    Roles of transcriptional and translational control mechanisms in regulation of ribosomal protein synthesis in Escherichia coli

    ABSTRACTBacterial ribosome biogenesis is tightly regulated to match nutritional conditions and to prevent formation of defective ribosomal particles. InEscherichia coli, most ribosomal protein (r-protein) synthesis is coordinated with rRNA synthesis by a translational feedback mechanism: when r-proteins exceed rRNAs, specific r-proteins bind to their own mRNAs and inhibit expression of the operon. It was recently discovered that the second messenger nucleotide guanosine tetra and pentaphosphate (ppGpp), which directly regulates rRNA promoters, is also capable of regulating many r-protein promoters. To examine the relative contributions of the translational and transcriptional control mechanisms to the regulation of r-protein synthesis, we devised a reporter system that enabled us to genetically separate thecis-acting sequences responsible for the two mechanisms and to quantify their relative contributions to regulation under the same conditions. We show that the synthesis of r-proteins from the S20 and S10 operons is regulated by ppGpp following shifts in nutritional conditions, but most of the effect of ppGpp required the 5â€Č region of the r-protein mRNA containing the target site for translational feedback regulation and not the promoter. These results suggest that most regulation of the S20 and S10 operons by ppGpp following nutritional shifts is indirect and occurs in response to changes in rRNA synthesis. In contrast, we found that the promoters for the S20 operon were regulated during outgrowth, likely in response to increasing nucleoside triphosphate (NTP) levels. Thus, r-protein synthesis is dynamic, with different mechanisms acting at different times.IMPORTANCEBacterial cells have evolved complex and seemingly redundant strategies to regulate many high-energy-consuming processes. InE. coli, synthesis of ribosomal components is tightly regulated with respect to nutritional conditions by mechanisms that act at both the transcription and translation steps. In this work, we conclude that NTP and ppGpp concentrations can regulate synthesis of ribosomal proteins, but most of the effect of ppGpp is indirect as a consequence of translational feedback in response to changes in rRNA levels. Our results illustrate how effects of seemingly redundant regulatory mechanisms can be separated in time and that even when multiple mechanisms act concurrently their contributions are not necessarily equivalent.</jats:p

    The evolution of the bi-modal colour distribution of galaxies in SDSS groups

    We analyse u−ru-r colour distributions for several samples of galaxies in groups drawn from the Fourth Data Release of the Sloan Digital Sky Survey. For all luminosity ranges and environments considered the colour distributions are well described by the sum of two Gaussian functions. We find that the fraction of galaxies in the red sequence is an increasing function of group virial mass. We also study the evolution of the galaxy colour distributions at low redshift, z≀0.18z\le0.18 in the field and in groups for galaxies brighter than Mr−5log⁥(h)=−20M_r-5\log(h)=-20, finding significant evidence of recent evolution in the population of galaxies in groups. The fraction of red galaxies monotonically increases with decreasing redshift, this effect implies a much stronger evolution of galaxies in groups than in the field.Comment: 7 pages, 6 figures, submited to MNRAS after minor revisio

    Redox supercapacitor performance of nanocrystalline molybdenum nitrides obtained by ammonolysis of chloride- and amide-derived precursors

    Reactions of MoCl5 or Mo(NMe2)4 with ammonia result in cubic ?-Mo2N or hexagonal ?1-MoN depending on reaction time and temperature. At moderate temperatures the cubic product from Mo(NMe2)4 exhibits lattice distortions. Fairly high surface areas are observed in the porous particles of the chloride-derived materials and high capacitances of up to 275 F g?1 are observed when electrodes made from them are cycled in aqueous H2SO4 or K2SO4 electrolytes. The cyclic voltammograms suggest charge is largely stored in the electrochemical double layer at the surface of these materials. Amide-derived molybdenum nitrides have relatively low surface areas and smaller capacitances, but do exhibit strong redox features in their cyclic voltammograms, suggesting that redox capacitance is responsible for a significant proportion of the charge stored

    Normal ordering and boundary conditions in open bosonic strings

    Boundary conditions play a non trivial role in string theory. For instance the rich structure of D-branes is generated by choosing appropriate combinations of Dirichlet and Neumann boundary conditions. Furthermore, when an antisymmetric background is present at the string end-points (corresponding to mixed boundary conditions) space time becomes non-commutative there. We show here how to build up normal ordered products for bosonic string position operators that satisfy both equations of motion and open string boundary conditions at quantum level.     Standard bootstrap method is used to generate confidence intervals (CIs) of impulse response functions of VAR and SVAR models in the pork sector. In the VAR model, the bootstrap method does not produce significant different results from Monte Carlo simulations. In the SVAR analysis, on the other hand, the bootstrap CIs are significantly different from Monte Carlo CIs after a six period forecast intervals. This suggests that the choice of method used to measure reliability of IRFs is not trivial. Furthermore, bootstrap CIs in SVAR model seem to be more stable than MC CIs, which tend to be wider in the longer horizons.Research Methods/ Statistical Methods,

    Nitrogen-rich transition metal nitrides

    The solid state chemistry leading to the synthesis and characterization of metal nitrides with N:M ratios &gt;1 is summarized. Studies of these compounds represent an emerging area of research. Most transition metal nitrides have much lower nitrogen contents, and they often form with non- or sub-stoichiometric compositions. These materials are typically metallic with often superconducting properties, and they provide highly refractory, high hardness materials with many technological applications. The higher metal nitrides should achieve formal oxidation states (OS) attaining those found among corresponding oxides, and they are expected to have useful semiconducting properties. Only a very few examples of such high OS nitrogen-rich compounds are known at present. The main group elements typically form covalently bonded nitride ceramics such as Si3N4, Ge3N4 and Sn3N4, and the early transition metals Zr and Hf produce Zr3N4 and Hf3N4. However, the only main example of a highly nitrided transition metal compound known to date is Ta3N5 that has a formal oxidation state +5 and is a semiconductor with visible light absorption leading to applications as a pigment and in photocatalysis. New synthesis routes are being explored to study the possible formation of other N-rich materials that are predicted to exist by ab initio calculations. There is a useful interplay between theoretical predictions and experimental synthesis studies at ambient and high pressure conditions, as we explore and establish the existence and structure–property relations of these new nitride compounds and polymorphs. Here we review the state of current investigations and indicate possible new directions for further work
