    Adaptive subspace sampling for class imbalance processing

    © 2016 IEEE. This paper presents a novel oversampling technique that addresses highly imbalanced data distribution. At present, the imbalanced data that have anomalous class distribution and underrepresented data are difficult to deal with through a variety of conventional machine learning technologies. In order to balance class distributions, an adaptive subspace self-organizing map (ASSOM) that combines the local mapping scheme and globally competitive rule is proposed to artificially generate synthetic samples focusing on minority class samples. The ASSOM is conformed with feature-invariant characteristics, including translation, scaling and rotation, and it retains the independence of basis vectors in each module. Specifically, basis vectors generated via each ASSOM module can avoid generating repeated representative features that offer nothing but heavy computational load. Several experimental results demonstrate that the proposed ASSOM method with supervised learning manner is superior to other existing oversampling techniques

    Fuzzy Integral with Particle Swarm Optimization for a Motor-Imagery-Based Brain-Computer Interface

    © 2016 IEEE. A brain-computer interface (BCI) system using electroencephalography signals provides a convenient means of communication between the human brain and a computer. Motor imagery (MI), in which motor actions are mentally rehearsed without engaging in actual physical execution, has been widely used as a major BCI approach. One robust algorithm that can successfully cope with the individual differences in MI-related rhythmic patterns is to create diverse ensemble classifiers using the subband common spatial pattern (SBCSP) method. To aggregate outputs of ensemble members, this study uses fuzzy integral with particle swarm optimization (PSO), which can regulate subject-specific parameters for the assignment of optimal confidence levels for classifiers. The proposed system combining SBCSP, fuzzy integral, and PSO exhibits robust performance for offline single-trial classification of MI and real-time control of a robotic arm using MI. This paper represents the first attempt to utilize fuzzy fusion technique to attack the individual differences problem of MI applications in real-world noisy environments. The results of this study demonstrate the practical feasibility of implementing the proposed method for real-world applications

    CLEC5A-mediated enhancement of the inflammatory response in myeloid cells contributes to influenza virus pathogenicity in vivo

    Human infections with influenza viruses exhibit mild to severe clinical outcomes as a result of complex virus-host interactions. Induction of inflammatory mediators via pattern recognition receptors may dictate subsequent host responses for pathogen clearance and tissue damage. We identified that human C-type lectin domain family 5 member A (CLEC5A) interacts with the hemagglutinin protein of influenza viruses expressed on lentiviral pseudoparticles through lectin screening. Silencing CLEC5A gene expression, blocking influenza-CLEC5A interactions with anti-CLEC5A antibodies, or dampening CLEC5A-mediated signaling using a spleen tyrosine kinase inhibitor consistently reduced the levels of proinflammatory cytokines produced by human macrophages without affecting the replication of influenza A viruses of different subtypes. Infection of bone marrow-derived macrophages from CLEC5A-deficient mice showed reduced levels of tumor necrosis factor alpha (TNF-α) and IP-10 but elevated alpha interferon (IFN-α) compared to those of wild-type mice. The heightened type I IFN response in the macrophages of CLEC5A-deficient mice was associated with upregulated TLR3 mRNA after treatment with double-stranded RNA. Upon lethal challenges with a recombinant H5N1 virus, CLEC5A-deficient mice showed reduced levels of proinflammatory cytokines, decreased immune cell infiltration in the lungs, and improved survival compared to the wild-type mice, despite comparable viral loads noted throughout the course of infection. The survival difference was more prominent at a lower dose of inoculum. Our results suggest that CLEC5A-mediated enhancement of the inflammatory response in myeloid cells contributes to influenza pathogenicity in vivo and may be considered a therapeutic target in combination with effective antivirals.     © 2020 IEEE. Kohonen's Adaptive Subspace Self-Organizing Map (ASSOM) learns several subspaces of the data where each subspace represents some invariant characteristics of the data. To deal with the imbalance classification problem, earlier we have proposed a method for oversampling the minority class using Kohonen's ASSOM. This investigation extends that study, clarifies some issues related to our earlier work, provides the algorithm for generation of the oversamples, applies the method on several benchmark data sets, and makes an application to a Brain Computer Interface (BCI) problem. First we compare the performance of our method using some benchmark data sets with several state-of-The-Art methods. Finally, we apply the ASSOM-based technique to analyze a BCI based application using electroencephalogram (EEG) datasets. Our results demonstrate the effectiveness of the ASSOM-based method in dealing with imbalance classification problem

    TRPV6 Determines the Effect of Vitamin D3 on Prostate Cancer Cell Growth

    Despite remarkable advances in the therapy and prevention of prostate cancer it is still the second cause of death from cancer in industrialized countries. Many therapies initially shown to be beneficial for the patients were abandoned due to the high drug resistance and the evolution rate of the tumors. One of the prospective therapeutical agents even used in the first stage clinical trials, 1,25-dihydroxyvitamin D3, was shown to be either unpredictable or inefficient in many cases. We have already shown that TRPV6 calcium channel, which is the direct target of 1,25-dihydroxyvitamin D3 receptor, positively controls prostate cancer proliferation and apoptosis resistance (Lehen'kyi et al., Oncogene, 2007). However, how the known 1,25-dihydroxyvitamin D3 antiproliferative effects may be compatible with the upregulation of pro-oncogenic TRPV6 channel remains a mystery. Here we demonstrate that in low steroid conditions 1,25-dihydroxyvitamin D3 upregulates the expression of TRPV6, enchances the proliferation by increasing the number of cells entering into S-phase. We show that these pro-proliferative effects of 1,25-dihydroxyvitamin D3 are directly mediated via the overexpression of TRPV6 channel which increases calcium uptake into LNCaP cells. The apoptosis resistance of androgen-dependent LNCaP cells conferred by TRPV6 channel is drastically inversed when 1,25-dihydroxyvitamin D3 effects were combined with the successful TRPV6 knockdown. In addition, the use of androgen-deficient DU-145 and androgen-insensitive LNCaP C4-2 cell lines allowed to suggest that the ability of 1,25-dihydroxyvitamin D3 to induce the expression of TRPV6 channel is a crucial determinant of the success or failure of 1,25-dihydroxyvitamin D3-based therapies

    Differential Effects of Concomitant Use of Vitamins C and E on Trophoblast Apoptosis and Autophagy between Normoxia and Hypoxia-Reoxygenation

    Concomitant supplementation of vitamins C and E during pregnancy has been reportedly associated with low birth weight, the premature rupture of membranes and fetal loss or perinatal death in women at risk for preeclampsia; however, the cause is unknown. We surmise that hypoxia-reoxygenation (HR) within the intervillous space due to abnormal placentation is the mechanism and hypothesize that concomitant administration of aforementioned vitamin antioxidants detrimentally affects trophoblast cells during HR.Using villous explants, concomitant administration of 50 microM of vitamins C and E was observed to reduce apoptotic and autophagic changes in the trophoblast layer at normoxia (8% oxygen) but to cause more prominent apoptosis and autophagy during HR. Furthermore, increased levels of Bcl-2 and Bcl-xL in association with a decrease in the autophagy-related protein LC3-II were noted in cytotrophoblastic cells treated with vitamins C and E under standard culture conditions. In contrast, vitamin treatment decreased Bcl-2 and Bcl-xL as well as increased mitochondrial Bak and cytosolic LC3-II in cytotrophoblasts subjected to HR.Our results indicate that concomitant administration of vitamins C and E has differential effects on the changes of apoptosis, autophagy and the expression of Bcl-2 family of proteins in the trophoblasts between normoxia and HR. These changes may probably lead to the impairment of placental function and suboptimal growth of the fetus

    Blood-based omic profiling supports female susceptibility to tobacco smoke-induced cardiovascular diseases

    We recently reported that differential gene expression and DNA methylation profiles in blood leukocytes of apparently healthy smokers predicts with remarkable efficiency diseases and conditions known to be causally associated with smoking, suggesting that blood-based omic profiling of human populations may be useful for linking environmental exposures to potential health effects. Here we report on the sex-specific effects of tobacco smoking on transcriptomic and epigenetic features derived from genome-wide profiling in white blood cells, identifying 26 expression probes and 92 CpG sites, almost all of which are affected only in female smokers. Strikingly, these features relate to numerous genes with a key role in the pathogenesis of cardiovascular disease, especially thrombin signaling, including the thrombin receptors on platelets F2R (coagulation factor II (thrombin) receptor; PAR1) and GP5 (glycoprotein 5), as well as HMOX1 (haem oxygenase 1) and BCL2L1 (BCL2-like 1) which are involved in protection against oxidative stress and apoptosis, respectively. These results are in concordance with epidemiological evidence of higher female susceptibility to tobacco-induced cardiovascular disease and underline the potential of blood-based omic profiling in hazard and risk assessment
