    A reinforced carbon-carbon thermal protection system is used on the space shuttle orbiter vehicle's nose cap and wing leading edge regions where temperatures reach 1538 C (2800 F). To verify the analyses used to certify reinforced carbon-carbon for the first flight and operational missions, a multi-environment incremental test program was developed and implemented through the combined efforts of Rockwell International and the National Aeronautics and Space Administration. Three separate facilities at the Johnson Space Center were used to subject full-scale nose cap and wing leading edge test articles to simulated critical launch, on-orbit, and atmospheric entry environments

    Experimental Data on Stagnation-Point Gas Injection Cooling on a Hemisphere-Cone in a Hypersonic Arc Tunnel

    Exploratory tests were made to determine the reduction of heat transfer resulting from ejection of gases at the stagnation point on a hemisphere-cone at a nominal Mach number of about 9 and at stagnation enthalpies up to 1600 BTU per pound. Helium, nitrogen, and argon gases were used as coolants. The ratio of mass flow of coolant to the mass flow of air swept out by the model projected area was less than 0.20 in all cases. The experimental heat-transfer data were normalized by the calculated stagnation-point values based on test conditions in the tunnel and are presented as a function of the surface distance from the stagnation point. The variation of shock standoff distances with mass flow and volumetric flow of the gaseous ejectants is presented along with photographs of the luminous flow field of the model during tests

    Brucella sero-prevalence and modifiable risk factors among predisposed cattle keepers and consumers of un-pasteurized milk in Mbarara and Kampala districts, Uganda

    Background: Brucellosis is an important zoonotic disease in developing countries yet it is often not recognized, goes unreported and does not attract public health action by these governments including Uganda.Objective: To estimate the sero-prevalence and assess modifiable risk factors associated with Brucella seropositivity in cattle keepers and consumers of unpasteurized milk in Uganda.Methods: One group comprised of 161 individuals randomly selected from households living on farms that had Brucella sero-positive cattle and/or goats in Mbarara District from an earlier survey; the second group comprised of 168 randomly selected individuals attending an HIV voluntary counseling and testing clinic in Kampala District. Sera samples were tested using Rapid Plate Agglutination Test, Standard Tube Agglutination Test and cELISA.Results: The sero-prevalence of brucellosis among exposed cattle keepers in Mbarara and consumers of unpasteurised milk in Kampala Districts was 5.8% (95%CI: 3.3%, 8.3%) and 9% (95%CI: 13.3%, 4.7%), respectively. Consumption of unboiled milk was significantly (p=0.004) associated with seropositivity in Mbarara District. There was no association between sero-positivity with age, sex and awareness of human brucellosis.Conclusion: Human brucellosis is prevalent among livestock rearing communities and consumers of unpasteurised milk.     Determinants of flow-mediated outward remodeling in female rodents: respective roles of age, estrogens, and timing

    OBJECTIVE: Flow (shear stress)-mediated outward remodeling (FMR) of resistance arteries is a key adaptive process allowing collateral growth after arterial occlusion but declining with age. 17-beta-estradiol (E2) has a key role in this process through activation of estrogen receptor alpha (ERalpha). Thus, we investigated the impact of age and timing for estrogen efficacy on FMR. APPROACH AND RESULTS: Female rats, 3 to 18 months old, were submitted to surgery to increase blood flow locally in 1 mesenteric artery in vivo. High-flow and normal-flow arteries were collected 2 weeks later for in vitro analysis. Diameter increased by 27% in high-flow arteries compared with normal-flow arteries in 3-month-old rats. The amplitude of remodeling declined with age (12% in 18-month-old rats) in parallel with E2 blood level and E2 substitution failed restoring remodeling in 18-month-old rats. Ovariectomy of 3-, 9-, and 12-month-old rats abolished FMR, which was restored by immediate E2 replacement. Nevertheless, this effect of E2 was absent 9 months after ovariectomy. In this latter group, ERalpha and endothelial nitric oxide synthase expression were reduced by half compared with age-matched rats recently ovariectomized. FMR did not occur in ERalpha(-/-) mice, whereas it was decreased by 50% in ERalpha(+/-) mice, emphasizing the importance of gene dosage in high-flow remodeling. CONCLUSIONS: E2 deprivation, rather than age, leads to decline in FMR, which can be prevented by early exogenous E2. However, delayed E2 replacement was ineffective on FMR, underlining the importance of timing of this estrogen action

    Whole-genome analysis of histone H3 lysine 27 trimethylation in Arabidopsis

    Trimethylation of histone H3 lysine 27 (H3K27me3) plays critical roles in regulating animal development, and in several cases, H3K27me3 is also required for the proper expression of developmentally important genes in plants. However, the extent to which H3K27me3 regulates plant genes on a genome-wide scale remains unknown. In addition, it is not clear whether the establishment and spreading of H3K27me3 occur through the same mechanisms in plants and animals. We identified regions containing H3K27me3 in the genome of the flowering plant Arabidopsis thaliana using a high-density whole-genome tiling microarray. The results suggest that H3K27me3 is a major silencing mechanism in plants that regulates an unexpectedly large number of genes in Arabidopsis (~4,400), and that the maintenance of H3K27me3 is largely independent of other epigenetic pathways, such as DNA methylation or RNA interference. Unlike in animals, where H3K27m3 occupies large genomic regions, in Arabidopsis, we found that H3K27m3 domains were largely restricted to the transcribed regions of single genes. Furthermore, unlike in animals systems, H3K27m3 domains were not preferentially associated with low-nucleosome density regions. The results suggest that different mechanisms may underlie the establishment and spreading of H3K27me3 in plants and animals
