    Modulating adipogenesis via signaling pathways and cellular redox state

    The findings of this thesis help to understand how nitric oxide (NO) plays a pivotal role in the cell physiology of human progenitor fat cells (preadipocytes). NO significantly promoted the differentiation process from immature fat cells to mature adipocytes. The proliferation of preadipocytes, however, was inhibited. The results of this thesis are especially helpful in the context of soft tissue engineering in form of preadipocytes embedded in 3-dimensional grafts for reconstructive purposes. These biohybrids represent a promising tool to replace soft tissue defects resulting from deep burns, tumor resections, or congenital deformities to restore the modeling and protecting effects of the subcutaneous fat layer. Future application of nitric oxide-donating agents may then improve the differentiation process and organization of progenitor fat cells in 3-dimensional constructs. Furthermore, NO is a key mediator of inflammatory processes as they may take place at the site of future implantations. The findings of this thesis broaden the understanding of possible complications after transplantation: NO and other inflammatory mediators might inhibit preadipocyte proliferation. Thus, it would be necessary to apply very densely-seeded implants to benefit from the differentiation-promoting NO effect, especially in the center of larger constructs where differentiation is often poor. Moreover, the results of this thesis are generally helpful for future research on adipogenesis and obesity-associated diseases: culture media were improved to fasten the differentiation process from 14 to 8-10 days on an average. An alternative culture medium was created by replacing ingredients of the differentiation medium with potent activators of adenylyl- and guanylylcyclase-dependent intracellular cascades which also facilitated the experimental set up. In the second part of this thesis clozapine, an atypical neuroleptic, and extracts from green tea (EGCG) were analyzed for their potential to influence preadipocyte differentiation. The results of this study show a significant promotion of adipogenesis which could be inhibited by EGCG. This effect is probably due to the radical scavenging effects of clozapine inside the cell which supports the differentiation process of preadipocytes independent from receptor-dependent cascades. The proadipogenic clozapine-effect than leads to an enlarged fat mass which could explain the massive weight gain of patients under clozapine therapy. This side effect often results in bad compliance of the patients and bears the risk of obesity-related diseases. In this study, I tried to find an easy and effective way to minimize the weight gain. A promising substance is extract from green tea (EGCG). Preadipocytes that were treated with clozapine and EGCG at the same time showed a significantly reduced differentiation. This effect might be due to activation of intracellular superoxide dismutase (Mn-SOD) which was supported by PCR analysis. Mn-SOD participates in the formation of reactive oxygen species. It elevates intracellular hydrogen peroxide levels which than inhibits preadipocyte differentiation. The weight-reducing abilities of EGCG has already been described in the literature and could be used for preventing excessive weight gain under clozapine therapy. Further research is therefore necessary. Future experiments of this work group will evaluate the proadipogenic effect of other neuroleptics and will further analyze their intracellular effects
