    Effects of manganese-excess on CO2 assimilation, ribulose-1,5-bisphosphate carboxylase/oxygenase, carbohydrates and photosynthetic electron transport of leaves, and antioxidant systems of leaves and roots in Citrus grandis seedlings

    <p>Abstract</p> <p>Background</p> <p>Very little is known about the effects of manganese (Mn)-excess on citrus photosynthesis and antioxidant systems. Seedlings of sour pummelo (<it>Citrus grandis</it>) were irrigated for 17 weeks with nutrient solution containing 2 μM (control) or 500 μM (excess) MnSO<sub>4</sub>. The objective of this study were to understand the mechanisms by which Mn-excess leads to a decrease in CO<sub>2 </sub>assimilation and to test the hypothesis that Mn-induced changes in antioxidant systems differ between roots and leaves.</p> <p>Results</p> <p>Mn-excess decreased CO<sub>2 </sub>assimilation and stomatal conductance, increased intercellular CO<sub>2 </sub>concentration, but did not affect chlorophyll (Chl) level. Both initial and total ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) activity in Mn-excess leaves decreased to a lesser extent than CO<sub>2 </sub>assimilation. Contents of glucose, fructose, starch and total nonstructural carbohydrates did not differ between Mn-excess leaves and controls, while sucrose content was higher in the former. Chl a fluorescence (OJIP) transients from Mn-excess leaves showed increased O-step and decreased P-step, accompanied by positive L- and K-bands. Mn-excess decreased maximum quantum yield of primary photochemistry (F<sub>v</sub>/F<sub>m</sub>) and total performance index (PI<sub>tot,abs</sub>), but increased relative variable fluorescence at I-steps (V<sub>I</sub>) and energy dissipation. On a protein basis, Mn-excess leaves displayed higher activities of monodehydroascorbate reductase (MDAR), glutathione reductase (GR), superoxide dismutase (SOD), catalase (CAT) and guaiacol peroxidase (GPX) and contents of antioxidants, similar ascorbate peroxidase (APX) activities and lower dehydroascorbate reductase (DHAR) activities; while Mn-excess roots had similar or lower activities of antioxidant enzymes and contents of antioxidants. Mn-excess did not affect malondialdehyde (MDA) content of roots and leaves.</p> <p>Conclusions</p> <p>Mn-excess impaired the whole photosynthetic electron transport chain from the donor side of photosystem II (PSII) up to the reduction of end acceptors of photosystem I (PSI), thus limiting the production of reducing equivalents, and hence the rate of CO<sub>2 </sub>assimilation. Both the energy dissipation and the antioxidant systems were enhanced in Mn-excess leaves, while the antioxidant systems in Mn-excess roots were not up-regulated, but still remained high activity. The antioxidant systems in Mn-excess roots and leaves provided sufficient protection to them against oxidative damage.</p

    NNAT and DIRAS3 genes are paternally expressed in pigs

    Although expression and epigenetic differences of imprinted genes have been extensively characterised in man and the mouse, little is known on livestock species. In this study, the polymorphism-based approach was used to detect the imprinting status of NNAT and DIRAS3 genes in five heterozygous pigs (based on SNP) of Large White and Meishan F1 hybrids. The results show that both genes were paternally expressed in all the tested tissues (heart, liver, spleen, lung, kidney, stomach, small intestine, skeletal muscle, fat, uterus, ovary and pituitary). In addition, the NNAT gene had two transcripts in all tested tissues, which is consistent with its counterpart in man and cattle

    Towards high-quality biodiesel production from microalgae using original and anaerobically-digested livestock wastewater

    In this study, we conducted proof-of-concept research towards the simultaneous treatment of livestock wastewater and the generation of high-quality biodiesel, through microalgae technology. Both original (OPE) and anaerobically-digested (DPE) piggery effluents were investigated for the culture of the microalgae, Desmodesmus sp. EJ8-10. After 14 days’ cultivation, the dry biomass from microalgae cultivated in OPE increased from an initial value of 0.01 g/L to 0.33-0.39 g/L, while those growing in DPE only achieved a final dried mass of 0.15-0.35 g/L, under similar initial ammonium nitrogen (NH4+-N) concentrations. The significantly higher microalgal biomass production achieved in the OPE medium may have been supported by the abundance of both macronutrient, such as phosphorus (P), and of micronutrients, such as trace elements, present in the OPE, which may not been present in similar quantities in the DPE. However, a higher lipid content was observed (19.4-28%) in microalgal cells from DPE cultures than those (18.7-22.3%) from OPE cultures. Moreover, the fatty acid compositions in the microalgae cultured in DPE contained high levels of monounsaturated fatty acids (MUFAs) and total C16-C18 acids, which would afford a superior potential for high-quality biodiesel production. The N/P ratio (15.4:1) in OPE was much closer to that indicated by previous studies to be the most suitable (16:1) for microalgae growth, when compared with that determined from the DPE culture medium.     Prevalence of syphilis infection in different tiers of female sex workers in China: implications for surveillance and interventions

    <p>Abstract</p> <p>Background</p> <p>Syphilis has made a dramatic resurgence in China during the past two decades and become the third most prevalent notifiable infectious disease in China. Female sex workers (FSWs) have become one of key populations for the epidemic. In order to investigate syphilis infection among different tiers of FSWs, a cross-sectional study was conducted in 8 sites in China.</p> <p>Methods</p> <p>Serum specimens (n = 7,118) were collected to test for syphilis and questionnaire interviews were conducted to obtain socio-demographic and behavioral information among FSWs recruited from different types of venues. FSWs were categorized into three tiers (high-, middle- and low-tier FSWs) based on the venues where they solicited clients. Serum specimens were screened with enzyme-linked immunosorbent assay (ELISA) for treponemal antibody followed by confirmation with non-treponemal toluidine red unheated serum test (TRUST) for positive ELISA specimens to determine syphilis infection. A logistic regression model was used to determine factors associated with syphilis infection.</p> <p>Results</p> <p>Overall syphilis prevalence was 5.0% (95%CI, 4.5-5.5%). Low-tier FSWs had the highest prevalence (9.7%; 95%CI, 8.3-11.1%), followed by middle-tier (4.3%; 95%CI, 3.6-5.0%, <it>P </it>< 0.001) and high-tier FSWs (2.2%; 95%CI, 1.6-2.9%, <it>P </it>< 0.001). Factors independently associated with syphilis infection included older age, lower education level, geographic location, lower tier of typology, and injection drug use.</p> <p>Conclusions</p> <p>This multi-site survey showed a high prevalence of syphilis infection among FSWs and substantial disparities in syphilis prevalence by the tier of FSWs. The difference in syphilis prevalence is substantial between different tiers of FSWs, with the highest rate among low-tier FSWs. Thus, current surveillance and intervention activities, which have low coverage in low-tier FSWs in China, should be further examined.</p

    Development and Preclinical Studies of Broad-Spectrum Anti-HIV Agent (3′ R ,4′ R )-3-Cyanomethyl-4-methyl-3′,4′-di- O -( S )-camphanoyl-(+)- cis -khellactone (3-Cyanomethyl-4-methyl-DCK)

    In prior investigation, we discovered that (3'R,4'R)-3-cyanomethyl-4-methyl-3',4'-di-O-(S)-camphanoyl-(+)-cis-khellactone (4, 3-cyanomethyl-4-methyl-DCK) showed promising anti-HIV activity. In these current studies, we developed and optimized successfully a practical ten-step synthesis for scale-up preparation to increase the overall yield of 4 from 7.8% to 32%. Furthermore, compound 4 exhibited broad-spectrum anti-HIV activity against wild-type and drug-resistant viral infection of CD4+ T cell lines as well as peripheral blood mononuclear cells by both laboratory-adapted and primary HIV-1 isolates with distinct subtypes and tropisms. Compound 4 was further subjected to in vitro and in vivo pharmacokinetic studies. These studies indicated that 4 has moderate cell permeability, moderate oral bioavailability and low systemic clearance. These results suggest that 4 should be developed as a promising anti-HIV agent for development as a clinical trial candidate

    CodeFuse-13B: A Pretrained Multi-lingual Code Large Language Model

    Code Large Language Models (Code LLMs) have gained significant attention in the industry due to their wide applications in the full lifecycle of software engineering. However, the effectiveness of existing models in understanding non-English inputs for multi-lingual code-related tasks is still far from well studied. This paper introduces CodeFuse-13B, an open-sourced pre-trained code LLM. It is specifically designed for code-related tasks with both English and Chinese prompts and supports over 40 programming languages. CodeFuse achieves its effectiveness by utilizing a high quality pre-training dataset that is carefully filtered by program analyzers and optimized during the training process. Extensive experiments are conducted using real-world usage scenarios, the industry-standard benchmark HumanEval-x, and the specially designed CodeFuseEval for Chinese prompts. To assess the effectiveness of CodeFuse, we actively collected valuable human feedback from the AntGroup's software development process where CodeFuse has been successfully deployed. The results demonstrate that CodeFuse-13B achieves a HumanEval pass@1 score of 37.10%, positioning it as one of the top multi-lingual code LLMs with similar parameter sizes. In practical scenarios, such as code generation, code translation, code comments, and testcase generation, CodeFuse performs better than other models when confronted with Chinese prompts.Comment: 10 pages with 2 pages for reference
