    City branding and industrial transformation from manufacturing to services: Which pathways do cities in Central China follow?

    A potentially attractive way for cities to maintain economic growth while reducing environmental harm is to let their production structures undergo industrial transformation, a process otherwise known as ecological modernization. This attraction lies mainly in the fact that residents, visitors and corporations prefer clean air, water and soil as a milieu to invest their resources in. Municipal governments can use city branding as an important instrument to force off such a transformation, if it is taken as a point of departure for the adoption of a strategy to which they are deeply committed and for the benefit of which they are willing to deploy their various policy instruments. In the literature on ecological modernization, five different pathways for industrial transformation in cities have been identified and these have been matched with city branding practices. In this contribution, the abovementioned conceptual framework is further detailed and specified to account for a variety in types of secondary and tertiary sector industries. In the empirical sections, all cities in the Chinese provinces Hubei and Hunan, where the transition from manufacturing to services is typically most pressing, are examined in terms of their industrial structures, pathways to industrial transformation and city branding choices. The results indicate, inter alia, that further subdivision of the secondary and tertiary economic sectors is useful in understanding key features of the transformation, and that different sub-pathways affect tradeoffs between economic expansion and ecological preservation differently. Branding practices among Hubei and Hunan cities also indicate that some industries are more easily embraced and utilized than others in establishing brand identities and adopting popular city labels

    Temporal Asynchrony of Trophic Status Between Mainstream and Tributary Bay Within a Giant Dendritic Reservoir: The Role of Local-Scale Regulators

    Limnologists have regarded temporal coherence (synchrony) as a powerful tool for identifying the relative importance of local-scale regulators and regional climatic drivers on lake ecosystems. Limnological studies on Asian reservoirs have emphasized that climate and hydrology under the influences of monsoon are dominant factors regulating seasonal patterns of lake trophic status; yet, little is known of synchrony or asynchrony of trophic status in the single reservoir ecosystem. Based on monthly monitoring data of chlorophyll a, transparency, nutrients, and nonvolatile suspended solids (NVSS) during 1-year period, the present study evaluated temporal coherence to test whether local-scale regulators disturb the seasonal dynamics of trophic state indices (TSI) in a giant dendritic reservoir, China (Three Gorges Reservoir, TGR). Reservoir-wide coherences for TSICHL, TSISD, and TSITP showed dramatic variations over spatial scale, indicating temporal asynchrony of trophic status. Following the concept of TSI differences, algal productivity in the mainstream of TGR and Xiangxi Bay except the upstream of the bay were always limited by nonalgal turbidity (TSICHL−TSISD <0) rather than nitrogen and phosphorus (TSICHL−TSITN <0 and TSICHL−TSITP <0). The coherence analysis for TSI differences showed that local processes of Xiangxi Bay were the main responsible for local asynchrony of nonalgal turbidity limitation levels. Regression analysis further proved that local temporal asynchrony for TSISD and nonalgal turbidity limitation levels were regulated by local dynamics of NVSS, rather than geographical distance. The implications of the present study are to emphasize that the results of trophic status obtained from a single environment (reservoir mainstream) cannot be extrapolated to other environments (tributary bay) in a way that would allow its use as a sentinel site

    Transgene Expression Is Associated with Copy Number and Cytomegalovirus Promoter Methylation in Transgenic Pigs

    Transgenic animals have been used for years to study gene function, produce important proteins, and generate models for the study of human diseases. However, inheritance and expression instability of the transgene in transgenic animals is a major limitation. Copy number and promoter methylation are known to regulate gene expression, but no report has systematically examined their effect on transgene expression. In the study, we generated two transgenic pigs by somatic cell nuclear transfer (SCNT) that express green fluorescent protein (GFP) driven by cytomegalovirus (CMV). Absolute quantitative real-time PCR and bisulfite sequencing were performed to determine transgene copy number and promoter methylation level. The correlation of transgene expression with copy number and promoter methylation was analyzed in individual development, fibroblast cells, various tissues, and offspring of the transgenic pigs. Our results demonstrate that transgene expression is associated with copy number and CMV promoter methylation in transgenic pigs

    Structural insights into anion selectivity and activation mechanism of LRRC8 volume-regulated anion channels

    Summary: Volume-regulated anion channels (VRACs) are hexamers of LRRC8 proteins that are crucial for cell volume regulation. N termini (NTs) of the obligatory LRRC8A subunit modulate VRACs activation and ion selectivity, but the underlying mechanisms remain poorly understood. Here, we report a 2.8-Ã… cryo-electron microscopy structure of human LRRC8A that displays well-resolved NTs. Amino-terminal halves of NTs fold back into the pore and constrict the permeation path, thereby determining ion selectivity together with an extracellular selectivity filter with which it works in series. They also interact with pore-surrounding helices and support their compact arrangement. The C-terminal halves of NTs interact with intracellular loops that are crucial for channel activation. Molecular dynamics simulations indicate that low ionic strength increases NT mobility and expands the radial distance between pore-surrounding helices. Our work suggests an unusual pore architecture with two selectivity filters in series and a mechanism for VRAC activation by cell swelling

    An efficient evolutionary multi-objective framework for MEMS design optimisation: validation, comparison and analysis

    The application of multi objective evolutionary algorithms (MOEA) in the design optimisation of microelectromechanical systems (MEMS) is of particular interest in this research. MOEA is a class of soft computing techniques of biologically inspired stochastic algorithms, which have proved to outperform their conventional counterparts in many design optimisation tasks. MEMS designers can utilise a variety of multi-disciplinary design tools that explore a complex design search space, however, still follow the traditional trial and error approaches. The paper proposes a novel framework, which couples both modelling and analysis tools to the most referenced MOEAs (NSGA-II and MOGA-II). The framework is validated and evaluated through a number of case studies of increasing complexity. The research presented in this paper unprecedentedly attempts to compare the performances of the mentioned algorithms in the application domain. The comparative study shows significant insights into the behaviour of both of the algorithms in the design optimisation of MEMS. The paper provides extended discussions and analysis of the results showing, overall, that MOGA-II outperforms NSGA-II, for the selected case studies
