    Liver injury and acute liver failure after bariatric surgery an overview of potential injury mechanisms

    The obesity epidemic has caused a surge in the use of bariatric surgery. Although surgery-induced weight loss is an effective treatment of nonalcoholic fatty liver disease, it may precipitate severe hepatic complications under certain circumstances. Acute liver injury (ALI) and acute liver failure (ALF) following bariatric surgery have been reported in several case series. Although rare, ALI and ALF tend to emerge several months after bariatric surgery. If so, it can result in prolonged hospitalization, may necessitate liver transplantation, and in some cases prove fatal. However, little is known about the risk factors for developing ALI or ALF after bariatric surgery and the mechanisms of liver damage in this context are poorly defined. This review provides an account of the available data on ALI and ALF caused by bariatric surgery, with emphasis on potential injury mechanisms and the outcomes of liver transplantation for ALF after bariatric surgery.Cellular mechanisms in basic and clinical gastroenterology and hepatolog

    Moderation of associations between interparental stress and (mal)adaptation by adolescents' personality: Contrasting differential susceptibility and diathesis–stress models

    Objective: This study aimed to contrast differential susceptibility and diathesis–stress models in examining adolescents' Big Five personality dimensions as moderators of longitudinal associations between interparental stress and (mal)adaptation in emerging adulthood (i.e., self-efficacy, externalizing and internalizing behavior). Method: Data from the large longitudinal Flemish Study on Parenting, Personality and Development were used (475 families, adolescents' Mage = 15.82, SDage = 1.15), with both parents reporting on their interparental stress and mothers reporting on the adolescent's personality and in 2009, and emerging adults reporting on their own (mal)adaptive functioning in 2009 and 2015 and their personality in 2015. Results: Multivariate models showed that extraversion, benevolence, emotional stability and imagination were uniquely related to (mal)adaptation across the 6-year interval. In general, our results exhibited no consistent moderating role for adolescents' personality. Only for girls, high levels of extraversion functioned as a “susceptibility maker” in associations between father's interparental stress and self-efficacy, and, low levels of emotional stability functioned as a “vulnerability marker” in associations between parents' interparental stress and self-efficacy. Conclusions: The interaction effects as well as their (restricted) generalizability across gender should be replicated before drawing firm conclusions.     Racial Differences in the Effects of Hormone Therapy on Incident Open-Angle Glaucoma in a Randomized Trial

    Purpose: We conducted a secondary analysis of a randomized, placebo-controlled trial to test if hormone therapy (HT) altered the risk of open-angle glaucoma (OAG), and if the risk reduction varied by race. Design: Secondary analysis of randomized controlled trial data. Methods: We linked Medicare claims data to 25 535 women in the Women's Health Initiative. Women without a uterus were randomized to receive either oral conjugated equine estrogens (CEE 0.625 mg/day) or placebo, and women with a uterus received oral CEE and medroxyprogesterone acetate (CEE 0.625 mg/day + MPA 2.5 mg/day) or placebo. We used Cox proportional hazards models to calculate hazard ratios (HR) and 95% confidence interval. Results: After exclusion of women with prevalent glaucoma or without claims for eye care provider visits, the final analysis included 8102 women (mean age = 68.5 ± 4.8 years). The OAG incidence was 7.6% (mean follow-up = 11.5 ± 5.2 years; mean HT duration = 4.4 ± 2.3 years). Increased age (P trend =.01) and African-American race (HR = 2.69, 95% CI = 2.13–3.42; white as a reference) were significant risk factors for incident OAG. We found no overall benefit of HT in reducing incident OAG (HR = 1.01, 95% CI = 0.79–1.29 in the CEE trial, and HR = 1.05, 95% CI = 0.85–1.29 in the CEE + MPA trial). However, race modified the relationship between CEE use and OAG risk (P interaction =.01), and risk was reduced in African-American women treated with CEE (HR = 0.49, 95% CI = 0.27–0.88), compared to placebo. Race did not modify the relation between CEE + MPA use and OAG risk (P interaction =.68). Conclusions: Analysis suggests that HT containing estrogen, but not a combination of estrogen and progesterone, reduces the risk of incident OAG among African-American women.     Establishment and optimization of genomic selection to accelerate the domestication and improvement of intermediate wheatgrass

Plant Genome, 9(1). doi:10.3835/plantgenome2015.07.0059Intermediate wheatgrass (IWG) is a perennial species and has edible and nutritious grain and desirable agronomic traits, including large seed size, high grain yield, and biomass. It also has the potential to provide ecosystem services and an economic return to farmers. However, because of its allohexaploidy and self-incompatibility, developing molecular markers for genetic analysis and molecular breeding has been challenging. In the present study, using genotyping-by-sequencing (GBS) technology, 3436 genomewide markers discovered in a biparental population with 178 genets, were mapped to 21 linkage groups (LG) corresponding to 21 chromosomes of IWG. Genomic prediction models were developed using 3883 markers discovered in a breeding population containing 1126 representative genets from 58 half-sib families. High predictive ability was observed for seven agronomic traits using cross-validation, ranging from 0.46 for biomass to 0.67 for seed weight. Optimization results indicated that 8 to 10 genets from each half-sib family can form a good training population to predict the breeding value of their siblings, and 1600 genomewide markers are adequate to capture the genetic variation in the current breeding population for genomic selection. Thus, with the advances in sequencing-based marker technologies, it was practical to perform molecular genetic analysis and molecular breeding on a new and challenging species like IWG, and genomic selection could increase the efficiency of recurrent selection and accelerate the domestication and improvement of IWG.A. © Crop Science Society of America

    Associations of autozygosity with a broad range of human phenotypes

    In many species, the offspring of related parents suffer reduced reproductive success, a phenomenon known as inbreeding depression. In humans, the importance of this effect has remained unclear, partly because reproduction between close relatives is both rare and frequently associated with confounding social factors. Here, using genomic inbreeding coefficients (F-ROH) for >1.4 million individuals, we show that F-ROH is significantly associated (p <0.0005) with apparently deleterious changes in 32 out of 100 traits analysed. These changes are associated with runs of homozygosity (ROH), but not with common variant homozygosity, suggesting that genetic variants associated with inbreeding depression are predominantly rare. The effect on fertility is striking: F-ROH equivalent to the offspring of first cousins is associated with a 55% decrease [95% CI 44-66%] in the odds of having children. Finally, the effects of F-ROH are confirmed within full-sibling pairs, where the variation in F-ROH is independent of all environmental confounding.Peer reviewe

    Mass spectrometry in clinical glycomics: the path from biomarker identification to clinical implementation

    Over the past decades, the genome and proteome have been widely explored for biomarker discovery and personalized medicine. However, there is still a large need for improved diagnostics and stratification strategies for a wide range of diseases. Post-translational modification of proteins by glycosylation affects protein structure and function, and glycosylation has been implicated in many prevalent human diseases. Numerous proteins for which the plasma levels are nowadays evaluated in clinical practice are glycoproteins. While the glycosylation of these proteins often changes with disease, their glycosylation status is largely ignored in the clinical setting. Hence, the implementation of glycomic markers in the clinic is still in its infancy. This is for a large part caused by the high complexity of protein glycosylation itself and of the analytical techniques required for their robust quantification. Mass spectrometry-based workflows are particularly suitable for the quantification of glycans and glycoproteins, but still require advances for their transformation from a biomedical research setting to a clinical laboratory. In this review, we describe why and how glycomics is expected to find its role in clinical tests and the status of current mass spectrometry-based methods for clinical glycomics.
