Get PDF
We report a novel superresolution microscopy approach for imaging fluorescence samples. The reported approach, termed virtual k-space modulation optical microscopy (VIKMOM), is able to improve the lateral resolution by a factor of 2, reduce the background level, improve the optical sectioning effect and correct for unknown optical aberrations. In the acquisition process of VIKMOM, we used a scanning confocal microscope setup with a 2D detector array to capture sample information at each scanned x-y position. In the recovery process of VIKMOM, we first modulated the captured data by virtual k-space coding and then employed a ptychography-inspired procedure to recover the sample information and correct for unknown optical aberrations. We demonstrated the performance of the reported approach by imaging fluorescent beads, fixed bovine pulmonary artery endothelial (BPAE) cells, and living human astrocytes (HA). As the VIKMOM approach is fully compatible with conventional confocal microscope setups, it may provide a turn-key solution for imaging biological samples with ∼100  nm lateral resolution, in two or three dimensions, with improved optical sectioning capabilities and aberration correcting.National Institutes of Health (U.S.) (9P41EB015871-26A1)National Institutes of Health (U.S.) (1R01HL121386-01A1

Abstract

Background

Quantitative knowledge on the anatomy of the medial collateral ligament (MCL) is important for treatment of MCL injury and for MCL release during total knee arthroplasty (TKA). The objective of this study was to quantitatively determine the morphology of the MCL of human knees.

Methods

10 cadaveric human knees were dissected to investigate the MCL anatomy. The specimens were fixed in full extension and this position was maintained during the dissection and morphometric measurements. The outlines of the insertion sites of the superficial MCL (sMCL) and deep MCL (dMCL) were digitized using a 3D digitizing system.

Results

The insertion areas of the superficial MCL (sMCL) were 348.6 ± 42.8 mm2 and 79.7 ± 17.6 mm2 on the tibia and femur, respectively. The insertion areas of the deep MCL (dMCL) were 63.6 ± 13.4 mm2 and 71.9 ± 14.8 mm2 on the tibia and femur, respectively. The distances from the centroids of the tibial and femoral insertions of the sMCL to the tibial and femoral joint line were 62.4 ± 5.5 mm and 31.1 ± 4.6 mm, respectively. The distances from the centroids of dMCL in the tibial insertion and the femoral insertion to the tibial and femoral joint line were 6.5 ± 1.3 mm and 20.5 ± 4.2 mm, respectively. The distal portion of the dMCL (meniscotibial ligament - MTL) was approximately 1.7 times wider than the proximal portion of the dMCL (meniscofemoral ligament - MFL), whereas the MFL was approximately 3 times longer than the MTL.

Conclusions

The morphologic data on the MCL may provide useful information for improving treatments of MCL-related pathology and performing MCL release during TKA.</p

Terraces are typical artificial landforms on the Loess Plateau, with ecological functions in water and soil conservation, agricultural production, and biodiversity. Recording the spatial distribution of terraces is the basis of monitoring their extent and understanding their ecological effects. The current terrace extraction method mainly relies on high-resolution imagery, but its accuracy is limited due to vegetation coverage distorting the features of terraces in imagery. High-resolution topographic data reflecting the morphology of true terrace surfaces are needed. Terraces extraction on the Loess Plateau is challenging because of the complex terrain and diverse vegetation after the implementation of “vegetation recovery”. This study presents an automatic method of extracting terraces based on 1 m resolution digital elevation models (DEMs) and 0.3 m resolution Worldview-3 imagery as auxiliary information used for object-based image analysis (OBIA). A multi-resolution segmentation method was used where slope, positive and negative terrain index (PN), accumulative curvature slope (AC), and slope of slope (SOS) were determined as input layers for image segmentation by correlation analysis and Sheffield entropy method. The main classification features based on DEMs were chosen from the terrain features derived from terrain factors and texture features by gray-level co-occurrence matrix (GLCM) analysis; subsequently, these features were determined by the importance analysis on classification and regression tree (CART) analysis. Extraction rules based on DEMs were generated from the classification features with a total classification accuracy of 89.96%. The red band and near-infrared band of images were used to exclude construction land, which is easily confused with small-size terraces. As a result, the total classification accuracy was increased to 94%. The proposed method ensures comprehensive consideration of terrain, texture, shape, and spectrum characteristics, demonstrating huge potential in hilly-gully loess region with similarly complex terrain and diverse vegetation covers.(VLID)219512

A Versatile Method of Engineering the Electron Wavefunction of Hybrid Quantum Devices

With the development of quantum technology, hybrid devices that combine superconductors (S) and semiconductors (Sm) have attracted great attention due to the possibility of engineering structures that benefit from the integration of the properties of both materials. However, until now, none of the experiments have reported good control of band alignment at the interface, which determines the strength of S-Sm coupling and the proximitized superconducting gap. Here, we fabricate hybrid devices in a generic way with argon milling to modify the interface while maintaining its high quality. First, after the milling the atomically connected S-Sm interfaces appear, resulting in a large induced gap, as well as the ballistic transport revealed by the multiple Andreev reflections and quantized above-gap conductance plateaus. Second, by comparing transport measurement with Schr\"odinger-Poisson (SP) calculations, we demonstrate that argon milling is capable of varying the band bending strength in the semiconducting wire as the electrons tend to accumulate on the etched surface for longer milling time. Finally, we perform nonlocal measurements on advanced devices to demonstrate the coexistence and tunability of crossed Andreev reflection (CAR) and elastic co-tunneling (ECT) -- key ingredients for building the prototype setup for realization of Kitaev chain and quantum entanglement probing. Such a versatile method, compatible with the standard fabrication process and accompanied by the well-controlled modification of the interface, will definitely boost the creation of more sophisticated hybrid devices for exploring physics in solid-state systems.Comment: 18 pages, 9 figure

Abstract

Introduction

This report presents a new imaging technique for non-invasive study of six degrees of freedom (DOF) knee kinematics during treadmill gait.

Materials and methods

A treadmill was integrated into a dual fluoroscopic imaging system (DFIS) to formulate a gait analysis system. To demonstrate the application of the system, a healthy subject walked on the treadmill at four different speeds (1.5, 2.0, 2.5 and 3.0 MPH) while the DFIS captured the knee motion during three strides under each speed. Characters of knee joint motion were analyzed in 6DOF during the treadmill walking.

Results

The speed of the knee motion was lower than that of the treadmill. Flexion amplitudes increased with increasing walking speed. Motion patterns in other DOF were not affected by increase in walking speed. The motion character was repeatable under each treadmill speed.

Conclusion

The presented technique can be used to accurately measure the 6DOF knee kinematics at normal walking speeds.</p

Understanding the relationship between land use change and ecosystem service values (ESVs) is the key for improving ecosystem health and sustainability. This study estimated the spatial and temporal variations of ESVs at town scale in relation to land use change in the Loess Plateau which is characterized by its environmental vulnerability, then analyzed and discussed the relationship between ESVs and land use pattern. The result showed that ESVs increased with land use change from 1982 to 2008. The total ESVs increased by 16.17% from US6.315millionat1982toUS 6.315 million at 1982 to US 7.336 million at 2002 before the start of the Grain to Green project, while increased significantly thereafter by 67.61% to US$ 11.275 million at 2008 along with the project progressed. Areas with high ESVs appeared mainly in the center and the east where largely distributing orchard and forestland, while those with low ESVs occurred mainly in the north and the south where largely distributing cropland. Correlation and regression analysis showed that land use pattern was significantly positively related with ESVs. The proportion of forestland had a positive effect on ESVs, however, that of cropland had a negative effect. Diversification, fragmentation and interspersion of landscape positively affected ESVs, while land use intensity showed a negative effect. It is concluded that continuing the Grain to Green project and encouraging diversified agriculture benefit to improve the ecosystem service

which permits unrestricted use, distribution, and reproduction in any medium, provided the original work is properly cited. The contourlet transform can deal effectively with images which have directional information such as contour and texture. In contrast to wavelets for which there exists many good filters, the contourlet filter design for image processing applications is still an ongoing work. Therefore, this paper presents an approach for designing the contourlet filter based on the Chebyshev best uniform approximation for achieving an efficient image denoising applications using hidden Markov tree models in the contourlet domain. Here, we design both the optimal 9/7 wavelet filter banks with rational coefficients and new pkva 12 filter. In this paper, the Laplacian pyramid followed by the direction filter banks decomposition in the contourlet transform using the two filter banks above and the image denoising applications in the contourlet hidden Markov tree model are implemented, respectively. The experimental results show that the denoising performance of the test image Zelda in terms of peak signal-to-noise ratio is improved by 0.33 dB than using CDF 9/7 filter banks with irrational coefficients on the JPEG2000 standard and standard pkva 12 filter, and visual effects are as good as compared with the research results of Duncan D.-Y.
