Genetische Anpassung einer natuerlichen Bakterienpopulation im Litoral der Deutschen Bucht an erhoehte Schwermetallgehalte Abschlussbericht

Investigations were carried out on the propagation of bacterial resistance towards lead and cadmium, the identification of these organisms and the elucidation of typical marine resistance mechanisms. To this purpose bacteria samplings from North Sea station 927 (low anthropogenic impact) and 933 (high anthropogenic impact) have been isolated and purified. The low specifity of genomic probes could be significantly improved by two methods: a) restriction of genomic DNA, DNA transfer to membranes with subsequent hybridization. Hybridization and resistance data provide information of plasmid- or chromosome-encoded heavy metal resistance. b) for the first time amplification products of the polymerase chain reaction have been used after marking as probes for the identification of marine isolates. Further investigations were carried out on the role of Cd resistance by the cation efflux system of plasmid pMOL30 from Alcaligenes eutrophus. For introduction of the unsaturated erucid acid into the sn-2 position of glycerol during the triacylglycerol biosynthesis in developing rape seeds, the 1-acylglycerol-3-phosphate acyltransferase (1-Acyl-GPAT) of rape has to be exchanged by the corresponding seed specific enzyme from limnanthes douglasii. Acyltransferases from rape and limnanthes can be isolated by heterologic complementation and by in situ enzymassay based on the functional expression of the cDNAs in yeast cells. Both methods have been developed and optimized avoiding thus the difficult enzyme purification step.
