    UV photon-counting CCD detectors that enable the next generation of UV spectroscopy missions: AR coatings that can achieve 80-90% QE

    We describe recent progress in the development of anti-reflection coatings for use at UV wavelengths on CCDs and other Si-based detectors. We have previously demonstrated a set of coatings which are able to achieve greater than 50% QE in 4 bands from 130nm to greater than 300nm. We now present new refinements of these AR-coatings which will improve performance in a narrower bandpass by 50% over previous work. Successful test films have been made to optimize transmission at 190nm, reaching 80% potential transmission

    A robust Pax7EGFP mouse that enables the visualization of dynamic behaviors of muscle stem cells.

    BACKGROUND: Pax7 is a transcription factor involved in the specification and maintenance of muscle stem cells (MuSCs). Upon injury, MuSCs leave their quiescent state, downregulate Pax7 and differentiate, contributing to skeletal muscle regeneration. In the majority of regeneration studies, MuSCs are isolated by fluorescence-activated sorting (FACS), based on cell surface markers. It is known that MuSCs are a heterogeneous population and only a small percentage of isolated cells are true stem cells that are able to self-renew. A strong Pax7 reporter line would be valuable to study the in vivo behavior of Pax7-expressing stem cells. METHODS: We generated and characterized the muscle properties of a new transgenic Pax7EGFP mouse. Utilizing traditional immunofluorescence assays, we analyzed whole embryos and muscle sections by fluorescence microscopy, in addition to whole skeletal muscles by 2-photon microscopy, to detect the specificity of EGFP expression. Skeletal muscles from Pax7EGFP mice were also evaluated in steady state and under injury conditions. Finally, MuSCs-derived from Pax7EGFP and control mice were sorted and analyzed by FACS and their myogenic activity was comparatively examined. RESULTS: Our studies provide a new Pax7 reporter line with robust EGFP expression, detectable by both flow cytometry and fluorescence microscopy. Pax7EGFP-derived MuSCs have identical properties to that of wild-type MuSCs, both in vitro and in vivo, excluding any positional effect due to the transgene insertion. Furthermore, we demonstrated high specificity of EGFP to label MuSCs in a temporal manner that recapitulates the reported Pax7 expression pattern. Interestingly, immunofluorescence analysis showed that the robust expression of EGFP marks cells in the satellite cell position of adult muscles in fixed and live tissues. CONCLUSIONS: This mouse could be an invaluable tool for the study of a variety of questions related to MuSC biology, including but not limited to population heterogeneity, polarity, aging, regeneration, and motility, either by itself or in combination with mice harboring additional genetic alterations

    LAT1 Protein Content Increases Following 12 Weeks of Resistance Exercise Training in Human Skeletal Muscle

    Introduction: Amino acid transporters are essential for cellular amino acid transport and promoting protein synthesis. While previous literature has demonstrated the association of amino acid transporters and protein synthesis following acute resistance exercise and amino acid supplementation, the chronic effect of resistance exercise and supplementation on amino acid transporters is unknown. The purpose herein was to determine if amino acid transporters and amino acid metabolic enzymes were related to skeletal muscle hypertrophy following resistance exercise training with different nutritional supplementation strategies. Methods: 43 college-aged males were separated into a maltodextrin placebo (PLA, n = 12), leucine (LEU, n = 14), or whey protein concentrate (WPC, n = 17) group and underwent 12 weeks of total-body resistance exercise training. Each group\u27s supplement was standardized for total energy and fat, and LEU and WPC supplements were standardized for total leucine (6 g/d). Skeletal muscle biopsies were obtained prior to training and ~72 h following each subject\u27s last training session. Results: All groups increased type I and II fiber cross-sectional area (fCSA) following training (p \u3c 0.050). LAT1 protein increased following training (p \u3c 0.001) and increased more in PLA than LEU and WPC (p \u3c 0.050). BCKDHα protein increased and ATF4 protein decreased following training (p \u3c 0.001). Immunohistochemistry indicated total LAT1/fiber, but not membrane LAT1/fiber, increased with training (p = 0.003). Utilizing all groups, the change in ATF4 protein, but no other marker, trended to correlate with the change in fCSA (r = 0.314; p = 0.055); however, when regression analysis was used to delineate groups, the change in ATF4 protein best predicted the change in fCSA only in LEU (r2 = 0.322; p = 0.043). In C2C12 myoblasts, LAT1 protein overexpression caused a paradoxical decrease in protein synthesis levels (p = 0.002) and decrease in BCKDHα protein (p = 0.001). Conclusions: Amino acid transporters and metabolic enzymes are affected by resistance exercise training, but do not appear to dictate muscle fiber hypertrophy. In fact, overexpression of LAT1 in vitro decreased protein synthesis

    "Even if the test result is negative, they should be able to tell us what is wrong with us": a qualitative study of patient expectations of rapid diagnostic tests for malaria.

    BACKGROUND: The debate on rapid diagnostic tests (RDTs) for malaria has begun to shift from whether RDTs should be used, to how and under what circumstances their use can be optimized. This has increased the need for a better understanding of the complexities surrounding the role of RDTs in appropriate treatment of fever. Studies have focused on clinician practices, but few have sought to understand patient perspectives, beyond notions of acceptability. METHODS: This qualitative study aimed to explore patient and caregiver perceptions and experiences of RDTs following a trial to assess the introduction of the tests into routine clinical care at four health facilities in one district in Ghana. Six focus group discussions and one in-depth interview were carried out with those who had received an RDT with a negative test result. RESULTS: Patients had high expectations of RDTs. They welcomed the tests as aiding clinical diagnoses and as tools that could communicate their problem better than they could, verbally. However, respondents also believed the tests could identify any cause of illness, beyond malaria. Experiences of patients suggested that RDTs were adopted into an existing system where patients are both physically and intellectually removed from diagnostic processes and where clinicians retain authority that supersedes tests and their results. In this situation, patients did not feel able to articulate a demand for test-driven diagnosis. CONCLUSIONS: Improvements in communication between the health worker and patient, particularly to explain the capabilities of the test and management of RDT negative cases, may both manage patient expectations and promote patient demand for test-driven diagnoses

    A VLBI receiving system for the South Pole Telescope

    The Event Horizon Telescope (EHT) is a very-long-baseline interferometry (VLBI) experiment that aims to observe supermassive black holes with an angular resolution that is comparable to the event horizon scale. The South Pole occupies an important position in the array, greatly increasing its north-south extent and therefore its resolution. The South Pole Telescope (SPT) is a 10-meter diameter, millimeter-wavelength telescope equipped for bolometric observations of the cosmic microwave background. To enable VLBI observations with the SPT we have constructed a coherent signal chain suitable for the South Pole environment. The dual-frequency receiver incorporates state-of-the-art SIS mixers and is installed in the SPT receiver cabin. The VLBI signal chain also includes a recording system and reference frequency generator tied to a hydrogen maser.     First Results from COPSS: The CO Power Spectrum Survey

    We present constraints on the abundance of carbon monoxide in the early universe from the CO Power Spectrum Survey. We utilize a data set collected between 2005 and 2008 using the Sunyaev–Zel'dovich Array (SZA), which was previously used to measure arcminute-scale fluctuations of the cosmic microwave background. This data set features observations of 44 fields, covering an effective area of 1.7 square degrees, over a frequency range of 27–35 GHz. Using the technique of intensity mapping, we are able to probe the CO(1–0) transition, with sensitivity to spatial modes between k = 0.5–2 h Mpc^(−1) over a range in redshift of z = 2.3–3.3, spanning a comoving volume of 3.6 × 10^6 h^(−3) Mpc^3. We demonstrate our ability to mitigate foregrounds, and present estimates of the impact of continuum sources on our measurement. We constrain the CO power spectrum to P_(CO) < 2.6 × 10^4 μK^2 (h^(−1) Mpc)^3, or Δ^2_(CO)(k = 1 h Mpc^(−1)) < 1.3 × 10^3 μK^2, at 95% confidence. This limit resides near optimistic predictions for the CO power spectrum. Under the assumption that CO emission is proportional to halo mass during bursts of active star formation, this corresponds to a limit on the ratio of CO(1–0) luminosity to host halo mass of A_(CO) < 1.2 × 10^(−5) L⊙_ M_⊙^(−1). Further assuming a Milky Way-like conversion factor between CO luminosity and molecular gas mass (α_(CO) = 4.3 M_⊙ (K km s^(−1) pc^(−2))^(−1)), we constrain the global density of molecular gas to ρ_(z~3) (M_H_2) ⩽ 2.8 x 10^8 M_☉ Mpc^(-3)

    Observations of High-Redshift X-Ray Selected Clusters with the Sunyaev-Zel'dovich Array

    We report measurements of the Sunyaev-Zel'dovich (SZ) effect in three high-redshift (0.89 ≤ z ≤ 1.03), X-ray selected galaxy clusters. The observations were obtained at 30 GHz during the commissioning period of a new, eight-element interferometer—the Sunyaev-Zel'dovich Array (SZA)—built for dedicated SZ effect observations. The SZA observations are sensitive to angular scales larger than those subtended by the virial radii of the clusters. Assuming isothermality and hydrostatic equilibrium for the intracluster medium and gas-mass fractions consistent with those for clusters at moderate redshift, we calculate electron temperatures, gas masses, and total cluster masses from the SZ data. The SZ-derived masses, integrated approximately to the virial radii, are 1.9^(+0.5)_(-0.4) × 10^(14) M_☉ for Cl J1415.1+3612, 3.4^(+0.6)_(-0.5) × 10^(14) M_☉ for Cl J1429.0+4241, and 7.2^(+1.3)_(-0.9) × 10^(14) M_☉ for Cl J1226.9+3332. The SZ-derived quantities are in good agreement with the cluster properties derived from X-ray measurements

    Sunyaev Zel'dovich Effect Observations of Strong Lensing Galaxy Clusters: Probing the Over-Concentration Problem

    We have measured the Sunyaev Zel'dovich (SZ) effect for a sample of ten strong lensing selected galaxy clusters using the Sunyaev Zel'dovich Array (SZA). The SZA is sensitive to structures on spatial scales of a few arcminutes, while the strong lensing mass modeling constrains the mass at small scales (typically < 30"). Combining the two provides information about the projected concentrations of the strong lensing clusters. The Einstein radii we measure are twice as large as expected given the masses inferred from SZ scaling relations. A Monte Carlo simulation indicates that a sample randomly drawn from the expected distribution would have a larger median Einstein radius than the observed clusters about 3% of the time. The implied overconcentration has been noted in previous studies with smaller samples of lensing clusters. It persists for this sample, with the caveat that this could result from a systematic effect such as if the gas fractions of the strong lensing clusters are substantially below what is expected.Comment: submitte

    A Minimal Model of Metabolism Based Chemotaxis

    Since the pioneering work by Julius Adler in the 1960's, bacterial chemotaxis has been predominantly studied as metabolism-independent. All available simulation models of bacterial chemotaxis endorse this assumption. Recent studies have shown, however, that many metabolism-dependent chemotactic patterns occur in bacteria. We hereby present the simplest artificial protocell model capable of performing metabolism-based chemotaxis. The model serves as a proof of concept to show how even the simplest metabolism can sustain chemotactic patterns of varying sophistication. It also reproduces a set of phenomena that have recently attracted attention on bacterial chemotaxis and provides insights about alternative mechanisms that could instantiate them. We conclude that relaxing the metabolism-independent assumption provides important theoretical advances, forces us to rethink some established pre-conceptions and may help us better understand unexplored and poorly understood aspects of bacterial chemotaxis
