Both in cytological and histological specimen, specific fluorescence of LDH-H and LDH-M were seen in the cytoplasm of cancer cells, and in most cases no remarkable difference could be demonstrated between the distribution of H and M subunit. But in some cases specific fluorescence of M-subunit was stronger than that of H-subunit. The fluorescence of M-subunit was demonstrated as fine granules diffusely in the cytoplasm. On the other hand the fluorescence of H-subunit was rather localized and demonstrated as rather coarse granules. And this observation was discussed

    Ascitic Bile Acids in Patients with Liver Cirrhosis

    Ascitic bile acids of 8 patients with liver cirrhosis were analyzed in order to elucidate the clinical significance. The results were as follows. 1) Ascitic bile acid concentrations in intractable ascites were high, although low in tractable ascites. 2) Among the 8 patients, 4 had serum bile acid analysis performed and a significant correlation of bile acid concentration was found between ascites and serum (P<0.01). 3) Ascitic bile acid concentration was positively correlative with ascitic protein concentration (P(0.05). 4) In one patient whose group separation on Piperidinohydroxypropyl Sephadex LH-20 was determined, free bile acids were found to predominate the major part of the ascitic bile acids. These results suggest that ascitic bile acids reflect serum bile acids, and that bile acids with protein are filtered into ascitic fluid. Further patient studies will be necessary to shed light on the relationship between intractability of ascites and high ascitic bile acid concentrations

    The effect of continuous arterial infusion of Gabexate Mesilate (FOY-007) on experimental acute pancreatitis

    The effectiveness of continuous arterial infusion of Gabexate Mesilate (FOY-007) on experimental acute pancreatitis was investigated. Acute necrotizing pancreatitis was induced by an injection of 10% Na-taurocholate (1ml/kg) into the main pancreatic duct of mongrel dogs. Animals were divided into three groups Group A: non-treated control, Group B: after the induction of pancreatitis, injected with FOY-007 intravenously (5mg/kg/hr), Group C: after the induction of pancreatitis, injectedwithFOY-007 via the celiac artery. The changes in the values of amylase and lipase in serum and ascites etc. were examined. A histological examination was done and the FOY-007 concentration of the pancreas was measured. In both groups B and C, the serum levels of amylase and lipase reached significantly to low levels compared with those in group A. The extents of pancreatic parenchyma necrosis in each group were 36.1, 25.3 and 19.5% , respectively, and were significantly improved in group C. In addition, theFOY-007levels in pancreas specimens in the intraarterial infusion group exceeded those in the intravenous infusion group by 32 times. The results suggest that continuous FOY-007 arterial infusion therapy is useful as a local treatment for severe acute pancreatitis

    Effects of Bepridil on Spiral Reentry

    Bepridil is effective for conversion of atrial fibrillation to sinus rhythm and in the treatment of drug-refractory ventricular tachyarrhythmias. We investigated the effects of bepridil on electrophysiological properties and spiral-wave (SW) reentry in a 2-dimensional ventricular muscle layer of isolated rabbit hearts by optical mapping. Ventricular tachycardia (VT) induced in the presence of bepridil (1 μM) terminated earlier than in the control. Bepridil increased action potential duration (APD) by 5% – 8% under constant pacing and significantly increased the space constant. There was a linear relationship between the wavefront curvature (κ) and local conduction velocity: LCV = LCV0 − D·κ (D, diffusion coefficient; LCV0, LCV at κ = 0). Bepridil significantly increased D and LCV0. The regression lines with and without bepridil crossed at κ = 20 – 40 cm−1, resulting in a paradoxical decrease of LCV at κ > 40 cm−1. Dye transfer assay in cultured rat cardiomyocytes confirmed that bepridil increased intercellular coupling. SW reentry in the presence of bepridil was characterized by decremental conduction near the rotation center, prominent drift, and self-termination by collision with boundaries. These results indicate that bepridil causes an increase of intercellular coupling and a moderate APD prolongation, and this combination compromises wavefront propagation near the rotation center of SW reentry, leading to its drift and early termination

    Reproducibility and validity of food group intake in a short food frequency questionnaire for the middle-aged Japanese population

    Purpose: The purpose of this study was to evaluate the reproducibility and validity of a short food frequency questionnaire (FFQ) for food group intake in Japan, the reproducibility and partial validity of which were previously confirmed for nutrients. Methods: A total of 288 middle-aged healthy volunteers from 11 different areas of Japan provided nonconsecutive 3-day weighed dietary records (DRs) at 3-month intervals over four seasons. We evaluated reproducibility based on the first (FFQ1) and second (FFQ2) questionnaires and their validity against the DRs by comparing the intake of 20 food groups. Spearman’s rank correlation coefficients (SRs) were calculated between energy-adjusted intake from the FFQs and that from the DRs. Results: The intake of 20 food groups estimated from the two FFQs was mostly equivalent. The median energy-adjusted SRs between the FFQ1 and FFQ2 were 0.61 (range 0.38–0.86) for men and 0.66 (0.45–0.84) for women. For validity, the median de-attenuated SRs between DRs and the FFQ1 were 0.51 (0.17–0.76) for men and 0.47 (0.23–0.77) for women. Compared with the DRs, the proportion of cross-classification into exact plus adjacent quintiles with the FFQ1 ranged from 58 to 86% in men and from 57 to 86% in women. According to the robust Z scores and the Bland–Altman plot graphs, the underestimation errors in the FFQ1 tended to be greater in individuals with high mean levels of consumption for meat for men and for other vegetables for both men and women. Conclusion: The FFQ demonstrated high reproducibility and reasonable validity for food group intake. This questionnaire is short and remains appropriate for identifying associations between diet and health/disease among adults in Japan

    The relationship between histopathology of liver and preoperative liver functional data was studied in 151 cases with cholelithiasis to clarify how gallstones effect on the hepatic tissues. The results were as follows. 1. Histologically, 80% of all the cases showed hepatic inflammation. 2. The group with cholecystolithiasis had the highest frequency of hepatic parenchymatous changes. 3. The data of liver functions in the group with cholecystolithiasis were found to be almost within normal limits. Conversely, hepatolithiasis group showed to be moderate to severe disorder. 4. Although both groups with stasis and cirrhosis had hepatic functional disorder, the former showed to be moderate to severe, the latter to be normal to mild, in the magnitude of liver functional disorder. As a conclusion, the grade of liver functional disorder in cholelithiasis patients depends upon that of biliary stasis
