    Analysis of SF-6D Index Data: Is Beta Regression Appropriate?

    AbstractBackgroundPreference-weighted index scores of health-related quality of life are commonly skewed to the left and bounded at one. Beta regression is used in various disciplines to address the specific features of bounded outcome variables such as heteroscedasticity, but has rarely been used in the context of health-related quality of life measures. We aimed to examine if beta regression is appropriate for analyzing the relationship between subject characteristics and SF-6D index scores.MethodsWe used data from the population-based German KORA F4 study. Besides classical beta regression, we also fitted extended beta regression models by allowing a regression structure on the precision parameter. Regression coefficients and predictive accuracy of the models were compared to those from a linear regression model with model-based and robust standard errors.ResultsThe beta distribution fitted the empirical distribution of the SF-6D index better than the normal distribution. Extended beta regression performed best in terms of predictive accuracy but confidence intervals of the fit measures suggested that no model was superior to the others. Age had a significant negative effect on the precision parameter indicating higher variation of health utilities in older age groups. The observations reporting perfect health had a high influence on model results.ConclusionsBeta regression, especially with precision covariates is a possible supplement to the methods currently used in the analysis of health utility data. In particular, it accounted for the boundedness and heteroscedasticity of the SF-6D index. A pitfall of the beta regression is that it does not work well in handling one-valued observations

    Identification of novel regulators in T-cell differentiation of aplastic anemia patients

    BACKGROUND: Aplastic anemia (AA) is a bone marrow failure syndrome mostly characterized by an immune-mediated destruction of marrow hematopoietic progenitor/stem cells. The resulting hypocellularity limits a detailed analysis of the cellular immune response. To overcome this technical problem we performed a microarray analysis of CD3(+ )T-cells derived from bone marrow aspirates and peripheral blood samples of newly diagnosed AA patients and healthy volunteers. Two AA patients were additionally analyzed after achieving a partial remission following immunosuppression. The regulation of selected candidate genes was confirmed by real-time RT-PCR. RESULTS: Among more than 22.200 transcripts, 583 genes were differentially expressed in the bone marrow of AA patients compared to healthy controls. Dysregulated genes are involved in T-cell mediated cytotoxicity, immune response of Th1 differentiated T-cells, and major regulators of immune function. In hematological remission the expression levels of several candidate genes tend to normalize, such as immune regulators and genes involved in proinflammatory immune response. CONCLUSION: Our study suggests a pivotal role of Th1/Tc1 differentiated T-cells in immune-mediated marrow destruction of AA patients. Most importantly, immune regulatory genes could be identified, which are likely involved in the recovery of hematopoiesis and may help to design new therapeutic strategies in bone marrow failure syndromes

    Tricyanidoferrates(−IV) and ruthenates(−IV) with non‐innocent cyanido ligands

    Exceptionally electron-rich, nearly trigonal-planar tricyanidometalate anions [Fe(CN)(3)](7-) and [Ru(CN)(3)](7-) were stabilized in LiSr3[Fe(CN)(3)] and AE(3.5)[M(CN)(3)] (AE=Sr, Ba; M=Fe, Ru). They are the first examples of group 8 elements with the oxidation state of -IV. Microcrystalline powders were obtained by a solid-state route, single crystals from alkali metal flux. While LiSr3[Fe(CN)(3)] crystallizes in P6(3)/m, the polar space group P6(3) with three-fold cell volume for AE(3.5)[M(CN)(3)] is confirmed by second harmonic generation. X-ray diffraction, IR and Raman spectroscopy reveal longer C-N distances (124-128 pm) and much lower stretching frequencies (1484-1634 cm(-1)) than in classical cyanidometalates. Weak C-N bonds in combination with strong M-C pi-bonding is a scheme also known for carbonylmetalates. Instead of the formal notation [Fe-IV(CN-)(3)](7-), quantum chemical calculations reveal non-innocent intermediate-valent CN1.67- ligands and a closed-shell d(10) configuration for Fe, that is, Fe2-

    Identification of critical parameters for PEMFC stack performance characterization and control strategies for reliable and comparable stack benchmarking

    This paper is focused on the identification of critical parameters and on the development of reliable methodologies to achieve comparable benchmark results. Possibilities for control sensor positioning and for parameter variation in sensitivity tests are discussed and recommended options for the control strategy are summarized. This ensures result comparability as well as stable test conditions. E.g., the stack temperature fluctuation is minimized to about 1 °C. The experiments demonstrate that reactants pressures differ up to 12 kPa if pressure control positions are varied, resulting in an average cell voltage deviation of 21 mV. Test parameters simulating different stack applications are summarized. The stack demonstrated comparable average cell voltage of 0.63 V for stationary and portable conditions. For automotive conditions, the voltage increased to 0.69 V, mainly caused by higher reactants pressures. A benchmarking concept is introduced using “steady-state” polarization curves. The occurring 20 mV hysteresis effect between the ascending and descending polarization curve can be corrected calculating the mean value of both voltages. This minimizes the influence of preceding load levels, current set points, and dwell times

    In situ airborne measurements of atmospheric and sea surface parameters related to offshore wind parks in the German Bight

    Between 6 September 2016 and 15 October 2017, meteorological measurement flights were conducted above the German Bight in the framework of the project WIPAFF (Wind Park Far Field). The scope of the measurements was to study long-range wakes with an extent larger than 10 km behind entire wind parks, and to investigate the interaction of wind parks and the marine atmospheric boundary layer. The research aircraft Dornier 128 of the Technische UniversitÀt (TU) Braunschweig performed in total 41 measurement flights during different seasons and different stability conditions. The instrumentation consisted of a nose boom with sensors for measuring the wind vector, temperature and humidity, and additionally sensors for characterizing the water surface, a surface temperature sensor, a laser scanner and two cameras in the visible and infrared wavelength range. A detailed overview of the aircraft, sensors, data post-processing and flight patterns is provided here. Further, averaged profiles of atmospheric parameters illustrate the range of conditions. The potential use of the data set has been shown already by first publications.     HICFD – Highly Efficient Implementation of CFD Codes for HPC Many-Core Architectures

    The objective of the German BMBF research project Highly Efficient Implementation of CFD Codes for HPC Many-Core Architectures (HICFD) is to develop new methods and tools for the analysis and optimization of the performance of parallel computational fluid dynamics (CFD) codes on high performance computer systems with many-core processors. In the work packages of the project it is investigated how the performance of parallel CFD codes written in C can be increased by the optimal use of all parallelism levels. On the highest level MPI is utilized. Furthermore, on the level of the many-core architecture, highly scaling, hybrid OpenMP/MPI methods are implemented. On the level of the processor cores the parallel SIMD units provided by modern CPUs are exploited
