    Involution of the mouse mammary gland is associated with an immune cascade and an acute-phase response, involving LBP, CD14 and STAT3

    INTRODUCTION: Involution of the mammary gland is a complex process of controlled apoptosis and tissue remodelling. The aim of the project was to identify genes that are specifically involved in this process. METHODS: We used Affymetrix oligonucleotide microarrays to perform a detailed transcript analysis on the mechanism of controlled involution after withdrawal of the pups at day seven of lactation. Some of the results were confirmed by semi-quantitative reverse transcriptase polymerase chain reaction, Western blotting or immunohistochemistry. RESULTS: We identified 145 genes that were specifically upregulated during the first 4 days of involution; of these, 49 encoded immunoglobulin genes. A further 12 genes, including those encoding the signal transducer and activator of transcription 3 (STAT3), the lipopolysaccharide receptor (CD14) and lipopolysaccharide-binding protein (LBP), were involved in the acute-phase response, demonstrating that the expression of acute-phase response genes can occur in the mammary gland itself and not only in the liver. Expression of LBP and CD14 was upregulated, at both the RNA and protein level, immediately after pup withdrawal; CD14 was strongly expressed in the luminal epithelial cells. Other genes identified suggested neutrophil activation early in involution, followed by macrophage activation late in the process. Immunohistochemistry and histological staining confirmed the infiltration of the involuting mammary tissue with neutrophils, plasma cells, macrophages and eosinophils. CONCLUSION: Oligonucleotide microarrays are a useful tool for identifying genes that are involved in the complex developmental process of mammary gland involution. The genes identified are consistent with an immune cascade, with an early acute-phase response that occurs in the mammary gland itself and resembles a wound healing process

    Proteus: A Hierarchical Portfolio of Solvers and Transformations

    In recent years, portfolio approaches to solving SAT problems and CSPs have become increasingly common. There are also a number of different encodings for representing CSPs as SAT instances. In this paper, we leverage advances in both SAT and CSP solving to present a novel hierarchical portfolio-based approach to CSP solving, which we call Proteus, that does not rely purely on CSP solvers. Instead, it may decide that it is best to encode a CSP problem instance into SAT, selecting an appropriate encoding and a corresponding SAT solver. Our experimental evaluation used an instance of Proteus that involved four CSP solvers, three SAT encodings, and six SAT solvers, evaluated on the most challenging problem instances from the CSP solver competitions, involving global and intensional constraints. We show that significant performance improvements can be achieved by Proteus obtained by exploiting alternative view-points and solvers for combinatorial problem-solving.Comment: 11th International Conference on Integration of AI and OR Techniques in Constraint Programming for Combinatorial Optimization Problems.     Constraints on Baryon-Nonconserving Yukawa Couplings in a Supersymmetric Theory

    The 1-loop evolution of couplings in the minimal supersymmetric standard model, extended to include baryon nonconserving (B ⁣ ⁣ ⁣/)(B\!\!\!/) operators through explicit RR-parity violation, is considered keeping only B ⁣ ⁣ ⁣/B\!\!\!/ superpotential terms involving the maximum possible number of third generation superfields. If all retained Yukawa couplings YiY_i are required to remain in the perturbative domain (Yi<1)(Y_i < 1) upto the scale of gauge group unification, upper bounds ensue on the magnitudes of the B ⁣ ⁣ ⁣/B\!\!\!/ coupling strengths at the supersymmetry breaking scale, independent of the model of unification. They turn out to be similar to the corresponding fixed point values reached from a wide range of YiY_i (including all YiY_i greater than unity) at the unification scale.     The influence of biochar position in a leach bed system anaerobically digesting chicken litter

    As a consequence of the rapidly growing poultry industry, chicken litter is becoming an abundant and problematic waste. Anaerobic digestion of chicken litter can mitigate environmental issues while producing valuable by-products. Recent studies have shown that leach bed reactor (LBR) systems are suitable for processing chicken litter and that anaerobic digestion can be enhanced using biochar. This study investigates the influence of biochar position within an LBR system on anaerobic digestion of chicken litter. Compared to a system without biochar, application of biochar in both the LBR (mixed in with the feedstock or as a layer below the feedstock) and coupled leachate tank (LT) increased methane yield by 6 to 8% at 51 days and accelerated VFA degradation and methane production. More significant differences in methane yield were observed at shorter solid retention times. Biochar mixed in feedstock in addition to a filter in the LT performed best in terms of both methane and hydrogen sulfide production, with a 77% reduction in hydrogen sulfide yield and hydrogen sulfide contents maintained below 500 ppm. The enhanced rates of VFA degradation and methane production when applying biochar in both reactors corresponds with observed differences in the methanogen population. Biochar application in both reactors increased the abundance of Methanobacteriales in digestate and Methanosarcinaceae in leachate compared to the control. Microbial attachment and activity on biochar also increased when mixed in feedstock. Increased diversity of the methanogen population throughout the system, as well as increased activity on biochar, may have facilitated the syntrophic relationship between acetogenic bacteria and methanogens, thus accelerating VFA degradation and methane production. These results suggest mixing biochar in feedstock, in addition to a biochar filter in the LT, to enhance anaerobic digestion of chicken litter in this system.Ben A. Collins, Cristian H.     b-physics signals of the lightest CP-odd Higgs in the NMSSM at large tan beta

    We investigate the low energy phenomenology of the lighter pseudoscalar A10A_1^0 in the NMSSM. The A10A_1^0 mass can naturally be small due to a global U(1)RU(1)_R symmetry of the Higgs potential, which is only broken by trilinear soft terms. The A10A_1^0 mass is further protected from renormalization group effects in the large tanβ\tan \beta limit. We calculate the bsA10b \to s A_1^0 amplitude at leading order in tanβ\tan \beta and work out the contributions to rare KK, BB and radiative Υ\Upsilon-decays and BBˉB -\bar B mixing. We obtain constraints on the A10A_1^0 mass and couplings and show that masses down to O(10){\cal{O}}(10) MeV are allowed. The bb-physics phenomenology of the NMSSM differs from the MSSM in the appearance of sizeable renormalization effects from neutral Higgses to the photon and gluon dipole operators and the breakdown of the MSSM correlation between the Bsμ+μB_s \to \mu^+ \mu^- branching ratio and BsBˉsB_s - \bar B_s mixing. For A10A_1^0 masses above the tau threshold the A10A_1^0 can be searched for in bsτ+τb \to s \tau^+ \tau^- processes with branching ratios \lsim 10^{-3}.Comment: 18 pages, 3 figures; references adde

    Uplifted supersymmetric Higgs region

    We show that the parameter space of the Minimal Supersymmetric Standard Model includes a region where the down-type fermion masses are generated by the loop-induced couplings to the up-type Higgs doublet. In this region the down-type Higgs doublet does not acquire a vacuum expectation value at tree level, and has sizable couplings in the superpotential to the tau leptons and bottom quarks. Besides a light standard-like Higgs boson, the Higgs spectrum includes the nearly degenerate states of a heavy spin-0 doublet which can be produced through their couplings to the bb quark and decay predominantly into \tau^+\tau^- or \tau\nu.Comment: 14 pages; Signs in Eqns.     Minimal Composite Higgs Model with Light Bosons

    We analyze a composite Higgs model with the minimal content that allows a light Standard-Model-like Higgs boson, potentially just above the current LEP limit. The Higgs boson is a bound state made up of the top quark and a heavy vector-like quark. The model predicts that only one other bound state may be lighter than the electroweak scale, namely a CP-odd neutral scalar. Several other composite scalars are expected to have masses in the TeV range. If the Higgs decay into a pair of CP-odd scalars is kinematically open, then this decay mode is dominant, with important implications for Higgs searches. The lower bound on the CP-odd scalar mass is loose, in some cases as low as \sim 100 MeV, being set only by astrophysical constraints.Comment: 33 pages, latex.     Proton-Antiproton Annihilation into a Lambda_c-Antilambda_c Pair

    The process p-pbar -> Lambda_c-Antilambda_c is investigated within the handbag approach. It is shown that the dominant dynamical mechanism, characterized by the partonic subprocess u-ubar -> c-cbar factorizes in the sense that only the subprocess contains highly virtual partons, a gluon to lowest order of perturbative QCD, while the hadronic matrix elements embody only soft scales and can be parameterized in terms of helicity flip and non-flip generalized parton distributions. Modelling these parton distributions by overlaps of light-cone wave functions for the involved baryons we are able to predict cross sections and spin correlation parameters for the process of interest.Comment: 39 pages, 7 figures, problems with printout of figures resolved, Ref. 33 and referring sentences in section 4 change
