    Relativistic Hartree approach with exact treatment of vacuum polarization for finite nuclei

    We study the relativistic Hartree approach with the exact treatment of the vacuum polarization in the Walecka sigma-omega model. The contribution from the vacuum polarization of nucleon-antinucleon field to the source term of the meson fields is evaluated by performing the energy integrals of the Dirac Green function along the imaginary axis. With the present method of the vacuum polarization in finite system, the total binding energies and charge radii of 16O and 40Ca can be reproduced. On the other hand, the level-splittings in the single-particle level, in particular the spin-orbit splittings, are not described nicely because the inclusion of vacuum effect provides a large effective mass with small meson fields. We also show that the derivative expansion of the effective action which has been used to calculate the vacuum contribution for finite nuclei gives a fairly good approximation.Comment: 15 pages, 8 figure

    Epitope Density Influences CD8+ Memory T Cell Differentiation

    The generation of long-lived memory T cells is critical for successful vaccination but the factors controlling their differentiation are still poorly defined. We tested the hypothesis that the strength of T cell receptor (TCR) signaling contributed to memory CD8(+) T cell generation.We manipulated the density of antigenic epitope presented by dendritic cells to mouse naïve CD8(+) T cells, without varying TCR affinity. Our results show that a two-fold decrease in antigen dose selectively affects memory CD8(+) T cell generation without influencing T cell expansion and acquisition of effector functions. Moreover, we show that low antigen dose alters the duration of the interaction between T cells and dendritic cells and finely tunes the expression level of the transcription factors Eomes and Bcl6. Furthermore, we demonstrate that priming with higher epitope density results in a 2-fold decrease in the expression of Neuron-derived orphan nuclear receptor 1 (Nor-1) and this correlates with a lower level of conversion of Bcl-2 into a pro-apoptotic molecule and an increased number of memory T cells.Our results show that the amount of antigen encountered by naïve CD8(+) T cells following immunization with dendritic cells does not influence the generation of functional effector CD8(+) T cells but rather the number of CD8(+) memory T cells that persist in the host. Our data support a model where antigenic epitope density sensed by CD8(+) T cells at priming influences memory generation by modulating Bcl6, Eomes and Nor-1 expression

    Effect of DMF on liver I/R

    AIM To investigate the hypothesis that treatment with dimethyl fumarate (DMF) may ameliorate liver ischemia/reperfusion injury (I/RI). METHODS Rats were divided into 3 groups: sham, control (CTL), and DMF. DMF (25 mg/kg, twice/d) was orally administered for 2 d before the procedure. The CTL and DMF rats were subjected to ischemia for 1 h and reperfusion for 2 h. The serum alanine aminotransferase (ALT) and malondialdehyde (MDA) levels, adenosine triphosphate (ATP), NO × metabolites, anti-oxidant enzyme expression level, anti-inflammatory effect, and anti-apoptotic effect were determined. RESULTS Histological tissue damage was significantly reduced in the DMF group (Suzuki scores: sham: 0 ± 0; CTL: 9.3 ± 0.5; DMF: 2.5 ± 1.2; sham vs CTL, P < 0.0001; CTL vs DMF, P < 0.0001). This effect was associated with significantly lower serum ALT (DMF 5026 ± 2305 U/L vs CTL 10592 ± 1152 U/L, P = 0.04) and MDA (DMF 18.2 ± 1.4 μmol/L vs CTL 26.0 ± 1.0 μmol/L, P = 0.0009). DMF effectively improved the ATP content (DMF 20.3 ± 0.4 nmol/mg vs CTL 18.3 ± 0.6 nmol/mg, P = 0.02), myeloperoxidase activity (DMF 7.8 ± 0.4 mU/mL vs CTL 6.0 ± 0.5 mU/mL, P = 0.01) and level of endothelial nitric oxide synthase expression (DMF 0.38 ± 0.05-fold vs 0.17 ± 0.06-fold, P = 0.02). The higher expression levels of anti-oxidant enzymes (catalase and glutamate-cysteine ligase modifier subunit and lower levels of key inflammatory mediators (nuclear factor-kappa B and cyclooxygenase-2 were confirmed in the DMF group. CONCLUSION DMF improved the liver function and the anti-oxidant and inflammation status following I/RI. Treatment with DMF could be a promising strategy in patients with liver I/RI

    Regional carbon fluxes from land use and land cover change in Asia, 1980–2009

Available from IOP Publishing via the DOI in this record.We present a synthesis of the land-atmosphere carbon flux from land use and land cover change (LULCC) in Asia using multiple data sources and paying particular attention to deforestation and forest regrowth fluxes. The data sources are quasi-independent and include the U.N. Food and Agriculture Organization-Forest Resource Assessment (FAO-FRA 2015; country-level inventory estimates), the Emission Database for Global Atmospheric Research (EDGARv4.3), the 'Houghton' bookkeeping model that incorporates FAO-FRA data, an ensemble of 8 state-of-the-art Dynamic Global Vegetation Models (DGVM), and 2 recently published independent studies using primarily remote sensing techniques. The estimates are aggregated spatially to Southeast, East, and South Asia and temporally for three decades, 1980–1989, 1990–1999 and 2000–2009. Since 1980, net carbon emissions from LULCC in Asia were responsible for 20%–40% of global LULCC emissions, with emissions from Southeast Asia alone accounting for 15%–25% of global LULCC emissions during the same period. In the 2000s and for all Asia, three estimates (FAO-FRA, DGVM, Houghton) were in agreement of a net source of carbon to the atmosphere, with mean estimates ranging between 0.24 to 0.41 Pg C yr−1, whereas EDGARv4.3 suggested a net carbon sink of −0.17 Pg C yr−1. Three of 4 estimates suggest that LULCC carbon emissions declined by at least 34% in the preceding decade (1990–2000). Spread in the estimates is due to the inclusion of different flux components and their treatments, showing the importance to include emissions from carbon rich peatlands and land management, such as shifting cultivation and wood harvesting, which appear to be consistently underreported.This work was supported by the Asia Pacific Network for Global Change Research (ARCP2013-01CMY-Patra/Canadell).     CD40-Activated B Cells Can Efficiently Prime Antigen-Specific Naïve CD8+ T Cells to Generate Effector but Not Memory T cells

    Background: The identification of the signals that should be provided by antigen-presenting cells (APCs) to induce a CD8 + T cell response in vivo is essential to improve vaccination strategies using antigen-loaded APCs. Although dendritic cells have been extensively studied, the ability of other APC types, such as B cells, to induce a CD8 + T cell response have not been thoroughly evaluated. Methodology/Principal Findings: In this manuscript, we have characterized the ability of CD40-activated B cells, stimulated or not with Toll-like receptor (TLR) agonists (CpG or lipopolysaccharide) to induce the response of mouse naïve CD8 + T cells in vivo. Our results show that CD40-activated B cells can directly present antigen to naïve CD8 + T cells to induce the generation of potent effectors able to secrete cytokines, kill target cells and control a Listeria monocytogenes infection. However, CD40-activated B cell immunization did not lead to the proper formation of CD8 + memory T cells and further maturation of CD40-activated B cells with TLR agonists did not promote the development of CD8 + memory T cells. Our results also suggest that inefficient generation of CD8 + memory T cells with CD40-activated B cell immunization is a consequence of reduced Bcl-6 expression by effectors and enhanced contraction of the CD8 + T cell response. Conclusions: Understanding why CD40-activated B cell immunization is defective for the generation of memory T cells and gaining new insights about signals that should be provided by APCs are key steps before translating the use of CD40-B cel

    Comment on Qian et al. 2008: La Niña and El Niño composites of atmospheric CO2 change

    It is well known that interannual extremes in the rate of change of atmospheric CO2 are strongly influenced by the occurrence of El Niño-Southern Oscillation (ENSO) events. Qian et al. presented ENSO composites of atmospheric CO2 changes. We show that their composites do not reflect the atmospheric changes that are most relevant to understanding the role of ENSO on atmospheric CO2 variability. We present here composites of atmospheric CO2 change that differ markedly from those of Qian et al., and reveal previously unreported asymmetries between the effects on the global carbon system of El Niño and La Niña events. The calendar-year timing differs; La Niña changes in atmospheric CO2 typically occur primarily over September–May, while El Niño changes occur primarily over December–August. And the net concentration change is quite different; La Niña changes are about half the size of El Niño changes. These results illustrate new aspects of the ENSO/global carbon budget interaction and provide useful global-scale benchmarks for the evaluation of Earth System Model studies of the carbon system
