    Quantification of bioactive phenols, like stilbenes and flavonols (SaF), has been conducted to evaluate the nutraceutical potential of red wines. However, there is still a lack of full validated, fast and accessible liquid chromatography methods offering high selectivity and a simple procedure. We present here the use of a high-resolution mass spectrometer to evaluate the selectivity of a feasible and traditional liquid chromatography technique (HPLC? DAD) to analyze markers of aglycone SaF in red wines. The SaF compounds were tested: trans-resveratrol, trans-eviniferin, quercetin, myricetin, and kaempferol, as well as trans-cinnamic acid, one of their precursors. System suitability and validation tests were employed for the selected conditions (octylsilane column, methanol mobile phase, and gradient elution). The validation process ensured the HPLC?DAD method was selective, linear, sensitive, precise, accurate and robust. The method was then applied to red wine samples from the Campanha Gau´cha region, Southern Brazil. The real samples contained different SaF levels, showing that the method is applicable to routine use. Furthermore, this was the first SaF characterization of red wines from the Campanha Gau´cha, contributing to regional and product development.     Validation of a chromatographic method to routine analysis of transresveratrol and quercetin in red wines.

    The objective of this work was to validate a fast method with low-solvent use, for the analysis of trans-resveratrol and quercetin in red wines. The wines were prepared, using a classical method, from the grape (Vitis vinifera) cultivars Cabernet Franc, Cabernet Sauvignon, Malbec, Merlot, Petit Verdot, Pinot Noir, Ruby Cabernet, Syrah, Tannat, and Tempranillo. Samples were filtered and analyzed by high-pressure liquid chromatography (HPLC) with a diode array detector (DAD), at 306 and 371 nm, for trans-resveratrol and quercetin, respectively. An octylsilane column was used, and the mobile phase was composed by a gradient of methanol, water, and formic acid. The method was validated according to the following figures of merit: specificity, linearity, limit of detection, limit of quantification, precision, accuracy, and robustness. The proposed HPLC-DAD method may be established for the analysis of trans-resveratrol and quercetin in red wines. Validação de método cromatográfico para análises de rotina de trans-resveratrol e quercetina em vinhos tintos O objetivo deste trabalho foi validar um método rápido, com baixo consumo de solventes, para a análise de trans-resveratrol e quercetina em vinhos tintos. Os vinhos foram elaborados por método clássico, a partir das cultivares de uva (Vitis vinifera) Cabernet Franc, Cabernet Sauvignon, Malbec, Merlot, Petit Verdot, Pinot Noir, Ruby Cabernet, Syrah, Tannat e Tempranillo. As amostras foram filtradas e analisadas por cromatografia líquida de alta eficiência (CLAE) com detector de arranjo de diodos (DAD), em 306 e 371 nm, quanto ao trans-resveratrol e à quercetina, respectivamente. Utilizou-se uma coluna octilsilano, e a fase móvel foi composta por um gradiente de metanol, água e ácido fórmico. O método foi validado de acordo com as seguintes figuras de mérito: especificidade, linearidade, limite de detecção, limite de quantificação, precisão, exatidão e robustez. O método CLAE-DAD proposto pode ser implantado para análises de transresveratrol e quercetina em vinhos tintos.     Young varietal and experimental wines from Vitis vinifera - Petit Verdot,Tempranillo and Syrah - cultivated in a tropical semiarid region (Northeast Brazil) were evaluated with regard to their phenolic composition and chromatic characteristics during a storage period of 12-month. The wine was stored in bottles and evaluated using spectrophotometry and high efficiency liquid chromatography to determine color, total polyphenols, anthocyanins, flavanols and flavonols. The Petit Verdot had a less color variation when compared to wines from temperate climates. Although the wines followed the traditional pattem of decreasing polyphenol concentration during storage, the concentrations of these compounds remained higher than those reported in literature, such as for the anthocyanins in Petit Verdot wines and transresveratrolin Syrah, showing possible beneficial health effects even after storag

    Equilibrium Times for the Multicanonical Method

    This work measures the time to equilibrium for the multicanonical method on the 2D-Ising system by using a new criterion, proposed here, to find the time to equilibrium, teq, of any sampling procedure based on a Markov process. Our new procedure gives the same results that the usual one, based on the magnetization, for the canonical Metropolis sampling on a 2D-Ising model at several temperatures. For the multicanonical method we found a power-law relationship with the system size, L, of teq=0.27(15) L^2.80(13), and with the number of energy levels to explore, kE, of teq=0.7(13) kE^1.40(11), in perfect agreement with the result just above. In addition, some kind of critical slowing down was observed around the critical energy. Our new procedure is completely general, and can be applied to any sampling method based on a Markov process.Comment: 7 pages, 5 eps figures, to be published in Int. J. Mod. Phys.

    Analogue neural networks on correlated random graphs

    We consider a generalization of the Hopfield model, where the entries of patterns are Gaussian and diluted. We focus on the high-storage regime and we investigate analytically the topological properties of the emergent network, as well as the thermodynamic properties of the model. We find that, by properly tuning the dilution in the pattern entries, the network can recover different topological regimes characterized by peculiar scalings of the average coordination number with respect to the system size. The structure is also shown to exhibit a large degree of cliquishness, even when very sparse. Moreover, we obtain explicitly the replica symmetric free energy and the self-consistency equations for the overlaps (order parameters of the theory), which turn out to be classical weighted sums of 'sub-overlaps' defined on all possible sub-graphs. Finally, a study of criticality is performed through a small-overlap expansion of the self-consistencies and through a whole fluctuation theory developed for their rescaled correlations: Both approaches show that the net effect of dilution in pattern entries is to rescale the critical noise level at which ergodicity breaks down.Comment: 34 pages, 3 figure
