School Districts’ Assessment of the French Language Proficiency of Prospective FSL Teachers
It is a widespread practice among Canadian school districts to assess the French language proficiency of candidates applying for French as a Second Language (FSL) teaching positions. This pan-Canadian study used surveys, interviews, and website data to examine the French language proficiency assessment practices and processes used by Canadian schools when hiring FSL teachers. The findings show that almost 90% of Canadian school districts assess French language proficiency when hiring FSL teachers and that the most common form of assessment is to ask one to three questions in French (with candidates responding in French) during the employment interview. Evaluation of the candidates’ responses is rarely informed by a language proficiency framework or rubric; instead, evaluators’ decisions are informed by their overall impressions of the candidate’s language proficiency. Other promising assessment practices are described, along with suggestions about how districts may improve their French language proficiency assessments when hiring FSL teachers.Presque tous les districts scolaires canadiens évaluent les compétences en français des candidats lors de l'embauche pour des postes d'enseignants en FLS. Ces évaluations sont conçues pour s'assurer que les enseignants de FLS qu'ils embauchent possèdent les compétences linguistiques requises pour enseigner le français d'une manière efficace. Cette étude a recueilli des données auprès de 112 districts scolaires (situés dans les 10 provinces) et a révélé que bien qu'il existe une gamme d'outils et de processus d'évaluation à travers le pays, la méthode d'évaluation la plus courante consiste pour un administrateur scolaire à poser une ou deux questions en français (avec la candidat répondant en français) lors de l'entretien d'embauche. Nous posons des questions sur la justesse de ces évaluations pour prendre des décisions d'emploi à enjeux élevés et offrons des preuves que les districts scolaires commencent à utiliser ces informations d'évaluation à des fins formatives ainsi que pour les décisions d'embauche
Norms of valence, arousal, dominance, and age of acquisition for 4300 Dutch words
This article presents norms of valence/pleasantness, activity/arousal, power/dominance, and age of acquisition for 4,300 Dutch words, mainly nouns, adjectives, adverbs, and verbs. The norms are based on ratings with a 7-point Likert scale by independent groups of students from two Belgian (Ghent and Leuven) and two Dutch (Rotterdam and Leiden-Amsterdam) samples. For each variable, we obtained high split-half reliabilities within each sample and high correlations between samples. In addition, the valence ratings of a previous, more limited study (Hermans & De Houwer, Psychologica Belgica, 34:115-139, 1994) correlated highly with those of the present study. Therefore, the new norms are a valuable source of information for affective research in the Dutch language
Imaging neural activity in the ventral nerve cord of behaving adult Drosophila
To understand neural circuits that control limbs, one must measure their activity during behavior. Until now this goal has been challenging, because limb premotor and motor circuits have been largely inaccessible for large-scale recordings in intact, moving animals-a constraint that is true for both vertebrate and invertebrate models. Here, we introduce a method for 2-photon functional imaging from the ventral nerve cord (VNC) of behaving adult Drosophila melanogaster. We use this method to reveal patterns of activity across nerve cord populations during grooming and walking and to uncover the functional encoding of moonwalker ascending neurons (MANs), moonwalker descending neurons (MDNs), and a previously uncharacterized class of locomotion-associated A1 descending neurons. Finally, we develop a genetic reagent to destroy the indirect flight muscles and to facilitate experimental access to the VNC. Taken together, these approaches enable the direct investigation of circuits associated with complex limb movements
Prevalence, incidence and recurrence of sexually transmitted infections in HIV-negative adult women in a rural South African setting
Objective: Sexually transmitted infections (STIs), including syphilis, chlamydia, gonorrhoea and trichomoniasis, are of global public health concern. While STI incidence rates in sub-Saharan Africa are high, longitudinal data on incidence and recurrence of STIs are scarce, particularly in rural areas. We determined the incidence rates of curable STIs in HIV-negative women during 96 weeks in a rural South African setting. Methods: We prospectively followed participants enrolled in a randomised controlled trial to evaluate the safety and efficacy of a dapivirine-containing vaginal ring for HIV prevention in Limpopo province, South Africa. Participants were included if they were female, aged 18–45, sexually active, not pregnant and HIV-negative. Twelve-weekly laboratory STI testing was performed during 96 weeks of follow-up. Treatment was provided based on vaginal discharge by physical examination or after a laboratory-confirmed STI. Results: A total of 119 women were included in the study. Prevalence of one or more STIs at baseline was 35.3%. Over 182 person-years at risk (PYAR), a total of 149 incident STIs were diagnosed in 75 (65.2%) women with incidence rates of 45.6 events/PYAR for chlamydia, 27.4 events/100 PYAR for gonorrhoea and 8.2 events/100 PYAR for trichomoniasis. Forty-four women developed ≥2 incident STIs. Risk factors for incident STI were in a relationship ≤3 years (adjusted hazard ratio [aHR]: 1.86; 95% confidece interval [CI]: 1.04–2.65) and having an STI at baseline (aHR: 1.66; 95% CI: 1.17–2.96). Sensitivity and specificity of vaginal discharge for laboratory-confirmed STI were 23.6% and 87.7%, respectively. Conclusion: This study demonstrates high STI incidence in HIV-negative women in rural South Africa. Sensitivity of vaginal discharge was poor and STI recurrence rates were high, highlighting the shortcomings of syndromic management in the face of asymptomatic STIs in this setting

Heart Failure, Female Sex, and Atrial Fibrillation Are the Main Drivers of Human Atrial Cardiomyopathy: Results From the CATCH ME Consortium
Background: Atrial cardiomyopathy (atCM) is an emerging prognostic factor in cardiovascular disease. Fibrotic remodeling, cardiomyocyte hypertrophy, and capillary density are hallmarks of atCM. The contribution of etiological factors and atrial fibrillation (AF) to the development of differential atCM phenotypes has not been quantified. This study aimed to evaluate the association between histological features of atCM and the clinical phenotype. Methods and results: We examined left atrial (LA, n=95) and right atrial (RA, n=76) appendages from a European cohort of patients undergoing cardiac surgery. Quantification of histological atCM features was performed following wheat germ agglutinin/CD31/vimentin staining. The contributions of AF, heart failure, sex, and age to histological characteristics were determined with multiple linear regression models. Persistent AF was associated with increased endomysial fibrosis (LA: +1.13±0.47 μm, P=0.038; RA: +0.94±0.38 μm, P=0.041), whereas total extracellular matrix content was not. Men had larger cardiomyocytes (LA: +1.92±0.72 μm, P<0.001), while women had more endomysial fibrosis (LA: +0.99±0.56 μm, P=0.003). Patients with heart failure showed more endomysial fibrosis (LA: +1.85±0.48 μm, P<0.001) and extracellular matrix content (LA: +3.07±1.29%, P=0.016), and a higher capillary density (LA: +0.13±0.06, P=0.007) and size (LA: +0.46±0.22 μm, P=0.044). Fuzzy k-means clustering of histological features identified 2 subtypes of atCM: 1 characterized by enhanced endomysial fibrosis (LA: +3.17 μm, P<0.001; RA: +2.86 μm, P<0.001), extracellular matrix content (LA: +3.53%, P<0.001; RA: +6.40%, P<0.001) and fibroblast density (LA: +4.38%, P<0.001), and 1 characterized by cardiomyocyte hypertrophy (LA: +1.16 μm, P=0.008; RA: +2.58 μm, P<0.001). Patients with fibrotic atCM were more frequently female (LA: odds ratio [OR], 1.33, P=0.002; RA: OR, 1.54, P=0.004), with persistent AF (LA: OR, 1.22, P=0.036) or heart failure (LA: OR, 1.62, P<0.001). Hypertrophic features were more common in men (LA: OR=1.33, P=0.002; RA: OR, 1.54, P=0.004). Conclusions: Fibrotic atCM is associated with female sex, persistent AF, and heart failure, while hypertrophic features are more common in men
A scientific algorithm to simultaneously retrieve carbon monoxide and methane from TROPOMI onboard Sentinel-5 Precursor
Carbon monoxide (CO) is an important atmospheric constituent affecting air quality, and methane (CH) is the second most important greenhouse gas contributing to human-induced climate change. Detailed and continuous observations of these gases are necessary to better assess their impact on climate and atmospheric pollution. While surface and airborne measurements are able to accurately determine atmospheric abundances on local scales, global coverage can only be achieved using satellite instruments. The TROPOspheric Monitoring Instrument (TROPOMI) onboard the Sentinel-5 Precursor satellite, which was successfully launched in October 2017, is a spaceborne nadirviewing imaging spectrometer measuring solar radiation reflected by the Earth in a push-broom configuration. It has a wide swath on the terrestrial surface and covers wavelength bands between the ultraviolet (UV) and the shortwave infrared (SWIR), combining a high spatial resolution with daily global coverage. These characteristics enable the determination of both gases with an unprecedented level of detail on a global scale, introducing new areas of application. Abundances of the atmospheric column-averaged dry air mole fractions XCO and XCH are simultaneously retrieved from TROPOMI’s radiance measurements in the 2:3 μm spectral range of the SWIR part of the solar spectrum using the scientific retrieval algorithm Weighting Function Modified Differential Optical Absorption Spectroscopy (WFMDOAS). This algorithm is intended to be used with the operational algorithms for mutual verification and to provide new geophysical insights. We introduce the algorithm in detail, including expected error characteristics based on synthetic data, a machine-learning-based quality filter, and a shallow learning calibration procedure applied in the post-processing of the XCH data. The quality of the results based on real TROPOMI data is assessed by validation with ground-based Fourier transform spectrometer (FTS) measurements providing realistic error estimates of the satellite data: the XCO data set is characterised by a random error of 5:1 ppb (5:8 %) and a systematic error of 1:9 ppb (2:1 %); the XCH data set exhibits a random error of 14:0 ppb (0:8 %) and a systematic error of 4:3 ppb (0:2 %). The natural XCO and XCH variations are well-captured by the satellite retrievals, which is demonstrated by a high correlation with the validation data (R = 0:97 for XCO and R D 0:91 for XCH based on daily averages).
We also present selected results from the mission start until the end of 2018, including a first comparison to the operational products and examples of the detection of emission sources in a single satellite overpass, such as CO emissions from the steel industry and CH emissions from the energy sector, which potentially allows for the advance of emission monitoring and air quality assessments to an entirely new level
Sensitive Detection of p65 Homodimers Using Red-Shifted and Fluorescent Protein-Based FRET Couples
BACKGROUND: Fluorescence Resonance Energy Transfer (FRET) between the green fluorescent protein (GFP) variants CFP and YFP is widely used for the detection of protein-protein interactions. Nowadays, several monomeric red-shifted fluorescent proteins are available that potentially improve the efficiency of FRET. METHODOLOGY/PRINCIPAL FINDINGS: To allow side-by-side comparison of several fluorescent protein combinations for detection of FRET, yellow or orange fluorescent proteins were directly fused to red fluorescent proteins. FRET from yellow fluorescent proteins to red fluorescent proteins was detected by both FLIM and donor dequenching upon acceptor photobleaching, showing that mCherry and mStrawberry were more efficient acceptors than mRFP1. Circular permutated yellow fluorescent protein variants revealed that in the tandem constructs the orientation of the transition dipole moment influences the FRET efficiency. In addition, it was demonstrated that the orange fluorescent proteins mKO and mOrange are both suitable as donor for FRET studies. The most favorable orange-red FRET pair was mKO-mCherry, which was used to detect homodimerization of the NF-kappaB subunit p65 in single living cells, with a threefold higher lifetime contrast and a twofold higher FRET efficiency than for CFP-YFP. CONCLUSIONS/SIGNIFICANCE: The observed high FRET efficiency of red-shifted couples is in accordance with increased Förster radii of up to 64 A, being significantly higher than the Förster radius of the commonly used CFP-YFP pair. Thus, red-shifted FRET pairs are preferable for detecting protein-protein interactions by donor-based FRET methods in single living cells
AmiA and AliA peptide ligands, found in Klebsiella pneumoniae, are imported into pneumococci and alter the transcriptome
Klebsiella pneumoniae releases the peptides AKTIKITQTR and FNEMQPIVDRQ, which bind the pneumococcal proteins AmiA and AliA respectively, two substrate-binding proteins of the ABC transporter Ami-AliA/AliB oligopeptide permease. Exposure to these peptides alters pneumococcal phenotypes such as growth. Using a mutant in which a permease domain of the transporter was disrupted, by growth analysis and epifluorescence microscopy, we confirmed peptide uptake via the Ami permease and intracellular location in the pneumococcus. By RNA-sequencing we found that the peptides modulated expression of genes involved in metabolism, as pathways affected were mostly associated with energy or synthesis and transport of amino acids. Both peptides downregulated expression of genes involved in branched-chain amino acid metabolism and the Ami permease; and upregulated fatty acid biosynthesis genes but differed in their regulation of genes involved in purine and pyrimidine biosynthesis. The transcriptomic changes are consistent with growth suppression by peptide treatment. The peptides inhibited growth of pneumococcal isolates of serotypes 3, 8, 9N, 12F and 19A, currently prevalent in Switzerland, and caused no detectable toxic effect to primary human airway epithelial cells. We conclude that pneumococci take up K. pneumoniae peptides from the environment via binding and transport through the Ami permease. This changes gene expression resulting in altered phenotypes, particularly reduced growth
