Maize-Produced Ag2 as a Subunit Vaccine for Valley Fever
Coccidioides is the causative agent of San Joaquin Valley fever, a fungal disease prevalent in the semiarid regions of the Americas. Efforts to develop a fungal vaccine over the last 2 decades were unsuccessful. A candidate antigen, Antigen 2 (Ag2), is notoriously difficult to express in Escherichia coli, and this study sought to accumulate the antigen at high levels in maize. Transformed maize lines accumulated recombinant Ag2 at levels \u3e 1 g/kg. Mice immunized with this antigen and challenged with live Coccidioides arthroconidia showed a reduction in the fungal load when Ag2 derived from either E. coli or maize was loaded into glucan chitin particles. A fusion of Ag2 to dendritic cell carrier peptide (DCpep) induced a T-helper type 17 response in the spleen when orally delivered, indicative of a protective immune response. The maize production platform and the glucan chitin particle adjuvant system show promise for development of a Coccidioides vaccine, but further testing is needed to fully assess the optimal method of administration
Performance of layers fed diets containing different oils
A trial was carried out at the Animal Science Department of the Federal University of Viçosa to evaluate the performance of heavy and light layers. During four periods of twenty-eight days, 432 layers in the second production cycle, being 216 Hy Line W36 (light birds) and 216 Hy Line Brown (heavy birds), were used. A completely randomized experimental design with a 1+(4x2x2) factorial arrangement (one control treatment, four oil sources, two inclusion levels, and two genetic lines) with four replicates of six birds per experimental unit, was applied. The treatments consisted of diets containing 16.5% crude protein, formulated to supply the nutritional requirements, and the inclusion of each oil source at 2 or 4%. Soybean, canola, linseed and fish oils were used. The control diet did not contain any oil. The studied parameters were: feed intake (g/hen/day), egg production (%), egg weight (g), egg mass (g/hen/day), feed conversion ratio (kg feed /dozen of eggs and per kg eggs). Oil levels and sources did not influence egg mass, feed conversion ratio (per kg eggs and dozen eggs), or egg production. During the first three periods, independently of oil source or genetics, eggs from layers fed 4% oil were the heaviest, except for those of light birds fed soybean oil during the third period, which egg weight was similar for the 2 and 4% levels. Light layers presented lower feed intake and better feed conversion ratio (per kg eggs and dozen eggs) as compared to heavy layers, and there was no influence of the evaluated oil sources on feed intake
Water intake and digestive metabolism of broilers fed all-vegetable diets containing acidulated soybean soapstock
A study was conducted to compare live performance and digestive metabolism of broiler chickens fed all-vegetable diets (All-Veg) compared to a regular diet including animal by-products. Three feeds were formulated and provided to broilers according to the feeding program: pre-starter from 1 to 10 days, starter from 11 to 21 days, and grower from 21 to 35 days. All feeds had corn and soybean meal as major ingredients; however, two of them were all-vegetable diets having either Degummed Soybean Oil (DSO) or Acidulated Soybean Soapstock (ASS) as fat sources. The third diet included poultry by-product and poultry fat. A total number of 360 day-old broiler chicks were allocated to 1m² battery cages, 10 chicks in each, and 12 replicates per treatment. Live performance was similar between groups of birds receiving the different diets with the exception of weight gain, which was increased for birds fed the All-Veg diet with ASS. Birds fed All-Veg diets had increased water intake and produced more excreta with a concurrent reduced feed metabolizability at both ages, regardless of fat source. Metabolizable Energy was not different for the three diets
