    Alterations of the giant pyramidal neurons (Betz cells) in brain cortex of rat offspring born from gestational diabetic dams: A morphometric study [Alteraciones de las Neuronas Piramidales Gigantes de la Corteza Cerebral en Crías de Ratas Nacidas de Hembras con Diabetes Gestacional: Un Estudio Morfométrico]

    A few studies reported the adverse effects of gestational diabetes on hippocampus and spinal cord of rat offspring. Giant pyramidal neurons are giant pyramidal neurons located in fifth layers of the gray matter in the primary motor cortex. Therefore, this study was conducted to determine the effect of gestational diabetes on the giant pyramidal neurons and the thickness of internal pyramidal layer in the brain cortex of rat offspring. On day 1 of gestation, 10 Wistar rat dams were randomly allocated into two control and diabetic groups. Five animals in diabetic group received 40 mg/kg/BW of Streptozotocin (intraperitoneally) and control animals received normal saline. We randomly selected six offspring of every subject in both groups at day 28, 56 and 84. Rat offspring were scarified and then coronal sections were taken from the right brain cortex and sections were stained with Cresyl violet. The density of giant pyramidal neurons in brain cortex and thickness of internal pyramidal layer of brain cortex were evaluated. In P28, P56, P84 the Betz cells density of brain cortex were significantly reduced from 107.6±6.2, 131.6±4.6 and 143.5±4.0 in controls to 84.96±2.1, 109.8±7.3 and 121.05±5.6 in cases (p<0.05), respectively. The thickness of the internal pyramidal layer of brain cortex in P28, 56 and P84 was significantly higher in gestational diabetic group in comparison with the control group (p<0.05). This study showed that uncontrolled gestational diabetes reduces the giant pyramidal neurons density and internal pyramidal layer thickness in brain cortex of rat offspring. © 2015, International Journal of Morphology.     Possible role of TORCH agents in congenital malformations in Gorgan, northern Islamic Republic of Iran

    This descriptive, cross-sectional study was carried out to explore the frequency of contamination with TORCH agents in neonates with congenital malformations in a referral centre in Gorgan city, Islamic Republic of Iran. Blood samples were taken from 64 neonates and their mothers over a 20-month period in 2003-04. Serologic tests showed that 4/64 infants born with congenital malformations (6%) had positive IgM antibody titres for Toxoplasma gondii (2 cases), rubella virus (1 case) and cytomegalovirus (1 case). IgM was positive in 9/63 mothers (14%), also for T. gondii (3 cases), rubella virus (3 cases) and cytomegalovirus (3 cases). No cases of herpes simplex virus type II or Treponema pallidum were found

    Effect of Vitamin E on Oocytes Apoptosis in nicotine-treated Mice

    Objective(s): Cigarette and nicotine enhances embryogenesis, fertility, pregnancy loss and ultrastructure alterations of oocyte. This study was performed to determine the effect of daily supplementation of vitamin E on oocytes apoptosis in nicotine-treated mice. Materials and Methods: In this experimental study, 24 NMARI adult female mice were randomly allocated into four experimental groups. For 30 days, animals in control group (C) were received saline through subcutaneous injection, group I received vitamin E (60 mg/kg/day orally), group II received nicotine (5 mg/kg/day, subcutaneous) and animals of group III received nicotine with vitamin E (60 mg/kg/day orally). After 30 days, the animals were superovulated with PSMG (10 Units) and HCG (10 Units). Next day animals were sacrificed and oocytes were flushed. Collected oocytes were examined through TUNEL assay for the determination of apoptosis through the use of fluorescent microscope. Results: The number of retrieved oocytes was 139, 148, 97 and 127 in control, experimental group I, II and III, respectively. Nicotine treatment increased apoptosis in oocytes up to 13.4% whereas oocytes apoptosis was 3.6% in controls. Supplementation with vitamin E in nicotine-treated mice reduced the oocytes apoptosis to 5.5%. Conclusion: This study showed that nicotine exposure (5 mg/kg/day for 30 days) can increase apoptosis in oocytes, and supplementation with vitamin E (60 mg/kg/day orally) can reduce the oocytes apoptosis in nicotine-treated mice

    Epidemiology of neural tube defects in northern Iran, 1998-2003

    We determined the rates of neural tube defects at a referral hospital in Gorgan, north Islamic Republic of Iran, and the relations of these abnormalities to sex, maternal ethnicity, maternal age and season. During 1998-2003, there were 109 cases among 37951 births, a prevalence of 28.7 per 10000 (24.8 and 32.8 per 10000 among males and females respectively). The rates in Turkmen, native Fars and Sistani ethnic groups were 40.5, 25.2 and 30.8 per 10000 respectively. The rates of spina bifida and anencephaly were 16.3 and 11.3 per 10000 respectively.     Maternal serum zinc deficiency in cases of neural tube defect in Gorgan, north Islamic Republic of Iran

    Previous studies have shown a high rate of neural tube defects (NTD) in Gorgan, northern Islamic Republic of Iran. This case-control study during 2003-04 compared serum zinc levels and other variables in 23 mothers of neonates affected with NTD and 36 mothers with normal healthy neonates in Dezyani hospital in Gorgan. Mean serum zinc levels in the case and control groups were 13.43 μmol/L (SD 6.3) and 11.41 μmol/L (SD 6.3) respectively. Zinc deficiency was found in 13 (36.5%) of the cases and 7 (19.4%) of the controls. Logistic regression analysis showed an association between the presence of NTD and zinc deficiency (OR 5.06; 95% CI: 1.51-16.94)

    Maternal risk factors for oral clefts: A case-control study

    Introduction: A cleft lip with or without a cleft palate is one of the major congenital anomalies observed in newborns. This study explored the risk factors for oral clefts in Gorgan, Northern Iran. Materials and Methods: This hospital-based case-control study was performed in three hospitals in Gorgan, Northern Iran between April 2006 and December 2009. The case group contained 33 newborns with oral clefts and the control group contained 63 healthy newborns. Clinical and demographic factors, including date of birth, gender of the newborns, type of oral cleft, consanguinity of the parents, parental ethnicity, and the mother's parity, age, education and intake of folic acid were recorded for analysis. Results: A significant association was found between parity higher than 2 and the risk of an oral cleft (OR= 3.33, CI 95% [1.20, 9.19], P> 0.02). According to ethnicity, the odds ratio for oral clefts was 0.87 in Turkmens compared with Sistani people (CI 95% [0.25, 2.96]) and 1.11 in native Fars people compared with Sistani people (CI 95% [0.38, 3.20]). A lack of folic acid consumption was associated with an increased risk of oral clefts but this was not statistically significant (OR = 1.42, CI 95% [0.58, 3.49]). There were no significant associations between sex (OR boy/girl = 0.96, CI 95% [0.41, 2.23]), parent familial relations (OR = 1.07, CI 95% [0.43, 2.63]), mother's age and oral clefts. Conclusions: The results of this study indicate that higher parity is significantly associated with an increased risk of an oral cleft, while Fars ethnicity and a low intake of folic acid increased the incidence of oral clefts but not significantly

    Preventing effect of vitamin E on oocytes apoptosis in morphinetreated mice

    Several studies have shown that Morphine Sulfate affects on fertility, embryogenesis and consequent pregnancy loss and ultrastructural alterations of oocytes in animal model. This study was done to determine the effect of morphine sulfate on oocytes apoptosis and preventive role of daily supplementation of Vitamin E on oocytes apoptosis in morphine sulfate -treated mice. Twenty-four NMARI female mice were randomly allocated into four experimental groups. For 15 days, control group received saline (0.2 ml/day by subcutaneous injection), group I Vitamin E (60 mg/kg/day orally), group II Morphine Sulfate (10 mg/kg/day by subcutaneous injection) and group III Morphine Sulfate with Vitamin E (60 mg/kg/day orally). Then, animals were superovulated with PSMG (10 Units) and 10 Unites of HCG. The next day the animals were sacrificed, oocytes were flushed from each fallopian tube. The collected oocytes were subjected to determine apoptosis by Tunnel assay with using Fluorescent Microscope. According to our results, the number of retrieved oocytes were 121, 132, 86 and 114 in control, experimental group I, II and III, respectively. Morphine Sulfate treatment increased apoptosis in oocytes to 17.44% whereas oocytes apoptosis was 4.13% in Controls. Supplementation with Vitamin E in Morphine Sulfate -treated mice reduced the oocytes apoptosis to 7.01%. This study showed that Morphine can increase apoptosis in oocytes and Vitamin E treatment significantly reduces oocytes apoptosis in the Morphine Sulfate -treated mice

    Study of embryotoxicity of mentha piperita l. during organogenesis in balb/c mice

    Mentha piperita (Labiatae), commonly known as peppermint is a native Iranian herb which is used in folk medicine for various purposes. This study was carried out to reveal the teratogenic effect of Mentha piperita on mice fetuses. In this experimental study, pregnant Balb/c mice divided to four groups. Case group received 600 (treatment I) and 1200 (treatment II) mg/kg/day the hydroalcoholic extract of Mentha piperita during 6-15 of gestational days and one control group received normal saline during GD6-GD15 by gavages and other control group did not receive any matter during 6-15 of gestational days. Mice sacrificed at GD18 and embryos were collected. Macroscopic observation was done by stereomicroscope. 20 fetuses of each group were stained by Alizarin red-S and Alcian blue staining method. The Mean weight of fetuses decreased in treatment groups rather than control (P<0.05) but CRL there was no significant difference between treatments and controls groups. In the treatment I (600 mg/kg/day) and treatment II (1200 mg/kg/day), normal saline and control group, no gross congenital malformations were observed in fetuses. Treated fetuses also had no delayed bone ossification as determined by Alizarin red-S and Alcian blue staining method. This study showed that the hydroalcoholic extract of Mentha piperita (600 and 1200 mg/ kg/day) has no teratogenic effect in mice fetuses if used continuously during embryonic period

    In this article, a full-term male neonate with Harlequin fetus has been reported. It was born from a 20 year old mother by the Caesarean operation. In the clinical examination, broad, thick and hard hyperkeratotic disks were observed all troughs the body with extended gashes among them in the skin. The deformity of the face, the eyes server ectropion, the orbital chemosis and the flattening of the nose and the ears were among other disorders to be observed. The lips were overturned. There were no nails or hairs to be seen. The movements of the joint were limited. Hands and legs, in particular in the extremities were on a state of fixed gangrene and seemed to be ischemic. The patient died in the third day of hospitalization due to indicators of respiratory infections and the sepsis
