Influence of the Melissa officinalis
Melissa officinalis (MO, English: lemon balm, Lamiaceae), one of the oldest and still most popular aromatic medicinal plants, is used in phytomedicine for the prevention and treatment of nervous disturbances. The aim of our study was to assess the effect of subchronic (28-fold) administration of a 50% ethanol extract of MO leaves (200 mg/kg, p.o.) compared with rosmarinic acid (RA, 10 mg/kg, p.o.) and huperzine A (HU, 0.5 mg/kg, p.o.) on behavioral and cognitive responses in scopolamine-induced rats. The results were linked with acetylcholinesterase (AChE), butyrylcholinesterase (BuChE), and beta-secretase (BACE-1) mRNA levels and AChE and BuChE activities in the hippocampus and frontal cortex of rats. In our study, MO and HU, but not RA, showed an improvement in long-term memory. The results were in line with mRNA levels, since MO produced a decrease of AChE mRNA level by 52% in the cortex and caused a strong significant inhibition of BACE1 mRNA transcription (64% in the frontal cortex; 50% in the hippocampus). However, the extract produced only an insignificant inhibition of AChE activity in the frontal cortex. The mechanisms of MO action are probably more complicated, since its role as a modulator of beta-secretase activity should be taken into consideration
Oznaczanie zawartości flawan-3-oli w korzeniu Rhodiola kirilowii
According to a new classification, nearly 90 species belong to the Rhodiola family. However,
only three of them are most frequently used in alternative medicine. In addition to R. rosea
and R. quadrifida, R. Kirilowii can also be included in this group. This species was investigated
for flavan-3-ol content. Due to beneficial properties of this class of compounds
(free radical scavenging and reactive oxygen forms), the researchers decided to examine
5 substances in the root and in two extracts, alcoholic and aqueous ones. Depending on
the matrix analysed, the content of particular components varies. Furthermore, it is also
affected by extraction time as well as a particular extraction solvent employed. An ultra
performance liquid chromatograph connected to a tandem mass spectrometer (Waters)
was used for the assay. The application of this analytical method allows to detect very
small amounts of analytes.Zgodnie z nową klasyfikacją do rodziny Rhodiola należy ok. 90 gatunków. Jednak gatunkami
najczęściej wykorzystywanymi w medycynie niekonwencjonalnej są jedynie trzy z
nich. Oprócz R. rosea i R. quadrifida do tej grupy można zaliczyć R. Kirilowii. Gatunek ten
został poddany badaniom na zawartość flawan-3-oli. Ze względu na bardzo korzystne
właściwości tej klasy związków (zmiatania wolnych rodników i reaktywnych form tlenu),
postanowiono zbadać zawartość 5 substancji w korzeniu i dwóch wyciągach: wodnym i
alkoholowym. Zawartość poszczególnych składników różni się w zależności od analizowanej
matrycy. Wpływ na to ma czas ekstrakcji, jak również zastosowanie konkretnego
ekstrahentu. Do analizy wykorzystano ultrasprawny chromatograf cieczowy sprzężony z
tandemowym spektrometrem mas (Waters). Zastosowanie tej metody analitycznej pozwala
na wykrycie bardzo małych ilości analizowanych substancji
Zmienność fitochemiczna podbiału (Tussilago farfara L.) w Polsce
Coltsfoot leaves are a traditional raw material, rich in polysaccharides and phenolics. The
variability of the contents of these main compounds was determined, using plant material
originated from 22 natural populations of Tussilago farfara L. growing in various regions of Poland.
In the years 2008–2009, plants from each investigated population were collected in the
Garden of Medicinal Plants in Plewiska near Poznań (Poland). Coltsfoot leaves were harvested
in the middle of June and July of 2010, and then dried at room temperature. In these raw
material we quantified swelling index (describing mucilage content) and spectrophotometrically:
the amounts of total polyphenols, polyphenols unadsorbed on hide powder (non-tannin
phenolics) and tannins (expressed as pyrogallol equivalent) as well as the sum of hydroxycinnamic
acid derivatives (expressed as rosmarinic acid) and flavonoids (expressed as quercetin).
The demonstrated results show the relatively high and balanced contents of the basic active
compounds, especially flavonoids (0.7–1.3%) and polysaccharides (swelling index: 8.0–14.5).
In addition, it was found that flavonoids and mucilage in coltsfoot leaves fluctuate in only
a small range (V=11–13%), regardless of overshadow and the harvest time of raw material.Liście podbiału stanowią tradycyjny surowiec zielarski, bogaty w polisacharydy i związki
fenolowe. W niniejszych badaniach określono poziom zróżnicowania zawartości wspomnianych
związków czynnych. Jako materiał roślinny wykorzystano 22 naturalne populacje
Tussilago farfara L., pochodzące z różnych regionów Polski. W latach 2008–2009 pobrano
rośliny z każdej badanej populacji i założono kolekcję pochodzeniową w Ogrodzie
Roślin Leczniczych w Plewiskach koło Poznania. Liście podbiału do analiz fitochemicznych
zbierano w połowie czerwca i lipca 2010 r., a następnie suszono w temperaturze pokojowej.
W pozyskanym surowcu zielarskim oznaczono wskaźnik pęcznienia (określający
zawartość śluzów) oraz spektrofotometrycznie: poziom polifenoli ogółem, polifenoli nie
wiążących się z proszkiem skórzanym i garbników (w przeliczeniu na pirogalol), a także
sumę pochodnych kwasu hydroksycynamonowego (w przeliczeniu na kwas rozmarynowy)
i flawonoidów (w przeliczeniu na kwercetynę). Prezentowane wyniki badań wskazują
na relatywnie wysoką i wyrównaną zawartość głównych związków czynnych, szczególnie
flawonoidów (0,7–1,3%) i polisacharydów (wskaźnik pęcznienia: 8,0–14,5). Stwierdzono,
iż poziom wspomnianych grup związków zmienia się w małym zakresie (V=11–13%) i nie
zależy od ocienienia roślin i terminu zbioru surowca
Zmienność biometryczna i fitochemiczna różeńca górskiego (Rhodiola rosea L.) w uprawie
Roseroot (Rhodiola rosea L.) is an adaptogenic plant, widely used in the traditional medicine
of Scandinavia, Russia, China and Mongolia. The aim of the study was to determine
the biometric and phytochemical co-variability of this species under field cultivation in
western Poland (Plewiska near Poznań). The plant material originated from four-year-old
cultivation established twice by rhizome division in autumn 2007 and 2008. In the study,
46 individuals (23 plants in 2011 and in 2012) were used. The biometric analysis included
measurements of the basic characteristics of plants related to the size (luxuriance)
of clumps, shoots, leaves and rhizomes with roots (raw material). The amounts of total
polyphenols (expressed as gallic acid), tannins (expressed as pyrogallol) and flavonoids
(expressed as quercetin) were determined spectrophotometrically. The obtained results
showed high variation of Rh. rosea, especially in the level of flavonoids (0.01–0.20% DM)
and in the weight of raw material (113–1156 g FM/plant). There were observed correlations
between the phytochemical (flavonoid and total phenolic content) and biometric
(water content, leaf number, shoot and clump size) features.Różeniec górski (Rhodiola rosea L.) jest rośliną o działaniu adaptogennym, szeroko stosowaną
w tradycyjnej medycynie Skandynawii, Rosji, Chin i Mongolii. Celem prezentowanych
badań było opisanie współzmienności biometrycznych i fitochemicznych cech tego
gatunku, w warunkach uprawy polowej, prowadzonej w zachodniej Polsce (Plewiska koło
Poznania). Materiał do analiz pochodził z czteroletnich upraw, założonych jesienią 2007
i 2008 r., przez podział kłączy macierzystych roślin. W latach 2011 i 2012 zebrano po 23
osobniki omawianego gatunku. Biometria obejmowała pomiary podstawowych cech roślin,
opisujących wielkość (bujność) tworzonych kęp, pędów, liści oraz kłączy z korzeniami
(surowca zielarskiego). Badania fitochemiczne wykonano metodą spektrofotometryczną,
oznaczając zawartość polifenoli ogółem (w przeliczeniu na kwas galusowy), garbników
(w przeliczeniu na pirogalol) i flawonoidów (w przeliczeniu na kwercetynę). Otrzymane
wyniki wskazują na dużą zmienność Rh. rosea, szczególnie w przypadku poziomu flawonoidów
(0,01–0,20% s.m.) i masy surowca (113–1156 g św.m./roślina). Stwierdzono również
istotne statystycznie korelacje między cechami fitochemicznymi (zawartość flawonoidów
i fenoli ogółem) a biometrycznymi (zawartość wody, liczba liści, wielkość pędów i kęp)
Phenolic acids improve the antioxidant activity of ceruloplasmin isolated from plasma of healthy volunteers and atherosclerotic patients
The aim of the present study was to estimate the ability of individual phenolic acids to
eliminate Fe(II) ions from the solution. Moreover, the influence of phenolic acids on the ferroxidase
activity of ceruloplasmin (Cp) isolated from blood plasma of healthy volunteers (CpC) and
patients with atherosclerosis obliterans (CpAO) was established in vitro. Phenolic acids demonstrated
a ferroxidase-like activity, i.e. the ability to eliminate Fe(II) ions, within the studied
concentration range of 2.0-17.0 mol 10-5 dm-3, in the following order of decreasing effectiveness:
caffeic acid (CA)>ellagic acid (EA)>chlorogenic acid (ChA)>ferulic acid (FA) ≈ p-coumaric acid
(PcA) = sinapic acid (SA). The study showed that the ability of phenolic acids to eliminate Fe(II) ions by oxidation or chelation was related to the structure of the former, to the presence of orto
-OH groups, especially. Furthermore, the effect of the molar ratio of phenolic acid to Fe(II) ion
was observed. EA and ChA, both containing two orto-OH groups and the highest number of –OH
groups (4 and 5, respectively), demonstrated the greatest ability to eliminate Fe(II) ions, especially
at the Fe(II) to phenolic acid molar ratio of 6:1. Phenolic acids added to samples with a
constant amount of Cp caused decrease in the concentration of Fe(II) ions. Therefore, it may be
assumed that the addition of phenolic acids to CpC and CpAO, even in low concentrations, caused
a significant decrease in the concentration of Fe(II) ions
Plon i zawartość związków fenolowych w kwiatostanach kocanek piaskowych Helichrysum arenarium (l.) Moench zbieranych ze stanu naturalnego
Introduction: The inflorescence of Helichrysum arenarium (L.) Moench shows anti-inflammatory,
antioxidant, detoxifying properties and is traditionally used in liver and biliary
tract diseases. Because of its difficult and expensive cultivation, the plant raw material is
mainly harvested from natural sites. Objective: The research aimed to determine the local
variation in yield and content of flavonoids and phenolic acids in the yellow everlasting
inflorescence against the background of the layer structure of vegetation as the rate of
plant succession. Methods: The plant raw material was collected from 30 plots of 1 m2,
established for three separate populations developing on sandy fallows near Zielona Góra
(western Poland). For each study area, percentage cover of the moss-lichen and herb layers,
the height, cover and yield of H. arenarium as well as the height and cover of other
herbaceous plants were determined. Total contents of flavonoids (expressed as quercetin)
and phenolic acids (calculated as caffeic acid) were measured spectrophotometrically, according
to Polish Pharmacopoeia. Results: Everlastings reached a cover of up to 70% and
the maximum air-dry matter yield of 46.42 g/m2. The height, coverage and yield of H.
arenarium were correlated with the parameters describing the herb layer. The content of
flavonoids ranged from 0.56 to 0.99%, while that of phenolic acids from 0.82 to 1.80% DM.
Conclusions: Yellow everlasting is an important species of early fallows on poor sandy soils and these habitats constitute a rich natural source of herbal raw material. Inflorescences
harvested from natural sites are distinguished by a high and similar content of polyphenols
and usually meet the requirements of Polish Pharmacopoeia.Wstęp: Kwiatostany Helichrysum arenarium (L.) Moench są tradycyjnie stosowane w chorobach
wątroby i dróg żółciowych, wykazując właściwości przeciwzapalne, antyoksydacyjne
oraz odtruwające. Ze względu na trudności w uprawie tego gatunku i wysokie koszty
prowadzenia plantacji, surowiec zielarski wciąż jest pozyskiwany głównie ze stanu naturalnego.
Cel: W prezentowanych badaniach określono lokalną zmienność plonowania oraz
zawartości flawonoidów i fenolokwasów ogółem w kwiatostanach kocanek piaskowych
na tle struktury warstwowej roślinności jako wskaźnika zaawansowania procesu sukcesji.
Metody: Surowiec zielarski zebrano z 30 poletek (1 m2), wyznaczonych w obrębie trzech
populacji kocanek występujących na piaszczystych odłogach w okolicach Zielonej Góry.
Na każdej powierzchni badawczej opisano strukturę warstwową roślinności: procentowe
pokrycie warstwy porostowo-mszystej i zielnej, wysokość, pokrycie i plon H. arenarium
oraz przeciętną wysokość i pokrycie pozostałych gatunków zielnych. Całkowitą zawartość
flawonoidów (w przeliczeniu na kwercetynę) i fenolokwasów (w przeliczeniu na kwas
kawowy) oznaczono metodą spektrofotometryczną według Farmakopei Polskiej. Wyniki:
Kocanki uzyskiwały pokrycie do 70%, a maksymalny plon powietrznie suchej masy kwiatostanów
wynosił 46,42 g/m2. Wysokość, pokrycie i plon H. arenarium były skorelowane
z parametrami opisującymi warstwę zielną. Poziom flawonoidów w surowcu mieścił się
w granicach od 0,56 do 0,99% suchej masy (s.m.), natomiast kwasów fenolowych: od 0,82
do 1,80% s.m. Wnioski: Kocanki stanowią ważny element roślinności wczesnych odłogów
powstałych na ubogich, piaszczystych glebach. Dlatego też, wspomniane siedliska mogą
być zasobnym źródłem surowca zielarskiego. Kwiatostany zbierane ze stanu naturalnego
charakteryzują się wysoką i wyrównaną zawartością związków polifenolowych i zwykle
spełniają wymagania Farmakopei Polskiej
Wplyw ekstraktu z pestek porzeczki czarnej [Ribus nigrum] na skutecznosc ludzkiej ceruloplazminy w eliminowaniu Fe[II]
Ions of irons, especially ferrous ions may be harmful for living organisms, because
they generate reactive oxygen species like O2
•- or •OH. Probability of the risk rises especially
in pathological conditions, in which high level of iron is observed. For this reason
scientists try to establish new methods that can support organism in eliminating reactive
ferrous ions.
Nowadays, attention focuses on substances present in plants, especially polyphenols,
whose administration prevents oxidative damages in iron overloading.
This new approach requires some research on behavior of plant-derived compounds
in human organism, within a system containing other biomolecules involved in iron metabolism.
The aim of this study has been to investigate the influence of black currant (Ribes
nigrum) seed extract, a source of polyphenols, on the activity of ceruloplasmin, an enzyme
participating in Fe(II) elimination from blood plasma in human organism. Depletion of
Fe(II) caused by ceruloplasmin isolated from healthy blood donors was compared to its decrease
in a system containing both ceruloplasmin and the extract. The results have shown
that addition of a particular amount of the extract elevates the effectiveness of ceruloplasmin
in eliminating Fe(II) from the sample but only under physiological condition (pH 7.4;
T 37°C). In a weak acidic solution, addition of the extract does not lead to any change in

stanach patologicznych, którym towarzyszy przeładowanie organizmu żelazem, ryzyko
pojawienia się wolnych jonów Fe(II) inicjujących wiele procesów wolnorodnikowych jest
bardzo prawdopodobne. W tym celu dąży się do opracowania terapii, które w chorobach
żelazozależnych wspomogą naturalne mechanizmy usuwania reaktywnych jonów żelaza
z organizmu. Ostatnio dużą uwagę poświęca się aktywnym substancjom obecnym w roślinach,
szczególnie związkom polifenolowym, ze względu na ich silne właściwości chelatujące. Ten nowy kierunek badań wymaga jednak wnikliwej analizy, która wyjaśni mechanizm
współdziałania substancji roślinnych z innymi biomolekułami zaangażowanymi w prawidłowy
metabolizm żelaza w organizmie oraz rozstrzygnie o bezpieczeństwie ich stosowania.
Celem badań było ustalenie wpływu ekstraktu z pestek porzeczki czarnej (Ribus nigrum),
bogatego w związki polifenolowe, na aktywność ludzkiej ceruloplazminy, enzymu uczestniczącego w usuwaniu Fe(II) w organizmie ludzkim. Ubytek Fe(II) obserwowany w obecności
ceruloplazminy porównywano z jego ubytkiem w mieszaninie ceruloplazmina-ekstrakt.
Stwierdzono, że dodatek ekstraktu podnosi skuteczność ceruloplazminy w eliminowaniu
Fe(II) ze środowiska. Istnieje więc przypuszczenie, że związki pochodzenia roślinnego takie
jak polifenole mogą wspomagać naturalne mechanizmy eliminowania Fe(II) w organizmie
Antioxidant Potential of Hemp and Flax Fibers Depending on Their Chemical Composition
Flax and hemp fibers are known as textile raw materials with pro-health properties. This paper presents results of research aimed at investigating the antioxidant activity of the fibers in order to explain a mechanism for the favorable influence of textiles made of these fibers when in contact with human skin. The study presents a new approach in evaluation of textile fibers in terms of their inherent pro-health properties. Antioxidant properties of flax and hemp fibers were tested with the use of FRAP and DPPH methods. The content of phenolic acids: syringic, sinapinic, p-coumaric and ferulic acid was determined with the use of HPLC. The results proved that the antioxidant activity of the tested fibers depends on a type and variety of fibrous plants, a method of fiber extraction and subsequent stages of preliminary processing of the fibers. Both decorticated flax and hemp fibers showed higher antioxidant activity in comparison to the fibers obtained with other extraction methods, while decorticated flax of different varieties presented the highest value of the FRAP. Porównanie zawartości proantocyjanidyn w korzeniach Rhodiola kirilowii i Rhodiola rosea z wykorzystaniem metody UPLC-MS/MS
The purpose of presented study was the comparison of flavan-3-ol content in the roots
of Rhodiola kirilowii and Rhodiola rosea with use of UPLC-MS/MS method. Two kinds of extract
were prepared: aqueous extract and 50% v/v ethanol extract. The worked out UPLC
MS/MS method allowed to determine the content of five flavan-3-ols: (+)-catechin, (-)-epicatechin,
(-)-epigallocatechin, (-)-epicatechin gallate (ECG) and (-)-epigallocatechin gallate
(EGCG). The obtained results shown that the content of measured catechins was higher in
R. kirilowii roots than in R. rosea. Both Rhodiola roots contain EGCG as a main proanthocyanidin
compound – the content in R. kirilowii roots is about 0.14%. Our results indicate
that application of ultra performance liquid chromatograph connected to a tandem mass
spectrometer (UPLC MS/MS method) allows to determine the proanthocyanidins content
in tested samples with satisfactory precision and can be used in the Rhodiola sp. phytochemical
investigations.Celem przedstawionych badań było porównanie za pomocą opracowanej metodyki wykorzystującej
ultrasprawny chromatograf cieczowy sprzężony z tandemowym spektrometrem
mas (Waters) zawartości flawan-3-oli w korzeniach Rhodiola kirilowii i Rhodiola rosea.
Badano wodne i alkoholowo-wodne (50% EtOH) wyciągi z korzeni. Opracowana metoda
UPLC MS/MS pozwoliła na określenie stężeń pięciu flawan-3-oli: (+)-katechiny, (-)-epikatechiny,
(-)-epigalokatechiny, galusanu (-)-epikatechiny (ECG) oraz galusanu (-)-epigalokatechiny
(EGCG). Otrzymane wyniki wskazują, że zawartość tych katechin jest wyższa w korzeniach
R. kirilowii niż w korzeniach R. rosea. Oba surowce zawierają galusan (-)-epigallokatechiny
jako główną proantocyjanidynę – jej zawartość w korzeniach R. kirilowii wynosi
ok. 0,14%. Uzyskane przez nas wyniki wskazują, że metoda analityczna z wykorzystaniem
ultrasprawnego chromatografu cieczowego sprzężonego z tandemowym spektrometrem
mas pozwala z zadawalającą precyzją oznaczyć zawartości proantocyjanidyn w analizowanych
próbkach i może być stosowana w badaniach rodzaju Rhodiola
Porównanie zawartości fenyloetanoidów w korzeniach Rhodiola kirilowii i Rhodiola rosea za pomocą metody UPLC-MS/MS
A concentration of two phenylethanoids in the roots of two species: Rhodiola kirilowii and
rosea were compared, aqueous and hydroalcoholic extracts from those plants were also
analyzed. In order to determine the content of p-tyrosol and salidroside, the ultra performance
liquid chromatography connected with a tandem mass spectrometry (UPLC-ESI
MS/MS, Waters) was used. The obtained results shown that content of measured phenylethanoids
depends on Rhodiola species. Roots of R. kirilowii contain more p-tyrosol,
while R. rosea roots are reacher in salidroside. Our results indicate that the application of
UPLC MS/MS method allows to determine the phenylethanoids content in tested samples
with satisfactory precision.Przedmiotem badań było zwalidowanie metody analitycznej oznaczenia zawartości fenyloetanoidów.
Do detekcji p-tyrozolu i salidrozydu w analizowanych dwóch gatunkach
różeńca wykorzystano wysokosprawny chromatograf cieczowy sprzężony z tandemowym
spektrometrem mas (UPLC-MS/MS). Analizie poddano Rhodiola kirilowii oraz Rhodiola rosea
uzyskane z hodowli gruntowej w 2009 r. w Instytucie Włókien Naturalnych i Roślin Zielarskich
w Poznaniu. Dodatkowo z surowca przygotowano dwa wyciągi suche: wyciąg wodny
