Rapid identification of Salmonella typhimurium, S. enteritidis and S. virchow isolates by Polymerase Chain Reaction based fingerprinting methods
In this study we used and evaluated three rapid molecular typing methods for the identification of three frequent, clinically significant Salmonella serovars on the basis of the ease, simplicity and reproducibility of the chosen methods. We determined the genetic diversity among several isolates of Salmonella enteritidis, S. typhimurium and S. virchow, and compared them with other enterobacteria by using the repetitive extragenic palindromic (REP) sequences, the enterobacterial repetitive intergenic consensus (ERIC) sequences, and the 16S–23S rDNA intergenic spacer region (ITS1). The objective was to evaluate their potential application to discriminate among members of the species Salmonella enterica subspecies enterica using the genetic diversity of the group found by genomic fingerprinting. The three different serovars of Salmonella studied gave reproducible and distinguishable profiles using whichever of the above mentioned polymerase chain reaction (PCR) methods assayed. The conserved patterns in each serovar allowed for easy differentiation from other serovars of Salmonella

La expresión plástica y desarrollo de motricidad fina en niños de cuatro años de la Institución Educativa N°21011 - Barranca, 2021
El objetivo principal del estudio fue determinar la relación entre expresión
plástica y motricidad fina en los escolares de cuatro años de la I.E. N°21011,
UGEL 16 – Barranca, 2021. Fue un estudio aplicado, cuantitativo fue el enfoque,
correlacional descriptivo el nivel de investigación y no experimental correlacional
descriptivo el diseño del estudio y de corte transaccional. Se tuvo como muestra
a 80 escolares de cuatro años. Se administró la observación como técnica y dos
guías de observación validados y confiables como instrumento. Entre los
resultados se determinó la relación entre expresión plástica y motricidad fina en
los escolares de cuatro años de la I.E. N°21011, UGEL 16 – Barranca, 2021,
evidenciándose el Rho=0,820, por lo tanto, se afirma la existencia la correlación
de nivel alto además positivo. También evidenciando la significancia pvalor=0.000<α=0.05 entonces, se rechaza la hipótesis nula del estudio y acepta
la hipótesis alterna, por lo tanto, se afirma que la expresión plástica y la
motricidad fina se relaciona significativamente en los escolares de cuatro años
A Growth Modulation Index-Based GEISTRA Score as a New Prognostic Tool for Trabectedin Efficacy in Patients with Advanced Soft Tissue Sarcomas: A Spanish Group for Sarcoma Research (GEIS) Retrospective Study
L-sarcoma; Índex de modulació del creixement; SarcomaL-sarcoma; Indice de modulación del crecimiento; SarcomaL-sarcoma; Growth modulation index; SarcomaThe aim of this study was to identify an easily reliable prognostic score that selects the subset of advanced soft tissue sarcoma (ASTS) patients with a higher benefit with trabectedin in terms of time to progression and overall survival. A retrospective series of 357 patients with ASTS treated with trabectedin as second- or further-line in 19 centers across Spain was analyzed. First, it was confirmed that patients with high growth modulation index (GMI > 1.33) were associated with the better clinical outcome. Univariate and multivariate analyses were performed to identify factors associated with a GMI > 1.33. Thus, GEISTRA score was based on metastasis free-interval (MFI ≤ 9.7 months), Karnofsky 1.33. The lowest GEISTRA score showed a median of time-to-progression (TTP) and overall survival (OS) of 5.7 and 19.5 months, respectively, whereas it was 1.8 and 3.1 months for TTP and OS, respectively, for the GEISTRA 4 score. This prognostic tool can contribute to better selecting candidates for trabectedin treatment in ASTS.This research was funded by the Spanish Group for Research on Sarcoma (grant number: NA) and partially by PharmaMar. Protection against Pseudomonas aeruginosa lung infection in mice by recombinant OprF-pulsed dendritic cell immunization
BACKGROUND: The Pseudomonas aeruginosa major constitutive outer membrane porin protein F (OprF) has been shown to be a protective antigen and was previously used to activate an immunological response in a mouse model of lung pneumonia. The purpose of our study was to demonstrate the ability of mouse dendritic cells pulsed with purified or recombinant OprF to protect mice against P. aeruginosa infection and inflammation.Both native (n-OprF), isolated and purified from PAO1 bacterial strain, and recombinant (histidin-conjugated) OprF (His-OprF), obtained by cloning of the oprF gene into the pET28a expression vector, were used to stimulate dendritic cells in vitro before adoptive transfer into prospective recipient mice with P. aeruginosa pulmonary infection. RESULTS: Similar to n-OprF, His-OprF activated dendritic cells in vitro, inducing the costimulatory molecule expression as well as cytokine production. Upon adoptive transfer in vivo, porin-pulsed dendritic cells (DCs) induced Th1-mediated resistance to infection and associated inflammatory pathology caused by either the PAO1 strain or a clinically-isolated mucoid strain. CONCLUSIONS: This study highlights the pivotal contribution of DCs to vaccine-induced protection against P. aeruginosa infection and associated inflammation
Plasma Molecular Signatures in Hypertensive Patients With Renin-Angiotensin System Suppression: New Predictors of Renal Damage and De Novo Albuminuria Indicators
Albuminuria is a risk factor strongly associated with cardiovascular disease, the first cause of death in the general population. It is well established that renin-angiotensin system suppressors prevent the development of new-onset albuminuria in naïf hypertensive patients and diminish its excretion, but we cannot forget the percentage of hypertensive patients who develop de novo albuminuria. Here, we applied multiple proteomic strategy with the purpose to elucidate specific molecular pathways involved in the pathogenesis and provide predictors and chronic organ damage indicators. Briefly, 1143 patients were followed up for a minimum period of 3 years. One hundred and twenty-nine hypertensive patients chronically renin-angiotensin system suppressed were recruited, classified in 3 different groups depending on their albuminuria levels (normoalbuminuria, de novo albuminuria, and sustained albuminuria), and investigated by multiple proteomic strategies. Our strategy allowed us to perform one of the deepest plasma proteomic analysis to date, which has shown 2 proteomic signatures: (1) with predictive value of de novo albuminuria and (2) sustained albuminuria indicator proteins. These proteins are involved in inflammation, immune as well as in the proteasome activation occurring in situations of endoplasmic reticulum stress. Furthermore, these results open the possibility of a future strategy based on anti-immune therapy to treat hypertension which could help to prevent the development of albuminuria and, hence, the progression of kidney damage.N
Comportamiento epidemiológico de los factores de riesgo asociados a enfermedades crónicas no transmisibles en estudiantes universitarios
Describir el comportamiento epidemiológico de los factores de riesgo relacionados a enfermedades crónicas no transmisibles asociados a hipertrigliceridemia presentes en los estudiantes del primer año de la carrera de Médico y Cirujano de la Universidad de San Carlos de Guatemala. Estudio descriptivo transversal realizado en 1097 estudiantes, en los cuales se aplicó el cuestionario del método paso a paso para la vigilancia de factores de riesgo de enfermedades crónicas (STEPS) modificado. Se encontró 610 (56%) estudiantes sexo femenino, la media de edad fue de 19 años (±1.56); en relación a los factores de riesgo modificables: 219 (20%) consumen cigarrillo, 274 (25%) consumen alcohol nocivamente, 838 (76%) se alimentan de forma inadecuada, 887 (81%) no practica actividad física
significativa; se identificó que 405 (37%) presentaron un índice cintura – cadera considerado de riesgo. En cuanto a los factores asociados a la presencia de hipertrigliceridemia: obesidad se identificó en 147 (13%) estudiantes (X2 de 104.98 y OR de 7), obesidad central en 204 (19%) (X2 de 55.74 y OR de 3), el sobrepeso en 330 (30%) (X2 de 53.4727 y OR de 3), presión arterial alta en 397 (36%) (X2 de 18.63 y OR de 2), y glucemia alterada en ayunas alta en 198 (18%) (X2 de 8.3 y OR de 2). La hipertrigliceridemia se presentó en 411 estudiantes (37%), siendo la edad más afectada la de 19 años. La mayoría de estudiantes estudiados es de sexo femenino. Existe una alta prevalencia de factores de riesgo modificables asociados a enfermedades crónicas no transmisibles, de estos se identificó que factores como obesidad, obesidad central, sobrepeso, presión arterial alta y glucemia alterada en ayunas alta tienen una significativa asociación al desarrollo de hipertrigliceridemia, con un aumento del riesgo de 7, 3, 3, 2 y 2, respectivamente
Un llamado ético a la inclusión de mujeres embarazadas en investigación: Reflexiones del Foro Global de Bioética en Investigación
El Foro Global de Bioética en Investigación (GFBR por sus siglas en inglés) se
reunió el 3 y 4 de noviembre en Buenos Aires, Argentina, con el objetivo de discutir
la ética de la investigación con mujeres embarazadas. El GFBR es una plataforma
mundial que congrega a actores clave con el objetivo de promover la investigación
realizada de manera ética, fortalecer la ética de la investigación en salud, particularmente
en países de ingresos bajos y medios, y promover colaboración entre
países del norte y del sur.a Los participantes en el GFBR provenientes de
Latinoamérica incluyeron a eticistas, investigadores, miembros de comités de ética
y representantes de autoridades sanitarias provenientes de Argentina, Brasil, Chile,
Colombia, Ecuador, El Salvador, Guatemala, Honduras, Panamá, Perú, Nicaragua
y la República Dominicana.
Una legítima preocupación por la protección de las mujeres embarazadas y sus
embriones o fetos ha llevado a la mayoría de los países de la Región de las Américas
a limitar la realización de estudios con mujeres embarazadas exclusivamente a
aquellos estudios específicos sobre el embarazo, y a requerir la exclusión sistemática
de las mujeres embarazadas o de las mujeres que quedan embarazadas en el
curso del estudio. Ciertamente, a lo largo de la historia de la ética de la investigación,
se ha creído erróneamente que proteger a una población es sinónimo de excluirla
de los estudios. Se sabe ahora que proceder así implica exponer a riesgos
mucho mayores a la población que se busca proteger.
El embarazo implica cambios fisiológicos sustantivos e impacta profundamente
la manera como el cuerpo metaboliza los medicamentos. Sin embargo,
por evitar hacer investigación con mujeres embarazadas, no se ha producido la
evidencia científica necesaria para tomar decisiones sobre tratamientos e intervenciones
preventivas con dosis eficaces y seguras para ellas y sus embriones o
fetos. A manera de ilustración, en el 2001 había en los Estados Unidos apenas
más de una docena de medicamentos aprobados para uso en el embarazo (1) y
en el 2011 la Food and Drug Administration (FDA) aprobó por primera vez en
15 años un medicamento para su uso en el embarazo (2). Como consecuencia
de no haber producido la evidencia necesaria, se pone en riesgo la salud de las
mujeres embarazadas cada vez que se les da atención médica. Las mujeres
embarazadas se enferman y las mujeres enfermas se embarazan, y no se sabe si
los medicamentos que se les da son eficaces o siquiera seguros para ellas y sus
embriones o fetos
Markers of endothelial damage in patients with chronic kidney disease on hemodialysis
Patients with Stage 5 chronic kidney disease who are on hemodialysis (HD) remain in a chronic inflammatory state, characterized by the accumulation of uremic toxins that induce endothelial damage and cardiovascular disease (CVD). Our aim was to examine microvesicles (MVs), monocyte subpopulations, and angiopoietins (Ang) to identify prognostic markers in HD patients with or without diabetes mellitus (DM). A total of 160 prevalent HD patients from 10 centers across Spain were obtained from the Biobank of the Nephrology Renal Network (Madrid, Spain): 80 patients with DM and 80 patients without DM who were matched for clinical and demographic criteria. MVs from plasma and several monocyte subpopulations (CD142+/CD16+, CD14+/CD162+) were analyzed by flow cytometry, and the plasma concentrations of Ang1 and Ang2 were quantified by ELISA. Data on CVD were gathered over the 5.5 yr after these samples were obtained. MV level, monocyte subpopulations (CD14+/CD162+ and CD142+/CD16+), and Ang2-to-Ang1 ratios increased in HD patients with DM compared with non-DM patients. Moreover, MV level above the median (264 MVs/µl) was associated independently with greater mortality. MVs, monocyte subpopulations, and Ang2-to-Ang1 ratio can be used as predictors for CVD. In addition, MV level has a potential predictive value in the prevention of CVD in HD patients.
