest multifactorial approaches for the investigation of school effectiveness. Therefore, in this study factors related to organizational and educational processes have been considered, with the aim of investigating differences between institutions with positive versus negative added value (developed by Invalsi). The study involved 1174 teachers, employed in 73 schools in three different Italian regions, who filled in an on-line questionnaire. The comparison between schools with a positive versus negative added value showed some differences. Particularly, schools with more positive learning outcomes showed significantly higher levels of supportive leadership and a participative decision-making style. Results and intervention proposals are discussed using a multifactorial and interdisciplinary approach.

Games people play with brands: An application of Transactional Analysis to marketplace relationships.
Relationships have been normalised in marketing theory as mutuality beneficial, long-term dyads. This obscures their emotional content, ignores critical conceptualisations of corporate exploitation, and fails to capture the range of possible marketplace relationship forms. In this paper we offer Berne’s (1964) Transactional Analysis (TA) as a way to uncover the biographical psychology that informs marketplace relationship structures and their accompanying emotions, and to provide a critique of such arrangements. We first explain TA, its origins, its relationship with psychoanalysis, its limitations, and contemporary extensions beyond therapy. We then present the structural basis of marketplace relationships from a TA perspective, before illustrating how TA Game Analysis can be applied through an analysis of the iPhone and related mobile phone contracts, and the Games If I didn’t Love Apple and Smallprint. Finally we discuss the implications of such an approach for transforming market practices based on recognition of Marketplace Games and their modification
Essential earth observation variables for high-level multi-scale indicators and policies
Several holistic approaches are based on the description of socio-ecological systems to address the sustainability challenge. Essential Variables (EVs) have the potential to support these approaches by describing the status of the Earth system through monitoring and modeling. The different classes of EVs can be organized along the environmental policy framework of Drivers, Pressures, States, Impacts and Responses. The EV concept represents an opportunity to strengthen monitoring systems by providing observations to seize the fundamental dimensions of the Earth system
The Group on Earth Observation (GEO) is a partnership of 113 nations and 134 participating organizations in 2021 that are dedicated to making Earth Observation (EO) data available globally to inform about the state of the environment and enable data-driven decision processes. GEO is building the Global Earth Observation System of Systems, a set of coordinated and independent EO, information and processing systems that interoperate to provide access to EO for users in the public and private sectors. The progresses made in the development of various classes of EVs are described with their main policy targets, Internet links and key references
The paper reviews the literature on EVs and describes the main contributions of the EU GEOEssential project to integrate EVs within the work plan of GEO in order to better address selected environmental policies and the SDGs. A new GEO-EVs community has been set to discuss about the current status of the EVs, exchange knowledge, experiences and assess the gaps to be solved in their communities of providers and users. A set of four traits characterizing an EV was put forward to describe the entire socio-ecological system of planet Earth: Essentiality, Evolvability, Unambiguity, and Feasibility. A workflow from the identification of EO data sources to the final visualization of SDG 15.3.1 indicators on land degradation is demonstrated, spanning through the use of different EVs, the definition of the knowledge base on this indicator, the implementation of the workflow in the VLab (a cloud-based processing infrastructure), the presentation of the outputs on a dedicated dashboard and the corresponding narrative through a story map.
The concept of EV started in the climate sphere and spread to other domains of the earth system but less so in socio-economic activities. More work is therefore needed to converge on a common definition and criteria in order to complete the implementation of EVs in all GEO focus areas. EVs should screen the entire Earth's social-ecological system, providing a trusted and long-term foundation for interdisciplinary approaches such as ecological footprinting, planetary boundaries, disaster risk reduction, and nexus frameworks, as well as many other policy frameworks such as the SDG
