    Process mapping of laser surface modification of AISI 316L stainless steel for biomedical applications

    A 1.5-kW CO2 laser in pulsed mode at 3 kHz was used to investigate the effects of varied laser process parameters and resulting morphology of AISI 316L stainless steel. Irradiance and residence time were varied between 7.9 to 23.6 MW/cm2 and 50 to 167 Āµs respectively. A strong correlation between irradiance, residence time, depth of processing and roughness of processed steel was established. The high depth of altered microstructure and increased roughness were linked to higher levels of both irradiance and residence times. Energy fluence and surface temperature models were used to predict levels of melting occurring on the surface through the analysis of roughness and depth of the region processed. Microstructural images captured by the SEM revealed significant grain structure changes at higher irradiances, but due to increased residence times, limited to the laser in use, the hardness values were not improved

    Reactions of [Mo(CO)6] under vacuum in Ī±-cages of Na56Y zeolite fully loaded with chemisorbed PMe3 form cis-[Mo(CO)4(PMe3)2] but mixtures of [Mo(CO)5(PMe3)] and cis-[Mo(CO)4(PMe3)2] are formed under CO. Reactions under vacuum exhibit low enthalpic and very negative entropic factors (Ī”H = 71.4 Ā± 3.5 kJ mol-1 and Ī”S = -102 Ā± 11 J K-1 mol-1) compared with much higher enthalpic and positive entropic factors for CO dissociative reactions with P(n-Bu)3 in xylene observed elsewhere. Reaction at 66 Ā°C under vacuum is ca.103 times faster than spontaneous CO dissociative reactions in solution. Intrazeolite substitution is concluded to occur by a zeolite assisted mechanism in which two oxide ions in the cavity walls simultaneously displace two neighbouring CO ligands from the [Mo(CO)6]. This contrasts with even lower enthalpy, and more negative entropy values, for simultaneous displacement of three neighbouring CO ligands in thermal decarbonylation reactions. The Ī±-cages behave as multidentate anionic zeolate ligands with varying numbers of O2- ions participating to create highly ordered transition states. These results emphasize the high degree to which such kinetic studies can reveal intimate details of the nature of these activating effects

    Utility of TERT Promoter Mutations for Cutaneous Primary Melanoma Diagnosis

    Telomerase reverse transcriptase (TERT) promoter mutations are commonly found in malignant melanomas but rare in melanocytic nevi. To assess its potential diagnostic utility for the distinction of melanoma from nevus, we determined the TERT promoter mutation status of 86 primary melanomas, 72 melanocytic nevi, and 40 diagnostically problematic melanocytic proliferations. Of the 86 melanomas, 67 (77.9%) were TERT-positive, defined as harboring a hotspot TERT promoter mutation at positions -124C>T, -124_125CC>TT, -138_139CC>TT, or -146C>T. Of the 72 nevi, only 1 (1.4%) was TERT-positive. Of the 40 diagnostically uncertain melanocytic proliferations, 2 (5.0%) were TERT-positive. TERT positivity as a test for melanoma versus nevus had an accuracy of 87.3% [95% confidence interval (CI), 81.1-92.1], a sensitivity of 77.9% (95% CI, 68.9-85.4), a specificity of 98.6% (95% CI, 95.8-100), a positive predictive value of 98.5% (95% CI, 95.6-100), and a negative predictive value of 78.9% (95% CI, 72.6-85.4). Our results indicate that hotspot TERT promoter mutation status may be a useful ancillary parameter for the diagnosis of melanoma. In particular, the high specificity of these mutations for melanoma indicates the presence of a TERT promoter mutation in a melanocytic neoplasm associated with diagnostic controversy, or uncertainty should increase concern for a melanoma

    Five new genes are important for common polysaccharide antigen biosynthesis in <em>Pseudomonas aeruginosa</em>

    ABSTRACT Common polysaccharide antigen (CPA) is a conserved cell surface polysaccharide produced by Pseudomonas aeruginosa. It contains a rhamnan homopolymer and is one of the two forms of O polysaccharide attached to P. aeruginosa lipopolysaccharide (LPS). Our laboratory has previously characterized an eight-gene cluster (pa5447-pa5454 in P. aeruginosa PAO1) required for biosynthesis of CPA. Here we demonstrate that an adjacent five-gene cluster pa5455-pa5459 is also involved. Using reverse transcriptase PCR (RT-PCR), we showed that the original eight-gene cluster and the new five-gene cluster are both organized as operons. We have analyzed the LPS phenotypes of in-frame deletion mutants made in each of the five genes, and the results verified that these five genes are indeed required for CPA biosynthesis, extending the CPA biosynthesis locus to contain 13 contiguous genes. By performing overexpression experiments of different sets of these biosynthesis genes, we were able to obtain information about their possible functions in CPA biosynthesis. IMPORTANCE Lipopolysaccharide (LPS) is an important cell surface structure of Gram-negative bacteria. The human opportunistic pathogen Pseudomonas aeruginosa simultaneously produces an O-antigen-specific (OSA) form and a common polysaccharide antigen (CPA) form of LPS. CPA, the focus of this study, is composed of Ī±-1-2, Ī±1-3-linked d-rhamnose sugars and has been shown to be important for attachment of the bacteria to human airway epithelial cells. Genome sequencing of this species revealed a new five-gene cluster that we predicted to be involved in CPA biosynthesis and modification. In this study, we have generated chromosomal knockouts by performing in-frame deletions and allelic replacements. Characterizing the function of each of the five genes is important for us to better understand CPA biosynthesis and the mechanisms of chain length termination and regulation of this unique D-rhamnan polysaccharide

    Contributions from SUSY-FCNC couplings to the interpretation of the HyperCP events for the decay \Sigma^+ \to p \mu^+ \mu^-

    The observation of three events for the decay Ī£+ā†’pĪ¼+Ī¼āˆ’\Sigma^+ \to p \mu^+ \mu^- with a dimuon invariant mass of 214.3Ā±0.5214.3\pm0.5MeV by the HyperCP collaboration imply that a new particle X may be needed to explain the observed dimuon invariant mass distribution. We show that there are regions in the SUSY-FCNC parameter space where the A10A^0_1 in the NMSSM can be used to explain the HyperCP events without contradicting all the existing constraints from the measurements of the kaon decays, and the constraints from the K0āˆ’KĖ‰0K^0-\bar{K}^0 mixing are automatically satisfied once the constraints from kaon decays are satisfied.Comment: 18 pages, 7 figure

    Prominent bulk pinning effect in the MgB_2 superconductor

    We report the magnetic-field dependence of the irreversible magnetization of the recently discovered binary superconductor MgB2_{2}. For the temperature region of T<0.9TcT< 0.9T_c, the contribution of the bulk pinning to the magnetization overwhelms that of the surface pinning. This was evident from the fact that the magnetization curves, M(H)M(H), were well described by the critical-state model without considering the surface pinning effect. It was also found that the M(H)M(H) curves at various temperatures scaled when the field and the magnetization were normalized by the characteristic scaling factors Hāˆ—(T)H^\ast(T) and Māˆ—(T)M^\ast(T), respectively.     Identification of a Robust Methylation Classifier for Cutaneous Melanoma Diagnosis

    Early diagnosis improves melanoma survival, yet the histopathological diagnosis of cutaneous primary melanoma can be challenging, even for expert dermatopathologists. Analysis of epigenetic alterations, such as DNA methylation, that occur in melanoma can aid in its early diagnosis. Using a genome-wide methylation screening, we assessed CpG methylation in a diverse set of 89 primary invasive melanomas, 73 nevi, and 41 melanocytic proliferations of uncertain malignant potential, classified based on interobserver review by dermatopathologists. Melanomas and nevi were split into training and validation sets. Predictive modeling in the training set using ElasticNet identified a 40-CpG classifier distinguishing 60 melanomas from 48 nevi. High diagnostic accuracy (area under the receiver operator characteristic curve = 0.996, sensitivity = 96.6%, and specificity = 100.0%)was independently confirmed in the validation set (29 melanomas, 25 nevi)and other published sample sets. The 40-CpG melanoma classifier included homeobox transcription factors and genes with roles in stem cell pluripotency or the nervous system. Application of the 40-CpG melanoma classifier to the diagnostically uncertain samples assigned melanoma or nevus status, potentially offering a diagnostic tool to assist dermatopathologists. In summary, the robust, accurate 40-CpG melanoma classifier offers a promising assay for improving primary melanoma diagnosis

    Particle Motion Around Tachyon Monopole

    Recently, Li and Liu have studied global monoole of tachyon in a four dimensional static space-time. We analyze the motion of massless and massive particles around tachyon monopole. Interestingly, for the bending of light rays due to tachyon monopole instead of getting angle of deficit we find angle of surplus. Also we find that the tachyon monopole exerts an attractive gravitational force towards matter.Comment: 14 pages, 7 figure
