Immunohistochemiczna detekcja receptorów dopaminowych D2 w guzach neuroendokrynnych
Background: Recently, dopamine D2 receptors (RD2) have been found to be expressed in neuroendocrine tumours (NET), the tumours
which arise from the diffuse neuroendocrine cells. Moreover, successful trials of the treatment of NET with cabergoline — D2 agonist,
have been reported. These findings increase the interest of investigating RD2 expression in NET.
Material and methods: The expression of RD2 was investigated immunohistochemically using the antibody which recognises both short
(S) and long (L) isoforms of the receptor in 17 NET samples taken from 15 patients.
Results: In 17 NET samples, a positive reaction with the anti-RD2 antibody occurred in 11 cases. In six cases, the localisation of the immunostaining
was cytoplasmic and in nine cases it was nuclear. Only in one case was the receptor cell membrane-located, and in two
cases the immunoreaction was also localised in the blood vessels walls. The relation between RD2 expression and the grade of malignancy
examined by means of Ki-67 antigen expression needs further study. However, preliminary observations indicate that the nuclear localisation
of RD2 is linked to higher tumour malignancy. The next investigated question was the co-expression of somatostatin and dopamine
receptors. This question seems important because of the perspectives of somatostatin-dopamine chimeras application in NET treatment.
In the samples examined by us, RD2 were co-expressed in 5/10 cases with sstr1, in 3/10 with sstr2A, in 2/9 with sstr2B, in 3/10 with sstr3,
and in 5/10 with sstr5.
Conclusion: Dopamine D2 receptors are revealed by means of immunohistochemistry in the majority of NET. They exhibit cytoplasmic
and/or nuclear localisations, the latter being possibly linked to a higher grade of malignancy, and are often co-expressed with somatostatin
receptors (mostly with subtypes1 and 5). (Pol J Endocrinol 2011; 62 (5): 388–391)Wstęp: Ostatnio wykryto występowanie receptorów dopaminowych D2 (RD2) w guzach neuroendokrynnych (NET) i doniesiono o skutecznych
próbach leczenia NET kabergoliną — agonistą tych receptorów. Zwiększa to znaczenie badań nad ekspresją RD2 w NET.
Materiał i metody: Zbadano 17 wycinków guzów NET pobranych od 15 pacjentów. Badania immunohistochemiczne przeprowadzono
z użyciem przeciwciała rozpoznającego zarówno krótką (S), jak i długą (L) izoformę receptora D2.
Wyniki: Pozytywny odczyn z przeciwciałem anty-RD2 występował w 11 spośród 17 badanych przypadków NET. W 6 na 17 badanych wycinków
był to odczyn o lokalizacji cytoplazmatycznej, w 9 na 17 odczyn jądrowy, a tylko w jednym przypadku odczyn o lokalizacji błonowej. Ponadto
w 2 przypadkach obserwowano odczyn w ścianach naczyń krwionośnych. Podjęto próbę oceny zależności między stopniem złośliwości
guza, określonym na podstawie badania antygenu Ki-67, a ekspresją RD2. Wstępne obserwacje wskazują, że lokalizacja jądrowa odczynu na
RD2 wiąże się z wyższym stopniem złośliwości nowotworu. Zależność ta wymaga dalszych badań na większym materiale. Ponadto zbadano
współwystępowanie receptorów somatostatynowych i dopaminowych. Zagadnienie to ma istotne znaczenie ze względu na perspektywy
stosowania chimer somatostatynowo-dopaminowych w leczeniu NET. W badanych przez autorów pracy guzach cytoplazmatyczne receptory
D2 występowały w 5 spośród 10 przypadków, łącznie z sstr1, w 3/10 z sstr2A, w 2/9 z sstr 2B, 3/10 z sstr3 i 5/10 z sstr5.
Wnioski: Receptory dopaminowe D2 wykrywa się za pomocą metody immunohistochemicznej w większości NET. Mają one lokalizację
cytoplazmatyczną i/lub jądrową. Lokalizacja jądrowa wydaje się wiązać z większym stopniem złośliwości nowotworu. RD2 wykazują
często koekspresję z receptorami somatostatynowymi, zwłaszcza podtypami 1 i 5.(Endokrynol Pol 2011; 62 (5): 388–391
Expression of α-Klotho protein in human thyroid cancers — an immunohistochemical study
Introduction: The α-Klotho protein was discovered as a gene controlling the process of aging, but further studies indicated that it also plays the role of a tumour suppressor. Although numerous studies were performed on the role of the α-Klotho gene and protein in neoplasia, the data on α-Klotho protein expression in thyroid cancers are very scarce. Our study presents the immunohistochemical investigation of α-Klotho expression in benign and malignant thyroid tumours.
Material and methods: The material included samples of benign (nodular hyperplasia, follicular adenoma), differentiated (follicular and papillary) cancers and aggressive thyroid cancers of low differentiation grade. The samples were immunostained using two different monoclonal anti-α-Klotho antibodies.
Results: From the two antibodies used in this study, one (EPR6856) reacted probably with the soluble form of Klotho and immunostained mostly the colloid filling thyroid follicles and intravascular or extravascular serum deposits. The other (A-9 antibody) immunostained the follicular epithelium in benign thyroid lesions as well as the epithelial tumoural cells in differentiated thyroid (follicular and papillary cancers). In the thyroid cancers of high malignancy, the immunostaining with A-9 anti-α-Klotho antibody was (except in one case) negative or very weak.
Conclusion: Our results indicate that lowered expression of a- Klotho is involved in the process of thyroid neoplasia

Introduction: The overexpression of nitric oxide synthase (NOS) has been found in tumours, including pituitary adenomas. It has also
been found that NOS is overexpressed in human spontaneous pituitary adenomas. The question arises whether NOS and its product,
nitric oxide (NO), are involved in pituitary tumourigenesis. To investigate this question, in the present paper we examine the effects of
NOS inhibition on the development of diethylstilbestrol (DES)-induced prolactin–secreting pituitary tumours in rats.
Material and methods: Thirty male Fisher 344 rats, four weeks old, were submitted to subcutaneous implantation of a silastic capsule
containing DES (10 mg/capsule) or of an empty capsule. Six weeks after implantation, some of the DES-treated animals received a NOS
inhibitor, N-nitro-l-arginine methyl ester (NAME), 1 mg/mL, in their drinking water, for the subsequent 14 days. Eight weeks after the
implantation, all the animals were sacrificed, their pituitaries were weighed, and samples of heart blood were collected for prolactin (PRL)
and vascular endothelial growth factor (VEGF) measurements.
Results: It was found that DES implantation significantly increased pituitary mass, as well as PRL and VEGF concentrations in blood
serum. On the other hand, the administration of NAME did not affect significantly either VEGF concentration or pituitary mass. On the
other hand, it did induce a further increase in PRL levels.
Conclusions: These findings indicate that NO is involved in oestrogen-induced hyperprolactinaemia, but does not play a crucial role in
oestrogen-induced pituitary tumourigenesis. (Pol J Endocrinol 2012; 63 (2): 115–118)Wstęp: W licznych nowotworach, w tym także gruczolakach przysadki, stwierdzono zwiększoną ekspresję syntazy tlenku azotu (NOS).
Powstaje pytanie, czy NOS oraz jej produkt — tlenek azotu (NO) biorą udział w patogenezie guzów przysadki. W celu zbadania tego
zagadnienia autorzy niniejszej pracy prześledzili wpływ zahamowania aktywności NOS na rozwój prolaktynowego guza przysadki
wywołanego dietylstilbestrolem (DES) u szczurów.
Materiał i metody: Czterotygodniowym szczurom samcom szczepu Fisher 344 implantowano podskórnie silastikowe kapsułki zawierające
10 mg DES w kapsułce lub puste kapsułki. Sześć tygodni po implantacji część zwierząt, którym implantowano kapsułki z DES, otrzymywała
inhibitor NOS — ester metylowy N-nitro-l-argininy (NAME) w wodzie do picia, w stężeniu 1 mg/ml, przez 14 dni. Osiem tygodni po
implantacji kapsułek zwierzęta uśmiercono, pobrano i zważono przysadki oraz próbki krwi z serca w celu oznaczenia stężeń prolaktyny
(PRL) i czynnika wzrostu śródbłonków naczyniowych (VEGF).
Wyniki: Stwierdzono, że implantacja DES znamiennie zwiększa masę przysadki oraz stężenia PRL i VEGF w surowicy krwi. Równoczesne
podawanie NAME nie ma istotnego wpływu na masę przysadki ani stężenie VEGF, natomiast powoduje dalszy wzrost stężenia PRL.
Wnioski: Uzyskane wyniki wskazują, że NO wpływa na hiperprolaktynemię indukowaną DES, ale nie odgrywa istotnej roli w rozwoju

Somatostatin receptors in human adrenal gland tumors--immunohistochemical study.

Somatostatin receptors in GH-secreting pituitary adenomas - their relationship to the response to octreotide
Zbadano 20 gruczolaków przysadki, usuniętych operacyjne u chorych z akromegalią. Na skrawkach parafinowych guzów wykonano
odczyny immunohistochemiczne z użyciem przeciwciał anty-GH i anty-PRL oraz przeciwciał przeciwko poszczególnym podtypom receptorów
somatostatynowych (rsst1–5). Nasilenie odczynu na rsst oceniano z użyciem następującej skali: 0 - odczyn negatywny,
1 - odczyn słaby, 2 - odczyn umiarkowany, 3 - odczyn silny. U 15 chorych zbadano korelacje między nasileniem odczynów na rsst
a odpowiedzią GH na podanie 200 mg oktreotydu. Wszystkie badane guzy były immunopozytywne dla GH, a większość wykazywała
także ko-ekspresję PRL (14/20). Wszystkie badane gruczolaki wykazywały ekspresję rsst2A (20/20) i rsst5 (12/12). Receptory sst2B i rsst3
występowały także we wszystkich badanych guzach poza jednym (odpowiednio: 19/20 i 11/12). W żadnym z badanych w tym kierunku
guzów nie stwierdzono obecności rsst4. Mieszane gruczolaki (GH/PRL) wykazywały tendencje do wyższej ekspresji rsst2A + rsst2B
i zarazem silniejszej odpowiedzi na oktreotyd, niż guzy monohormonalne. Stwierdzono znamienną korelację dodatnią między sumą
ekspresji rsst2A + rsst2B a spadkiem GH po podaniu oktreotydu. Podsumowując, hamujący wydzielanie GH efekt oktreotydu zależy od
nasilenia ekspresji receptorów sst2A i sst2B. Obie izoformy receptora sst2 w guzach wydzielających hormon wzrostu pośredniczą w tej
samej odpowiedzi biologicznej (hamowaniu wydzielania GH).Twenty pituitary adenomas, surgically removed from patients suffering from acromegaly, were studied. The tumours were immunostained
with anti-GH and anti-PRL antibodies and with antibodies raised against particular subtypes of somatostatin receptors (rsst1–5).
Expression of rsst immunoreactivity was scored using the following scale: 0 - negative reaction, 1 - weak reaction, 2 - moderate
reaction, 3 - strong reaction. In 15 patients in whom the GH response to the acute administration of 200 ug octreotide was tested the
correlation between the expression of rsst and the percentage of GH drop was estimated. All the tumours were GH-immunopositive and
in the majority (14/20) co-expression of PRL was also found. All the adenomas examined expressed rsst2A (20/20) and rsst5 (12/12) receptor
proteins. Receptors sst2B and rsst3 were found in all but one of the tumours examined (19/20 and 11/12, respectively). None of tumours
investigated presented rsst4 immunopositivity. The mixed (GH/PRL) adenomas showed a tendency to a higher expression of rsst2A +
+ rsst2B and a greater response to octreotide administration. A significant positive correlation was found between rsst2A + rsst2B expressions
and a drop in GH after octreotide.
To conclude, the GH-inhibiting effect of octreotide depends on the intensity of expression of both rsst2A and rsst2B. Both isoforms of rsst2
mediate the same biological response (inhibition of GH secretion) in GH-secreting and GH/PRL-secreting adenomas
Does the response of GH-secreting pituitary adenomas to octreotide depend on the cellular localization of the somatostatin receptor subtypes SSTR2 and SSTR5?
Wstęp: Badania immunohistochemicze ekspresji podtypów receptora somatostatynowego (SSTR1-5, somatostatin receptor 1-5) w guzach
endokrynnych wykazują ich obecność zarówno w błonie komórkowej, jak i na terenie cytoplazmy komórek guza. Szeroko stosowany
obecnie syntetyczny analog somatostatyny - oktreotyd wykazuje powinowactwo głównie do podtypów SSTR2 i SSTR5. W naszych
wcześniejszych badaniach zauważono pozytywną korelację między zsumowanym wskaźnikiem (score) nasilenia odczynu immunohistochemicznego
SSTR2A i 2B, a hamowaniem sekrecji hormonu wzrostu (GH, growth hormone) po podaniu oktreotydu u chorych z akromegalią,
niezależnie od lokalizacji receptorów w komórkach guza. Obecne badania autorów pracy dotyczą ewentualnej współzależności pomiędzy
hamowaniem GH w odpowiedzi na oktreotyd a komórkową lokalizacją SSTR2A, 2B i 5.
Materiał i metody: Zbadano 13 chorych z akromegalią, u których przed operacją wykonano krótki test z oktreotydem. W gruczolakach
przysadki określono immunohistochemicznie ich fenotyp hormonalny oraz występowanie i lokalizację podtypów SSTR.
Wyniki: W większości przypadków obserwowano cytoplazmatyczną ekspresję badanych podtypów SSTR, aczkolwiek występowała również
lokalizacja błonowa i mieszana błonowo-cytoplazmatyczna. Spadek wydzielenia GH w teście hamowania oktreotydem zawierał się
w przedziale pomiędzy 57,1-96,7% (średnio 82,1%). Największy znamienny spadek wydzielenia GH (92,0 ± 7,0%) po podaniu oktreotydu
wystąpił u pacjentów z cytoplazmatyczną lokalizacją SSTR5 w komórkach guza. Zauważono także tendencję do silniejszej odpowiedzi
u pacjentów, u których również SSTR2A i 2B miały położenie cytoplazmatyczne (odpowiednio 86,8% i 87,0%).
Wnioski: Wydaje się, że cytoplazmatyczne, a nie błonowe położenie SSTR2A, 2B i 5, jest powiązane z silniejszym hamowaniem GH
w teście z oktreotydem. Możliwe, że oktreotyd w procesie internalizacji receptora zmienia jego położenie z błonowego na cytoplazmatyczne
i zwiększa jego immunopozytywność w cytoplazmie badanych komórek guza.
(Endokrynol Pol 2010; 61 (2): 178-181)Introduction: The immunohistochemical examination of somatostatin receptor (SSTR) subtypes expression in different endocrine tumours,
including pituitary adenomas, revealed membranous or cytoplasmic distribution of SSTR1-5. Currently used long-acting somatostatin
analogue octreotide prefers SSTR2 and SSTR5 subtypes. In an earlier study a positive correlation between the summarized score of
SSTR2A + SSTR2B expressions and growth hormone (GH) response to octreotide administration was found, independently of receptor
distribution within the cell. In this study we searched for the relationship between the GH inhibitory response to acute octreotide administration
and SSTR2A, SSTR2B, and SSTR5 cellular localization.
Material and methods: Thirteen acromegalic patients underwent a test of acute administration of octreotide before surgery. The drop in
GH was defined as the percentage of the basal value. The pituitary adenomas from these patients were immunostained to determine the
hormonal phenotype and expression of SSTR subtypes. The subcellular distribution pattern of SSTR subtypes - membranous or cytoplasmic
- was determined.
Results: In the majority of specimens, cytoplasmic localization of receptor subtypes was observed, although membrane or mixed cytoplasmicmembranous
localized immunopositivity also occurred. The drop in GH after octreotide administration varied between 57.1-96.7% (mean
82.1%). Among the patients with the cytoplasmic localization of SSTR5, the decrease in GH was significantly higher (92.0 ± 7.0%). A tendency
towards the higher response in patients with cytoplasmic localization of SSTR2A and 2B was also observed (86.8% and 87.0%, respectively).
Conclusions: It seems that cytoplasmic localization of SSTR5, SSTR2A, and SSTR2B is connected with enhanced GH inhibition after
octreotide administration. It is possible that this somatostatin analogue alters the localization of subtypes SSTR2A and SSTR2B through the
receptor internalization. As a consequence, the SSTR-immunopositivity in cell cytoplasm is increased. The cytoplasmic but not the membranous
localization is connected with the higher responsiveness to the octreotide in somatotropinomas.

Ekspresja różnych podtypów receptora somatostatynowego w gruczolakach przysadki - badania immunohistochemiczne
Background: The highly variable expression of SSTR subtypes in pituitary adenomas (PA) may partially explain why the subgroup of
somatotropinomas or other adenomas do not respond to the therapeutic action of currently used long-acting somatostatin analogues like
octreotide or lanreotide.
Material and methods: Our study summarizes the data on expression of all somatostatin receptor subtypes (SSTR 1-5), extended for
2A and 2B SSTR isoforms, revealed by means of immunohistochemistry in dependence to different hormonal phenotype of the tumour.
Results: The pattern of SSTR immunostaining (estimated according to the percentage frequency of appearance) was in acromegaly: SSTR 5
> SSTR 1 > SSTR 2A = SSTR 3 > SSTR 2B, in prolactinomas: SSTR 2B = SSTR 3 = SSTR 5 > SSTR 1 = SSTR 2A, in gonadotropinomas:
SSTR 3 > SSTR 2B > SSTR 1 = SSTR 2A > SSTR 5, in corticotropinomas: SSTR 2A > SSTR 1 = SSTR 3 > SSTR 5 > SSTR 2B.
In PA immunonegative for pituitary hormones, we noticed only a weak staining of all receptor subtypes including SSTR 4. In plurihormonal
adenomas with positive GH phenotype the staining pattern was: SSTR 5 > SSTR 1 = SSTR 2B and in plurihormonal PA with
negative GH phenotype: SSTR 1 = SSTR 5 > SSTR 2A = SSTR 2B = SSTR 3. In plurihormonal adenoma with ACTH immunopositivity,
the staining pattern was: SSTR = SSTR 2A = SSTR 3 = SSTR 5. SSTR 1 and SSTR 5 were the most frequent subtypes of somatostatin
receptor in plurihormonal adenomas without ACTH expression.
Conclusions: Human PA represents a group of tumours with a much more differentiated appearance of somatostatin receptor subtypes.
It is very important to determine the SSTR profile individually for each tumour to make an appropriate decision as to therapeutic strategy
choice. Apart from applying SSTR 2 and SSTR 5-preferring octreotide and lanreotide - newly synthesized multiligand analogues, such as
SOM 230, KE 108, or other SST selective analogues, may represent a further useful approach for the treatment, especially in cases other
than somatotropinoma or thyrotropinoma.Wstęp: Duża różnorodność ekspresji podtypów receptora somatostatynowego SSTR 1-5 w gruczolakach przysadki (PA) może częściowo
wyjaśniać, dlaczego w guzach somatotropowych lub innych obserwuje się brak odpowiedzi na działanie analogów somatostatyny
o przedłużonym działaniu, takich jak octreotyd i lanreotyd, wiążących się głównie z podtypem receptora 2 i 5.
Materiał i metody: W pracy podsumowano wyniki badania metodą immunohistochemiczną ekspresji wszystkich podtypów receptora
somatostatynowego SSTR 1-5, łącznie z izoformami 2A i 2B, w zależności od fenotypu hormonalnego guza.
Wyniki: Wzory immunoekspresji SSTR 1-5, określone na podstawie procentowej częstości występowania, przedstawiały się następująco:
w akromegalii - SSTR 5 > SSTR 1 > SSTR 2A = SSTR 3 > SSTR 2B, w guzach prolaktynowych: SSTR 2B = SSTR 3 = SSTR 5 > SSTR 1 =
= SSTR 2A, w guzach gonadotropowych - SSTR 3 > SSTR 2B > SSTR 1 = SSTR 2A > SSTR 5, w guzach kortykotropowych - SSTR 2A
> SSTR 1 = SSTR 3 > SSTR 5 > SSTR 2B. W guzach immunonegatywnych dla hormonów przysadkowych zanotowano jedynie słaby
odczyn dla wszystkich podtypów receptora, łącznie z SSTR 4. W guzach wielohormonalnych GH-dodatnich, ekspresja receptora była
następująca - SSTR 5 > SSTR 1 = SSTR 2B, a w guzach wielohormonalnych GH-ujemnych - SSTR 1 = SSTR 5 > SSTR 2A = SSTR 2B = SSTR 3. W guzach wielohormonalnych o fenotypie ACTH-dodatnim - SSTR 1 = SSTR 2A = SSTR 3 = SSTR 5.
W guzach wielohormonalnych ACTH-ujemnych najczęściej występowały SSTR 1 i SSTR 5.
Wnioski: Podtypy receptora somatatostatynowego SSTR 1 i SSTR 5 występują w ludzkich gruczolakach przysadki w sposób bardzo
zróżnicowany. Dlatego też, aby podjąć właściwą decyzję dotyczącą sposobu ich leczenia, należy określić indywidualny dla każdego guza,
profil SSTR. Nowo zsyntetyzowane multiligandy, takie jak SOM 230, KE 180 lub selektywne analogi, mogą być użyteczne w leczeniu
zwłaszcza innych guzów, niż te wydzielające GH lub TSH
Immunohistochemical detection of FSH receptors in pituitary adenomas and adrenal tumors
<p><strong> </strong></p><p><strong><em>Objectives</em></strong>. Follicle stimulating hormone (FSH) receptors (FSHR) are physiologically expressed in the ovary and testis. It is well known that FSHR are also expressed in gonadal cancers, but the data on their incidence in extra-gonadal tumors are scarce. Recently, the expression of FSHR in the vascular endothelium within different human cancers was found, but nothing is known on FSHR appearance in non-gonadal endocrine tumors. The present paper reports on the immunohistochemical detection of FSHR in human pituitary adenomas and adrenal tumors.</p><p><strong>Materials and methods</strong>. The study included samples of 28 pituitary adenomas and 36 adrenal tumors. Moreover, 2 samples of non-tumoral adrenal glands were also studied.</p><p>FSH receptor immunostaining was performed on paraffin sections using the rabbit anti-human FSH-R polyclonal antibody raised against 1-190 amino acid sequence from the human FSH-R (sc-13935). The pituitary adenomas were immunostained to reveal the pituitary hormones and the proliferation marker Ki-67.</p><p><strong>Results</strong>. In the pituitary adenomas, positive immunostaining with anti-FSHR antibody occurred in the adenoma cells cytoplasm and endothelia of the intra- and peritumoral blood vessels. The cytoplasmic immunostaining was found in the majority of investigated tumors but the intensity of staining was weak to moderate. There is some tendency towards the higher cytoplasmic FSHR score in tumors with higher Ki-67 index (atypical adenomas). In contrast to the cytoplasm, the FSHR immunostaining in blood vessels is strong and concerns all the investigated samples. Strong FSHR immunostaining is present in the endothelium of intra- and/or peritumoral blood vessels in the majority of pheochromocytomas, approximatively one half of the adrenocortical adenomas and both cases of the adrenal cancers. The immunostaining is detectable also in the tumoral cell cytoplasm in all but one examined pheochromocytomas.. All the investigated adrenocortical adenomas presented strong immunostaining of cell membranes. No immunostained cell membranes were found. in adrenal cancers. The positive immunostaining was found in glandular cells, but not in blood vessels, of non-tumoral adrenal cortex and medulla.</p><strong>Conclusions</strong>. Immunostaining of FSHR often occurs in the endothelium of intra- and/or peritumoral blood vessels of pituitary adenomas and benign and malignant adrenal tumors. The immunostaining may be also present in tumoral cells. A role of FSHR expression in these tumors (stimulation of angiogenesis? stimulation of cell growth?) needs further studies to be clarified
Expression of somatostatin receptor subtypes in human thyroid tumors: the immunohistochemical and molecular biology (RT-PCR) investigation
Human endocrine tumors often express the somatostatin receptors SSTR 1–5 with different intensity. It has been widely investigated their distribution in pituitary adenomas, brain tumors, adrenal tumors and neuroendocrine tumors in gastrointestinal tract (NET). Some of studies also concern the expression of SSTRs in thyroid tumors but they are mainly limited to parafollicular C cells – derived medullary thyroid carcinomas (MTC). Results of SSTR 1–5 detection in other thyroid pathologies like follicular adenomas and papillary cancers are still scarce and often controversial, depending of investigation method used. The aim of this study was to report the presence of all the 5 subtypes of SSTR (including 2A and 2B SSTR isoforms) in some surgically treated human thyroid tumors by means of immunohistochemistry and real-time PCR method and to correlate the results obtained with both techniques. SSTR 1 protein was expressed in 88.8% of investigated cases, SSTR 2A and 2B both in 44.4%, SSTR 3 in 55.5%, SSTR 4 in 11.2% and SSTR 5 in 33.3%. SSTR 1 is the dominant form in the thyroid gland tumor and hyperplasia. We found positive confirmation of both methods in 88.8% for SSTR 1, 2A, 3 subtypes, in 22.2% for SSTR 4 and in 100% for SSTR 5. It suggests that somatostatin multiligand analogs or selective SSTR 1 agonists may be used in thyroid tumors treatment
Immunohistochemical detection of FSH receptors in pituitary adenomas and adrenal tumors
Objectives. Follicle stimulating hormone (FSH) receptors (FSHR) are physiologically expressed in the ovary and testis. It is well known that FSHR are also expressed in gonadal cancers, but the data on their incidence in extra-gonadal tumors are scarce. Recently, the expression of FSHR in the vascular endothelium within different human cancers was found, but nothing is known on FSHR appearance in non-gonadal endocrine tumors. The present paper reports on the immunohistochemical detection of FSHR in human pituitary adenomas and adrenal tumors.Materials and methods. The study included samples of 28 pituitary adenomas and 36 adrenal tumors. Moreover, 2 samples of non-tumoral adrenal glands were also studied.FSH receptor immunostaining was performed on paraffin sections using the rabbit anti-human FSH-R polyclonal antibody raised against 1-190 amino acid sequence from the human FSH-R (sc-13935). The pituitary adenomas were immunostained to reveal the pituitary hormones and the proliferation marker Ki-67.Results. In the pituitary adenomas, positive immunostaining with anti-FSHR antibody occurred in the adenoma cells cytoplasm and endothelia of the intra- and peritumoral blood vessels. The cytoplasmic immunostaining was found in the majority of investigated tumors but the intensity of staining was weak to moderate. There is some tendency towards the higher cytoplasmic FSHR score in tumors with higher Ki-67 index (atypical adenomas). In contrast to the cytoplasm, the FSHR immunostaining in blood vessels is strong and concerns all the investigated samples. Strong FSHR immunostaining is present in the endothelium of intra- and/or peritumoral blood vessels in the majority of pheochromocytomas, approximatively one half of the adrenocortical adenomas and both cases of the adrenal cancers. The immunostaining is detectable also in the tumoral cell cytoplasm in all but one examined pheochromocytomas.. All the investigated adrenocortical adenomas presented strong immunostaining of cell membranes. No immunostained cell membranes were found. in adrenal cancers. The positive immunostaining was found in glandular cells, but not in blood vessels, of non-tumoral adrenal cortex and medulla.Conclusions. Immunostaining of FSHR often occurs in the endothelium of intra- and/or peritumoral blood vessels of pituitary adenomas and benign and malignant adrenal tumors. The immunostaining may be also present in tumoral cells. A role of FSHR expression in these tumors (stimulation of angiogenesis? stimulation of cell growth?) needs further studies to be clarified
