    Visualization of microbe-dietary remnant interactions in digesta from pigs, by fluorescence in situ hybridization and staining methods; effects of a dietary arabinoxylan-rich wheat fraction

    This study investigated how the addition of a specific hydrocolloid, arabinoxylan (AX), to the pig diets containing red meat, affected the gut microbiota in terms of changes to the bacterial community dynamics. Fluorescence in situ hybridization (FISH) was used to enumerate both broad groups and particular bacterial species, and showed systematic differences in pigs fed AX. In addition, the role of bacteria in the fermentation of dietary substrates was studied by visualizing direct associations between the bacteria and substrate particles. To achieve this, novel methods of combining histological staining or enzymatic labelling with FISH protocols were established. In this way, undigested and/or unfermented remnants of meat, endogenous mucin, and several plant cell wall hydrocolloids (cellulose, lignin and arabinoxylan) were distinguished, together with large intestine microbiota under brightfield and/or confocal laser scanning microscopy (CLSM). It was evident that no apparent direct associations occurred between specific classes or groups of bacteria and meat, mucin, lignin or AX remnants. In contrast, bacteria belonging to Clostridium clusters XIVa and XIVb formed a strong (P < 0.01) direct association with cellulose remnants in the pig digesta in the caecum

    Radio Detection of High Energy Particles: Coherence Versus Multiple Scales

    Radio Cherenkov emission underlines detection of high energy particles via a signal growing like the particle-energy-squared. Cosmic ray-induced electromagnetic showers are a primary application. While many studies have treated the phenomenon approximately, none have attempted to incorporate all the physical scales involved in problems with time- or spatially- evolving charges. We find it is possible to decompose the calculated fields into the product of a form factor, characterizing a moving charge distribution, multiplying a general integral which depends on the charge evolution. In circumstances of interest for cosmic ray physics, the resulting expressions can be evaluated explicitely in terms of a few parameters obtainable from shower codes. The classic issues of Frauhofer and Fresnel zones play a crucial role in the coherence.Comment: 25 pages, 10 figure

    Measurement of the cosmic ray spectrum above 4×10184{\times}10^{18} eV using inclined events detected with the Pierre Auger Observatory

    A measurement of the cosmic-ray spectrum for energies exceeding 4×10184{\times}10^{18} eV is presented, which is based on the analysis of showers with zenith angles greater than 6060^{\circ} detected with the Pierre Auger Observatory between 1 January 2004 and 31 December 2013. The measured spectrum confirms a flux suppression at the highest energies. Above 5.3×10185.3{\times}10^{18} eV, the "ankle", the flux can be described by a power law EγE^{-\gamma} with index γ=2.70±0.02(stat)±0.1(sys)\gamma=2.70 \pm 0.02 \,\text{(stat)} \pm 0.1\,\text{(sys)} followed by a smooth suppression region. For the energy (EsE_\text{s}) at which the spectral flux has fallen to one-half of its extrapolated value in the absence of suppression, we find Es=(5.12±0.25(stat)1.2+1.0(sys))×1019E_\text{s}=(5.12\pm0.25\,\text{(stat)}^{+1.0}_{-1.2}\,\text{(sys)}){\times}10^{19} eV.Comment: Replaced with published version.     Sigma E Regulators Control Hemolytic Activity and Virulence in a Shrimp Pathogenic Vibrio harveyi

    Members of the genus Vibrio are important marine and aquaculture pathogens. Hemolytic activity has been identified as a virulence factor in many pathogenic vibrios including V. cholerae, V. parahaemolyticus, V. alginolyticus, V. harveyi and V. vulnificus. We have used transposon mutagenesis to identify genes involved in the hemolytic activity of shrimp-pathogenic V. harveyi strain PSU3316. Out of 1,764 mutants screened, five mutants showed reduced hemolytic activity on sheep blood agar and exhibited virulence attenuation in shrimp (Litopenaeus vannamei). Mutants were identified by comparing transposon junction sequences to a draft of assembly of the PSU3316 genome. Surprisingly none of the disrupted open reading frames or gene neighborhoods contained genes annotated as hemolysins. The gene encoding RseB, a negative regulator of the sigma factor (σE), was interrupted in 2 out of 5 transposon mutants, in addition, the transcription factor CytR, a threonine synthetase, and an efflux-associated cytoplasmic protein were also identified. Knockout mutations introduced into the rpoE operon at the rseB gene exhibited low hemolytic activity in sheep blood agar, and were 3-to 7-fold attenuated for colonization in shrimp. Comparison of whole cell extracted proteins in the rseB mutant (PSU4030) to the wild-type by 2-D gel electrophoresis revealed 6 differentially expressed proteins, including two down-regulated porins (OmpC-like and OmpN) and an upregulated protease (DegQ) which have been associated with σE in other organisms. Our study is the first report linking hemolytic activity to the σE regulators in pathogenic Vibrio species and suggests expression of this virulence-linked phenotype is governed by multiple regulatory pathways within the V. harveyi

    Nutrition and cancer: A review of the evidence for an anti-cancer diet

    It has been estimated that 30–40 percent of all cancers can be prevented by lifestyle and dietary measures alone. Obesity, nutrient sparse foods such as concentrated sugars and refined flour products that contribute to impaired glucose metabolism (which leads to diabetes), low fiber intake, consumption of red meat, and imbalance of omega 3 and omega 6 fats all contribute to excess cancer risk. Intake of flax seed, especially its lignan fraction, and abundant portions of fruits and vegetables will lower cancer risk. Allium and cruciferous vegetables are especially beneficial, with broccoli sprouts being the densest source of sulforophane. Protective elements in a cancer prevention diet include selenium, folic acid, vitamin B-12, vitamin D, chlorophyll, and antioxidants such as the carotenoids (α-carotene, β-carotene, lycopene, lutein, cryptoxanthin). Ascorbic acid has limited benefits orally, but could be very beneficial intravenously. Supplementary use of oral digestive enzymes and probiotics also has merit as anticancer dietary measures. When a diet is compiled according to the guidelines here it is likely that there would be at least a 60–70 percent decrease in breast, colorectal, and prostate cancers, and even a 40–50 percent decrease in lung cancer, along with similar reductions in cancers at other sites. Such a diet would be conducive to preventing cancer and would favor recovery from cancer as well
