Detection of equine atypical myopathy-associated hypoglycin A in plant material: Optimisation and validation of a novel LC-MS based method without derivatisation
Hypoglycin A (HGA) toxicity, following ingestion of material from certain plants, is linked to an acquired multiple acyl-CoA dehydrogenase deficiency known as atypical myopathy, a commonly fatal form of equine rhabdomyolysis seen worldwide. Whilst some plants are known to contain this toxin, little is known about its function or the mechanisms that lead to varied HGA concentrations between plants. Consequently, reliable tools to detect this amino acid in plant samples are needed. Analytical methods for HGA detection have previously been validated for the food industry, however, these techniques rely on chemical derivatisation to obtain accurate results at low HGA concentrations. In this work, we describe and validate a novel method, without need for chemical derivatisation (accuracy = 84–94%; precision = 3–16%; reproducibility = 3–6%; mean linear range R2 = 0.999). The current limit of quantitation for HGA in plant material was halved (from 1μg/g in previous studies) to 0.5μg/g. The method was tested in Acer pseudoplatanus material and other tree and plant species. We confirm that A. pseudoplatanus is most likely the only source of HGA in trees found within European pastures
Separation of Parallel Encoded Complex-Valued Slices (SPECS) From A Single Complex-Valued Aliased Coil Image
Purpose
Achieving a reduction in scan time with minimal inter-slice signal leakage is one of the significant obstacles in parallel MR imaging. In fMRI, multiband-imaging techniques accelerate data acquisition by simultaneously magnetizing the spatial frequency spectrum of multiple slices. The SPECS model eliminates the consequential inter-slice signal leakage from the slice unaliasing, while maintaining an optimal reduction in scan time and activation statistics in fMRI studies. When the combined k-space array is inverse Fourier reconstructed, the resulting aliased image is separated into the un-aliased slices through a least squares estimator. Without the additional spatial information from a phased array of receiver coils, slice separation in SPECS is accomplished with acquired aliased images in shifted FOV aliasing pattern, and a bootstrapping approach of incorporating reference calibration images in an orthogonal Hadamard pattern. The aliased slices are effectively separated with minimal expense to the spatial and temporal resolution. Functional activation is observed in the motor cortex, as the number of aliased slices is increased, in a bilateral finger tapping fMRI experiment. Conclusion
The SPECS model incorporates calibration reference images together with coefficients of orthogonal polynomials into an un-aliasing estimator to achieve separated images, with virtually no residual artifacts and functional activation detection in separated images
Conformational transformations induced by the charge-curvature interaction at finite temperature
The role of thermal fluctuations on the conformational dynamics of a single
closed filament is studied. It is shown that, due to the interaction between
charges and bending degrees of freedom, initially circular aggregates may
undergo transformation to polygonal shape. The transition occurs both in the
case of hardening and softening charge-bending interaction. In the former case
the charge and curvature are smoothly distributed along the chain while in the
latter spontaneous kink formation is initiated. The transition to a
non-circular conformation is analogous to the phase transition of the second

Crystallization dynamics of a single layer complex plasma
We report a series of complex (dusty) plasma experiments, aimed at the study
of the detailed time evolution of the re-crystallisation process following a
rapid quench of a two dimensional dust liquid. The experiments were accompanied
by large-scale (million particle) molecular dynamics simulations, assuming
Yukawa type inter-particle interaction. Both experiment and simulation show a
(power law) dependence of the linear crystallite domain size
as measured by the bond-order correlation length, translational correlation
length, dislocation (defect) density, and a direct size measurement algorithm.
The results show two stages of order formation: on short time-scales individual
particle motion dominates; this is a fast process characterized by
. At longer time-scales, small crystallites undergo
collective rearrangement, merging into bigger ones, resulting in a smaller

The acute management of trauma hemorrhage: a systematic review of randomized controlled trials.
PublishedResearch Support, Non-U.S. Gov'tReviewINTRODUCTION: Worldwide, trauma is a leading cause of death and disability. Haemorrhage is responsible for up to 40% of trauma deaths. Recent strategies to improve mortality rates have focused on optimal methods of early hemorrhage control and correction of coagulopathy. We undertook a systematic review of randomized controlled trials (RCT) which evaluated trauma patients with hemorrhagic shock within the first 24 hours of injury and appraised how the interventions affected three outcomes: bleeding and/or transfusion requirements; correction of trauma induced coagulopathy and mortality. METHODS: Comprehensive searches were performed of MEDLINE, EMBASE, CENTRAL (The Cochrane Library Issue 7, 2010), Current Controlled Trials, ClinicalTrials.gov, the World Health Organization International Clinical Trials Registry Platform (ICTRP) and the National Health Service Blood and Transplant Systematic Review Initiative (NHSBT SRI) RCT Handsearch Database. RESULTS: A total of 35 RCTs were identified which evaluated a wide range of clinical interventions in trauma hemorrhage. Many of the included studies were of low methodological quality and participant numbers were small. Bleeding outcomes were reported in 32 studies; 7 reported significantly reduced transfusion use following a variety of clinical interventions, but this was not accompanied by improved survival. Minimal information was found on traumatic coagulopathy across the identified RCTs. Overall survival was improved in only three RCTs: two small studies and a large study evaluating the use of tranexamic acid. CONCLUSIONS: Despite 35 RCTs there has been little improvement in outcomes over the last few decades. No clear correlation has been demonstrated between transfusion requirements and mortality. The global trauma community should consider a coordinated and strategic approach to conduct well designed studies with pragmatic endpoints.This research project was funded by the National Institute for Health

Clinical assessors' working conceptualisations of undergraduate consultation skills: a framework analysis of how assessors make expert judgements in practice.
Undergraduate clinical assessors make expert, multifaceted judgements of consultation skills in concert with medical school OSCE grading rubrics. Assessors are not cognitive machines: their judgements are made in the light of prior experience and social interactions with students. It is important to understand assessors' working conceptualisations of consultation skills and whether they could be used to develop assessment tools for undergraduate assessment. To identify any working conceptualisations that assessors use while assessing undergraduate medical students' consultation skills and develop assessment tools based on assessors' working conceptualisations and natural language for undergraduate consultation skills. In semi-structured interviews, 12 experienced assessors from a UK medical school populated a blank assessment scale with personally meaningful descriptors while describing how they made judgements of students' consultation skills (at exit standard). A two-step iterative thematic framework analysis was performed drawing on constructionism and interactionism. Five domains were found within working conceptualisations of consultation skills: Application of knowledge; Manner with patients; Getting it done; Safety; and Overall impression. Three mechanisms of judgement about student behaviour were identified: observations, inferences and feelings. Assessment tools drawing on participants' conceptualisations and natural language were generated, including 'grade descriptors' for common conceptualisations in each domain by mechanism of judgement and matched to grading rubrics of Fail, Borderline, Pass, Very good. Utilising working conceptualisations to develop assessment tools is feasible and potentially useful. Work is needed to test impact on assessment quality
Neutron spin structure with polarized deuterons and spectator proton tagging at EIC
The neutron's deep-inelastic structure functions provide essential
information for the flavor separation of the nucleon parton densities, the
nucleon spin decomposition, and precision studies of QCD phenomena in the
flavor-singlet and nonsinglet sectors. Traditional inclusive measurements on
nuclear targets are limited by dilution from scattering on protons, Fermi
motion and binding effects, final-state interactions, and nuclear shadowing at
x << 0.1. An Electron-Ion Collider (EIC) would enable next-generation
measurements of neutron structure with polarized deuteron beams and detection
of forward-moving spectator protons over a wide range of recoil momenta (0 <
p_R < several 100 MeV in the nucleus rest frame). The free neutron structure
functions could be obtained by extrapolating the measured recoil momentum
distributions to the on-shell point. The method eliminates nuclear
modifications and can be applied to polarized scattering, as well as to
semi-inclusive and exclusive final states. We review the prospects for neutron
structure measurements with spectator tagging at EIC, the status of R&D
