Environmental monitoring of Mycobacterium bovis in badger feces and badger sett soil by real-time PCR, as confirmed by immunofluorescence, immunocapture, and cultivation
Real-time PCR was used to detect and quantify Mycobacterium bovis cells in
naturally infected soil and badger faeces. Immunomagnetic capture,
immunofluorescence and selective culture confirmed species identification and cell
viability. These techniques will prove useful for monitoring M. bovis in the
environment and for elucidating transmission routes between wildlife and cattle
A qualitative study of primary care professionals’ views of case finding for depression in patients with diabetes or coronary heart disease in the UK
<p>Background
Routinely conducting case finding (also commonly referred to as screening) in patients with chronic illness for depression in primary care appears to have little impact. We explored the views and experiences of primary care nurses, doctors and managers to understand how the implementation of case finding/screening might impact on its effectiveness.</p>
<p>Methods
Two complementary qualitative focus group studies of primary care professionals including nurses, doctors and managers, in five primary care practices and five Community Health Partnerships, were conducted in Scotland.</p>
<p>Results
We identified several features of the way case finding/screening was implemented that may lead to systematic under-detection of depression. These included obstacles to incorporating case finding/screening into a clinical review consultation; a perception of replacing individualised care with mechanistic assessment, and a disconnection for nurses between management of physical and mental health. Far from being a standardised process that encouraged detection of depression, participants described case finding/screening as being conducted in a way which biased it towards negative responses, and for nurses, it was an uncomfortable task for which they lacked the necessary skills to provide immediate support to patients at the time of diagnosis.</p>
<p>Conclusion
The introduction of case finding/screening for depression into routine chronic illness management is not straightforward. Routinized case finding/screening for depression can be implemented in ways that may be counterproductive to engagement (particularly by nurses), with the mental health needs of patients living with long term conditions. If case finding/screening or engagement with mental health problems is to be promoted, primary care nurses require more training to increase their confidence in raising and dealing with mental health issues and GPs and nurses need to work collectively to develop the relational work required to promote cognitive participation in case finding/screening.</p>
Investigation of Anti-Relaxation Coatings for Alkali-Metal Vapor Cells Using Surface Science Techniques
Many technologies based on cells containing alkali-metal atomic vapor benefit
from the use of anti-relaxation surface coatings in order to preserve atomic
spin polarization. In particular, paraffin has been used for this purpose for
several decades and has been demonstrated to allow an atom to experience up to
10,000 collisions with the walls of its container without depolarizing, but the
details of its operation remain poorly understood. We apply modern surface and
bulk techniques to the study of paraffin coatings, in order to characterize the
properties that enable the effective preservation of alkali spin polarization.
These methods include Fourier transform infrared spectroscopy, differential
scanning calorimetry, atomic force microscopy, near-edge X-ray absorption fine
structure spectroscopy, and X-ray photoelectron spectroscopy. We also compare
the light-induced atomic desorption yields of several different paraffin
materials. Experimental results include the determination that crystallinity of
the coating material is unnecessary, and the detection of C=C double bonds
present within a particular class of effective paraffin coatings. Further study
should lead to the development of more robust paraffin anti-relaxation
coatings, as well as the design and synthesis of new classes of coating

Algebraic Bethe ansatz for the gl(12) generalized model II: the three gradings
The algebraic Bethe ansatz can be performed rather abstractly for whole
classes of models sharing the same -matrix, the only prerequisite being the
existence of an appropriate pseudo vacuum state. Here we perform the algebraic
Bethe ansatz for all models with , rational, gl(12)-invariant
-matrix and all three possibilities of choosing the grading. Our Bethe
ansatz solution applies, for instance, to the supersymmetric t-J model, the
supersymmetric model and a number of interesting impurity models. It may be
extended to obtain the quantum transfer matrix spectrum for this class of
models. The properties of a specific model enter the Bethe ansatz solution
(i.e. the expression for the transfer matrix eigenvalue and the Bethe ansatz
equations) through the three pseudo vacuum eigenvalues of the diagonal elements
of the monodromy matrix which in this context are called the parameters of the

Traversable wormholes coupled to nonlinear electrodynamics
In this work we explore the possible existence of static, spherically
symmetric and stationary, axisymmetric traversable wormholes coupled to
nonlinear electrodynamics. Considering static and spherically symmetric (2+1)
and (3+1)-dimensional wormhole spacetimes, we verify the presence of an event
horizon and the non-violation of the null energy condition at the throat. For
the former spacetime, the principle of finiteness is imposed, in order to
obtain regular physical fields at the throat. Next, we analyze the
(2+1)-dimensional stationary and axisymmetric wormhole, and also verify the
presence of an event horizon, rendering the geometry non-traversable.
Relatively to the (3+1)-dimensional stationary and axisymmetric wormhole
geometry, we find that the field equations impose specific conditions that are
incompatible with the properties of wormholes. Thus, we prove the non-existence
of the general class of traversable wormhole solutions, outlined above, within

Integrable multiparametric quantum spin chains
Using Reshetikhin's construction for multiparametric quantum algebras we
obtain the associated multiparametric quantum spin chains. We show that under
certain restrictions these models can be mapped to quantum spin chains with
twisted boundary conditions. We illustrate how this general formalism applies
to construct multiparametric versions of the supersymmetric t-J and U models.Comment: 17 pages, RevTe
Lineage-specific gene radiations underlie the evolution of novel betalain pigmentation in Caryophyllales.
Betalain pigments are unique to the Caryophyllales and structurally and biosynthetically distinct from anthocyanins. Two key enzymes within the betalain synthesis pathway have been identified: 4,5-dioxygenase (DODA) that catalyzes the formation of betalamic acid and CYP76AD1, a cytochrome P450 gene that catalyzes the formation of cyclo-DOPA. We performed phylogenetic analyses to reveal the evolutionary history of the DODA and CYP76AD1 lineages and in the context of an ancestral reconstruction of pigment states we explored the evolution of these genes in relation to the complex evolution of pigments in Caryophylalles. Duplications within the CYP76AD1 and DODA lineages arose just before the origin of betalain pigmentation in the core Caryophyllales. The duplications gave rise to DODA-α and CYP76AD1-α isoforms that appear specific to betalain synthesis. Both betalain-specific isoforms were then lost or downregulated in the anthocyanic Molluginaceae and Caryophyllaceae. Our findings suggest a single origin of the betalain synthesis pathway, with neofunctionalization following gene duplications in the CYP76AD1 and DODA lineages. Loss of DODA-α and CYP76AD1-α in anthocyanic taxa suggests that betalain pigmentation has been lost twice in Caryophyllales, and exclusion of betalain pigments from anthocyanic taxa is mediated through gene loss or downregulation. Data quality monitoring and performance metrics of a prospective, population-based observational study of maternal and newborn health in low resource settings
BACKGROUND: To describe quantitative data quality monitoring and performance metrics adopted by the Global Network´s (GN) Maternal Newborn Health Registry (MNHR), a maternal and perinatal population-based registry (MPPBR) based in low and middle income countries (LMICs). METHODS: Ongoing prospective, population-based data on all pregnancy outcomes within defined geographical locations participating in the GN have been collected since 2008. Data quality metrics were defined and are implemented at the cluster, site and the central level to ensure data quality. Quantitative performance metrics are described for data collected between 2010 and 2013. RESULTS: Delivery outcome rates over 95% illustrate that all sites are successful in following patients from pregnancy through delivery. Examples of specific performance metric reports illustrate how both the metrics and reporting process are used to identify cluster-level and site-level quality issues and illustrate how those metrics track over time. Other summary reports (e.g. the increasing proportion of measured birth weight compared to estimated and missing birth weight) illustrate how a site has improved quality over time. CONCLUSION: High quality MPPBRs such as the MNHR provide key information on pregnancy outcomes to local and international health officials where civil registration systems are lacking. The MNHR has measures in place to monitor data collection procedures and improve the quality of data collected. Sites have increasingly achieved acceptable values of performance metrics over time, indicating improvements in data quality, but the quality control program must continue to evolve to optimize the use of the MNHR to assess the impact of community interventions in research protocols in pregnancy and perinatal health.Fil: Goudar, Shivaprasad S.. Integrating informatics tools and portable sequencing technology for rapid detection of resistance to anti-tuberculous drugs
BACKGROUND: Mycobacterium tuberculosis resistance to anti-tuberculosis drugs is a major threat to global public health. Whole genome sequencing (WGS) is rapidly gaining traction as a diagnostic tool for clinical tuberculosis settings. To support this informatically, previous work led to the development of the widely used TBProfiler webtool, which predicts resistance to 14 drugs from WGS data. However, for accurate and rapid high throughput of samples in clinical or epidemiological settings, there is a need for a stand-alone tool and the ability to analyse data across multiple WGS platforms, including Oxford Nanopore MinION. RESULTS: We present a new command line version of the TBProfiler webserver, which includes hetero-resistance calling and will facilitate the batch processing of samples. The TBProfiler database has been expanded to incorporate 178 new markers across 16 anti-tuberculosis drugs. The predictive performance of the mutation library has been assessed using > 17,000 clinical isolates with WGS and laboratory-based drug susceptibility testing (DST) data. An integrated MinION analysis pipeline was assessed by performing WGS on 34 replicates across 3 multi-drug resistant isolates with known resistance mutations. TBProfiler accuracy varied by individual drug. Assuming DST as the gold standard, sensitivities for detecting multi-drug-resistant TB (MDR-TB) and extensively drug-resistant TB (XDR-TB) were 94% (95%CI 93-95%) and 83% (95%CI 79-87%) with specificities of 98% (95%CI 98-99%) and 96% (95%CI 95-97%) respectively. Using MinION data, only one resistance mutation was missed by TBProfiler, involving an insertion in the tlyA gene coding for capreomycin resistance. When compared to alternative platforms (e.g. Mykrobe predictor TB, the CRyPTIC library), TBProfiler demonstrated superior predictive performance across first- and second-line drugs. CONCLUSIONS: The new version of TBProfiler can rapidly and accurately predict anti-TB drug resistance profiles across large numbers of samples with WGS data. The computing architecture allows for the ability to modify the core bioinformatic pipelines and outputs, including the analysis of WGS data sourced from portable technologies. TBProfiler has the potential to be integrated into the point of care and WGS diagnostic environments, including in resource-poor settings
