    High-order rational-type solutions of the analogous (3+1)-dimensional Hirota-bilinear-like equation

    In this article, a new dynamical system equation named the (3+1)-dimensional Hirota-bilinear-like equation (HBLE) was constructed. The generalized Hirota bilinear method was applied to obtain this new HBLE in (3+1) dimensions. This new HBLE possesses a similar bilinear form to the original (3+1)-dimensional Hirota bilinear equation, but with additional nonlinear terms. A set of high-order rational solutions is constructed for the given equation, generated from polynomial solutions to the associated generalized bilinear equation. The analyticity conditions of the resulting solutions were investigated and six groups of general solutions were derived.     Transcriptomic and metabolomic analyses to study the key role by which Ralstonia insidiosa induces Listeria monocytogenes to form suspended aggregates

    Ralstonia insidiosa can survive in a wide range of aqueous environments, including food processing areas, and is harmful to humans. It can induce Listeria monocytogenes to form suspended aggregates, resulting from the co-aggregation of two bacteria, which allows for more persistent survival and increases the risk of L. monocytogenes contamination. In our study, different groups of aggregates were analyzed and compared using Illumina RNA sequencing technology. These included R. insidiosa under normal and barren nutrient conditions and in the presence or absence of L. monocytogenes as a way to screen for differentially expressed genes (DEGs) in the process of aggregate formation. In addition, sterile supernatants of R. insidiosa were analyzed under different nutrient conditions using metabolomics to investigate the effect of nutrient-poor conditions on metabolite production by R. insidiosa. We also undertook a combined analysis of transcriptome and metabolome data to further investigate the induction effect of R. insidiosa on L. monocytogenes in a barren environment. The results of the functional annotation analysis on the surface of DEGs and qPCR showed that under nutrient-poor conditions, the acdx, puuE, and acs genes of R. insidiosa were significantly upregulated in biosynthetic processes such as carbon metabolism, metabolic pathways, and biosynthesis of secondary metabolites, with Log2FC reaching 4.39, 3.96, and 3.95 respectively. In contrast, the Log2FC of cydA, cyoB, and rpsJ in oxidative phosphorylation and ribosomal pathways reached 3.74, 3.87, and 4.25, respectively. Thirty-one key components were identified while screening for differential metabolites, which mainly included amino acids and their metabolites, enriched to the pathways of biosynthesis of amino acids, phenylalanine metabolism, and methionine metabolism. Of these, aminomalonic acid and Proximicin B were the special components of R. insidiosa that were metabolized under nutrient-poor conditions

    SALL4, a Stem Cell Factor, Affects the Side Population by Regulation of the ATP-Binding Cassette Drug Transport Genes

    Our previous work shows that the stem cell factor SALL4 plays a central role in embryonic and leukemic stem cells. In this study, we report that SALL4 expression was higher in drug resistant primary acute myeloid leukemic patients than those from drug-responsive cases. In addition, while overexpression of SALL4 led to drug resistance in cell lines, cells with decreased SALL4 expression were more sensitive to drug treatments than the parental cells. This led to our investigation of the implication of SALL4 in drug resistance and its role in side population (SP) cancer stem cells. SALL4 expression was higher in SP cells compared to non-SP cells by 2–4 fold in various malignant hematopoietic cell lines. Knocking down of SALL4 in isolated SP cells resulted in a reduction of SP cells, indicating that SALL4 is required for their self-renewal. The SP phenotype is known to be mediated by members of the ATP-binding cassette (ABC) drug transport protein family, such as ABCG2 and ABCA3. Using chromatin-immunoprecipitation (ChIP), quantitative reverse transcription polymerase chain reaction (qRT-PCR) and electrophoretic mobility shift assay(EMSA), we demonstrated that SALL4 was able to bind to the promoter region of ABCA3 and activate its expression while regulating the expression of ABCG2 indirectly. Furthermore, SALL4 expression was positively correlated to those of ABCG2 and ABCA3 in primary leukemic patient samples. Taken together, our results suggest a novel role for SALL4 in drug sensitivity, at least in part through the maintenance of SP cells, and therefore may be responsible for drug-resistance in leukemia.     Diagnostic Significance of Serum IgG Galactosylation in CA19-9-Negative Pancreatic Carcinoma Patients

    Background: Although Carbohydrate antigen 19-9 (CA19-9) is considered clinically useful and informative for pancreatic carcinoma (PC), false positive results, and false negative results have restricted its clinical use. Especially missed or delayed diagnosis of PC patients with negative CA19-9 value limited the utility. To improve prognosis of PC patients, the discovery of reliable biomarkers to assist CA19-9 is desired. Serum IgG galactosylation based on our previous report was altered in PC patients comparing to healthy controls. The objective of this study was to explore the diagnostic significance of IgG galactosylation in assisting CA19-9 for PC in a comprehensive way.Methods: Serum IgG galactosylation profiles were analyzed by MALDI-MS in cohort 1 (n = 252) and cohort 2 in which all CA19-9 levels were negative (n = 133). In each cohort, not only healthy controls and PC patients but also benign pancreatic disease (BPD) patients were enrolled. Peaks were acquired by the software of MALDI-MS sample acquisition, followed by being processed and analyzed by the software of Progenesis MALDI. IgG Gal-ratio, which was calculated from the relative intensity of peaks G0, G1, and G2 according to the formula (G0/(G1+G2×2)), was employed as an index for indicating the distribution of IgG galactosylation.Results: The Gal-ratio was elevated in PC comparing with that in non-cancer group (healthy controls and BPD). The area under the receiver operating characteristic curve (AUC) of IgG Gal-ratio was higher than that of CA19-9 (0.912 vs. 0.814). The performance was further improved when Gal-ratio and CA19-9 were combined (AUC: 0.928). Meanwhile, Gal-ratio also had great diagnostic value with a sensitivity of 92.31% (AUC: 0.883) in detection of PC at early stage. Notably, IgG Gal-ratio has great sensitivity (90.63%) and specificity (76.81%) in CA19-9-negative PC patients.Conclusions: IgG Gal-ratio had a great performance in detection of PC and could be used to assist CA19-9 in improving diagnosis performance through early stage detection, differentiation from BPD, and PC diagnosis with CA19-9-negative level

    Mildly relativistic motion in the radio-quiet quasar PG 1351+640

    Measuring the proper motion of the emission component in radio-quiet quasars (RQQs) could help to distinguish between the origins of the radio emission and to understand whether the jet production mechanism is the same in radio-loud quasars and RQQs. PG 1351+640 is one of the few RQQs suitable for proper motion studies: it has two compact components on milli-arcsec scales, a flat-spectrum core and a steep-spectrum jet; both components are ≳2 mJy at 5 GHz and are well suited for Very Long Baseline Array (VLBA) observations. We compare recent VLBA observations with that made seventeen years ago and find no significant change in the core-jet separation between 2005 and 2015 (a proper motion of 0.003 mas yr-1). However, the core-jet separation increased significantly between 2015 and 2022, inferring a jet proper motion velocity of 0.063 mas yr-1, which corresponds to an apparent transverse velocity of. The result suggests that the jet of the RQQ PG 1351+640 is mildly relativistic and oriented at a relatively small viewing angle

    Interactions between the jet and disk wind in a nearby radio intermediate quasar III Zw 2

    Disk winds and jets are ubiquitous in active galactic nuclei (AGN), and how these two components interact remains an open question. We study the radio properties of a radio-intermediate quasar III Zw 2. We detect two jet knots J1 and J2 on parsec scales, which move at a mildly apparent superluminal speed of 1.35c1.35\,c. Two γ\gamma-ray flares were detected in III Zw 2 in 2009--2010, corresponding to the primary radio flare in late 2009 and the secondary radio flare in early 2010. The primary 2009 flare was found to be associated with the ejection of J2. The secondary 2010 flare occurred at a distance of \sim0.3 parsec from the central engine, probably resulting from the collision of the jet with the accretion disk wind. The variability characteristics of III Zw 2 (periodic radio flares, unstable periodicity, multiple quasi-periodic signals and possible harmonic relations between them) can be explained by the global instabilities of the accretion disk. These instabilities originating from the outer part of the warped disk propagate inwards and can lead to modulation of the accretion rate and consequent jet ejection. At the same time, the wobbling of the outer disk may also lead to oscillations of the boundary between the disk wind and the jet tunnel, resulting in changes in the jet-wind collision site. III Zw 2 is one of the few cases observed with jet-wind interactions, and the study in this paper is of general interest for gaining insight into the dynamic processes in the nuclear regions of AGN.Comment: accepted by Ap

    Clinical Study The Association of Weight Status with Physical Fitness among Chinese Children

    Objective. To investigate the association of weight status with physical fitness among Chinese children. Methods. A total of 6929 children aged 6-12 years were selected from 15 primary schools of 5 provincial capital cities in eastern China. The height and fasting body weight were measured. The age-, sex-specific BMI WHO criteria was used to define underweight, overweight and obesity. Physical fitness parameters including standing broad jump, 50 m sprint, and 50 m * 8 shuttle run were tested. Results. The prevalence of underweight, overweight, and obesity was 3.1%, 14.9%, and 7.8%, respectively. Boys performed better than girls, and the older children performed better than their younger counterparts for all physical fitness tests. No significant difference in all three physical fitness tests were found between children with underweight and with normal weight, and they both performed better than their counterparts with overweight and obese in all three physical fitness tests. The likelihood of achieving good performance was much lower among overweight and obese children in comparison with their counterparts with normal weight (OR = 0.13-0.54). Conclusions. An inverse association of obesity with cardiorespiratory fitness, muscle explosive strength, and speed was identified among Chinese children

    The Association of Hypertension with Obesity and Metabolic Abnormalities among Chinese Children

    A total of 8898 Chinese children (4580 boys and 4318 girls) aged 7–13 years in 6 cities of east China were recruited. Data on height, weight, waist circumference, blood pressure, serum lipid profiles, glucose, and insulin were collected. The overall prevalence of hypertension was 11.1%. Overweight and obese children had a higher risk of developing hypertension than their counterparts (29.1%, 17.4%, and 7.8%, resp.) (P=0.0001). The means levels of triglycerides, glucose, insulin, and HOMA-IR (1.0 mmol/L, 4.5 mmol/L, 8.4 mU/mL and 1.7, resp.) among hypertensive children were all significantly higher than their normotensive counterparts (0.8 mmol/L, 4.5 mmol/L, 5.9 mU/mL, and 1.2, resp.) (P=0.0001). Compared with the healthy children, the risk (odds ratio, OR) of having hypertension among children with high triglycerides, hyperglycemia, and metabolic syndrome was 1.4 (95% confidence interval (CI): 1.0–2.0, P=0.0334), 1.5 (95% CI: 0.9–2.5, P=0.0890), and 2.8 (95%CI: 1.5–5.4, P=0.0014), respectively, after controlling for age, gender, BMI, income level, parents' education level and puberty. In conclusion, overweight and obese children have higher risk of having hypertension and children with dyslipidemia, hyperglycemia, and metabolic syndrome and higher HOMA-IR have higher risk of developing hypertension
