    AGEs increase IL-6 and ICAM-1 expression

    Background and Objectives: Diabetes mellitus (DM) is a risk factor for periodontal diseases and may exacerbate the progression of the pathogenesis of periodontitis. Advanced glycation end-products (AGEs) cause DM complications relative to levels of glycemic control and larger amounts accumulate in the periodontal tissues of patients with periodontitis and DM. In the present study, we investigated the effects of AGEs on the expression of inflammation-related factors in human gingival fibroblasts (HGFs) in order to elucidate the impact of AGEs on DM-associated periodontitis. Materials and Methods: HGFs were cultured with or without AGEs. Cell viability was examined, and RNA and protein fractions were isolated from AGE-treated cells. The expression of IL-6, ICAM-1, and the receptor for AGE (RAGE) was investigated using RT-PCR, quantitative real-time PCR, and ELISA, and reactive oxygen species (ROS) activity was measured using a kit with 2’,7’-dichlorofluorescin diacetate. Human monocytic cells (THP-1) labelled with a fluorescent reagent were co-cultured with HGFs treated with AGEs and IL-6 siRNA, and the adhesive activity of THP-1 cells to HGFs was assessed. The expression of IL-6 and ICAM-1 was examined when HGFs were pretreated with recombinant human IL-6 (rhIL-6), the siRNAs of RAGE and IL-6, and inhibitors of MAPK and NF-κB, and then cultured with and without AGEs. The phosphorylation of MAPK and NF-κB was assessed using Western blotting. Results: AGEs increased the mRNA and protein expressions of RAGE, IL-6, ICAM-1 and ROS activity in HGFs, and promoted the adhesion of THP-1 cells to HGFs, but had no effect on cell viability until 72 h. rhIL-6 increased ICAM-1 expression in HGFs, while the siRNAs of RAGE and IL-6 inhibited AGE-induced IL6 and ICAM1 mRNA expression, and IL-6 siRNA depressed AGE-induced THP-1 cell adhesion. AGEs increased the phosphorylation of p38 and ERK MAPKs, p65 NF-κB, and IκBα, while inhibitors of p38, ERK MAPKs, and NF-κB significantly decreased AGE-induced IL-6 and ICAM-1 expression. Conclusions: AGEs increase IL-6 and ICAM-1 expression via the RAGE, MAPK and NF-κB pathways in HGFs and may exacerbate the progression of the pathogenesis of periodontal diseases

    The 2G allele of promoter region of Matrix metalloproteinase-1 as an essential pre-condition for the early onset of oral squamous cell carcinoma

    <p>Abstract</p> <p>Background</p> <p>Matrix metalloproteinase (<it>MMP</it>) is known to be involved in the initial and progressive stages of cancer development, and in the aggressive phenotypes of cancer. This study examines the association of single nucleotide polymorphisms in promoter regions of <it>MMP-1 </it>and <it>MMP-3 </it>with susceptibility to oral squamous cell carcinoma (OSCC).</p> <p>Methods</p> <p>We compared 170 Japanese OSCC cases and 164 healthy controls for genotypes of <it>MMP-1 </it>and <it>MMP-3</it>.</p> <p>Results</p> <p>The frequency of the <it>MMP-1 </it>2G allele was higher and that of the 1G homozygote was lower in the OSCC cases (<it>p </it>= 0.034). A multivariate logistic regression analysis revealed that subjects who were 45 years old or older had a significantly increased (2.47-fold) risk of OSCC (95%CI 1.47–4.14, <it>p </it>= 0.0006), and those carrying the <it>MMP-1 </it>2G allele had a 2.30-fold risk (95%CI 1.15–4.58, <it>p </it>= 0.018), indicating independent involvement of these factors in OSCC. One of the key discoveries of this research is the apparent reduction of the <it>MMP-1 </it>1G/1G and 1G/2G genotype distributions among the early onset OSCC cases under the ages of 45 years. It should be noted that the tongue was the primary site in 86.2% of these early onset cases. This could suggest the specific carcinogenic mechanisms, i.e. specific carcinogenic stimulations and/or genetic factors in the tongue.</p> <p>Conclusion</p> <p>Since the 2G allele is a majority of the <it>MMP-1 </it>genotype in the general population, it seems to act as a genetic pre-condition in OSCC development. However this report suggests a crucial impact of the <it>MMP-1 </it>2G allele in the early onset OSCC.</p

    Chelation ability of spironaphthoxazine with metal ions in silica gel

    Spironaphthoxazine (SNO) and three metal ions, Mg2+, Zn2+, and Al3+, were dispersed in silica gels by the sol-gel method. The chelation ability of SNO with the metal ions in silica gels was investigated by measuring the fluorescence spectra and was compared to that of 8-hydroxyquinoline (8-HQ) in ethanol and silica gels. A merocyanine-type isomer photoderived from SNO as well as 8-HQ easily formed complexes of the metal ions in the order of Al3+, Zn2+, and Mg2+ because the coordination ability of the metal ions to such ligands depended on their electron affinity. The changes in the fluorescence spectra of the silica gel samples during light irradiation were also investigated. The relative band intensity due to the intermediate species between the original SNO and the merocyanine species decreased and that of the complex increased with the UV irradiation time. The reverse process was observed during visible irradiation.     Accuracy of Dental Models Fabricated Using Recycled Poly-Lactic Acid

    Based on the hypothesis that the fabrication of dental models using fused deposition modeling and poly-lactic acid (PLA), followed by recycling and reusing, would reduce industrial waste, we aimed to compare the accuracies of virgin and recycled PLA models. The PLA models were recycled using a crusher and a filament-manufacturing machine. Virgin PLA was labeled R, and the first, second, and third recycles were labeled R1, R2, and R3, respectively. To determine the accuracies of the virgin and reused PLA models, identical provisional crowns were fitted, and marginal fits were obtained using micro-computed tomography. A marginal fit of 120 µm was deemed acceptable based on previous literature. The mesial, distal, buccal, and palatal centers were set at M, D, B, and P, respectively. The mean value of each measurement point was considered as the result. When comparing the accuracies of R and R1, R2, and R3, significant differences were noted between R and R3 at B, R and R2, R3 at P, and R and R3 at D (p < 0.05). No significant difference was observed at M. This study demonstrates that PLA can be recycled only once owing to accuracy limitations

    Fracture healing is a sequence of events that involves a combination of intramembranous and endochondral ossification. During fracture healing, a number of growth factors and important cytokines such as BMP-2, BMP-4, TGF-β, and PDGF-B and Osteopontin are present at elevated levels in and around the fracture site, suggesting that they play an active role in promoting fracture healing. The regulation of fracture repair by important growth factors and cytokines in a mandible fracture has not been clearly understood. Taking this into consideration, we want to know about the control mechanism that the coordinates recruitment, localization, and function of different cell populations at the mandible fracture. In this study, we present 48 Wistar rats that have their right ramus mandible fractured with a bending clamp. On days 3, 7, 14 and 21 after the operation the animals were sacrificed with perfusion technique. For immunohistochemistry we used goat polyclonal antiboy for BMP-2 and BMP-4, rabbit polyclonal antibody for TGF-β and PDGF-B, and mouse monoclonal antibody for Osteopontin as the primary antibody.     Verification of the accuracy of dynamic navigation for conventional and mouthpiece methods: in vivo study

    Abstract Background Dynamic navigation for implant placement is becoming popular under the concept of top-down treatment. The purpose of this study is to verify the accuracy of a dynamic navigation system for implant placement. Methods Implant placement was performed on 38 patients using 50 implant fixtures. Patients in group C were treated using a conventional method, in which thermoplastic clips were fixed to the teeth, and patients in group M were treated using thermoplastic clips fixed to a mouthpiece attached to the teeth. The groups were compared to verify whether an accuracy difference existed. A treatment planning support program for dental implants was used to superimpose the postoperative computed tomography data on the preoperative implant design data to measure the entry point, apex point, and angular deviation. Results The accuracy of group C was 1.36 ± 0.51 mm for entry point, 1.30 ± 0.59 mm for apex point, and 3.20 ± 0.74° for angular deviation. The accuracy of group M was 1.06 ± 0.31 mm for the entry point, 1.02 ± 0.30 mm for the apex point, and 2.91 ± 0.97° for angular deviation. Significant differences were observed in the entry and apex points between the two groups. Conclusions The results indicate that group M exhibited better accuracy than group C, indicating that the stability of the thermoplastic clip is important for ensuring the accuracy of the dynamic navigation system. No previous studies have verified the accuracy of this system using the mouthpiece method, and additional data is required to confirm its accuracy for dental implant placement. The mouthpiece method improves the accuracy of implant placement and provides a safer implant treatment than the conventional method.
