    Contours of Inclusion: Inclusive Arts Teaching and Learning

    The purpose of this publication is to share models and case examples of the process of inclusive arts curriculum design and evaluation. The first section explains the conceptual and curriculum frameworks that were used in the analysis and generation of the featured case studies (i.e. Understanding by Design, Differentiated Instruction, and Universal Design for Learning). Data for the cases studies was collected from three urban sites (i.e. Los Angeles, San Francisco, and Boston) and included participant observations, student and teacher interviews, curriculum documentation, digital documentation of student learning, and transcripts from discussion forum and teleconference discussions from a professional learning community.The initial case studies by Glass and Barnum use the curricular frameworks to analyze and understand what inclusive practices look like in two case studies of arts-in-education programs that included students with disabilities. The second set of precedent case studies by Kronenberg and Blair, and Jenkins and Agois Hurel uses the frameworks to explain their process of including students by providing flexible arts learning options to support student learning of content standards. Both sets of case studies illuminate curricular design decisions and instructional strategies that supported the active engagement and learning of students with disabilities in educational settings shared with their peers. The second set of cases also illustrate the reflective process of using frameworks like Universal Design for Learning (UDL) to guide curricular design, responsive instructional differentiation, and the use of the arts as a rich, meaningful, and engaging option to support learning.     Regulation of smooth muscle α-actin expression and hypertrophy in cultured mesangial cells.BackgroundMesangial cells during embryonic development and glomerular disease express smooth muscle α-actin (α-SMA). We were therefore surprised when cultured mesangial cells deprived of serum markedly increased expression of α-SMA. Serum-deprived mesangial cells appeared larger than serum-fed mesangial cells. We hypothesized that α-SMA expression may be more reflective of mesangial cell hypertrophy than hyperplasia.MethodsHuman mesangial cells were cultured in medium alone or with fetal bovine serum, thrombin, platelet-derived growth factor-BB (PDGF-BB) and/or transforming growth factor-β1 (TGF-β1). α-SMA expression was examined by immunofluorescence, Western blot, and Northern blot analysis. Cell size was analyzed by forward light scatter flow cytometry.Resultsα-SMA mRNA was at least tenfold more abundant after three to five days in human mesangial cells plated without serum, but β-actin mRNA was unchanged. Serum-deprived cells contained 5.3-fold more α-SMA after three days and 56-fold more after five days by Western blot. Serum deprivation also increased α-SMA in rat and mouse mesangial cells. The effects of serum deprivation on α-SMA expression were reversible. Mesangial cell mitogens, thrombin or PDGF-BB, decreased α-SMA, but TGF-β1 increased α-SMA expression and slowed mesangial cell proliferation in serum-plus medium. Flow cytometry showed that serum deprivation or TGF-β1 treatment caused mesangial cell hypertrophy. PDGF-BB, thrombin, or thrombin receptor-activating peptide blocked hypertrophy in response to serum deprivation.ConclusionsWe conclude that increased α-SMA expression in mesangial cells reflects cellular hypertrophy rather than hyperplasia

    Madagascar’s Mangroves: Quantifying Nation-Wide and Ecosystem Specific Dynamics, and Detailed Contemporary Mapping of Distinct Ecosystems

    Mangrove ecosystems help mitigate climate change, are highly biodiverse, and provide critical goods and services to coastal communities. Despite their importance, anthropogenic activities are rapidly degrading and deforesting mangroves world-wide. Madagascar contains 2% of the world’s mangroves, many of which have undergone or are starting to exhibit signs of widespread degradation and deforestation. Remotely sensed data can be used to quantify mangrove loss and characterize remaining distributions, providing detailed, accurate, timely and updateable information. We use USGS maps produced from Landsat data to calculate nation-wide dynamics for Madagascar’s mangroves from 1990 to 2010, and examine change more closely by partitioning the national distribution in to primary (i.e., \u3e1000 ha) ecosystems; with focus on four Areas of Interest (AOIs): Ambaro-Ambanja Bays (AAB), Mahajamba Bay (MHJ), Tsiribihina Manombolo Delta (TMD) and Bay des Assassins (BdA). Results indicate a nation–wide net-loss of 21% (i.e., 57,359 ha) from 1990 to 2010, with dynamics varying considerably among primary mangrove ecosystems. Given the limitations of national-level maps for certain localized applications (e.g., carbon stock inventories), building on two previous studies for AAB and MHJ, we employ Landsat data to produce detailed, contemporary mangrove maps for TMD and BdA. These contemporary, AOI-specific maps provide improved detail and accuracy over the USGS national-level maps, and are being applied to conservation and restoration initiatives through the Blue Ventures’ Blue Forests programme and WWF Madagascar West Indian Ocean Programme Office’s work in the region

    Bioclimatic envelope models predict a decrease in tropical forest carbon stocks with climate change in Madagascar

    Recent studies have underlined the importance of climatic variables in determining tree height and biomass in tropical forests. Nonetheless, the effects of climate on tropical forest carbon stocks remain uncertain. In particular, the application of process-based dynamic global vegetation models has led to contrasting conclusions regarding the potential impact of climate change on tropical forest carbon storage. Using a correlative approach based on a bioclimatic envelope model and data from 1771 forest plots inventoried during the period 1996–2013 in Madagascar over a large climatic gradient, we show that temperature seasonality, annual precipitation and mean annual temperature are key variables in determining forest above-ground carbon density. Taking into account the explicative climate variables, we obtained an accurate (R2 = 70% and RMSE = 40 Mg ha−1) forest carbon map for Madagascar at 250 m resolution for the year 2010. This national map was more accurate than previously published global carbon maps (R2 ≤ 26% and RMSE ≥ 63 Mg ha−1). Combining our model with the climatic projections for Madagascar from 7 IPCC CMIP5 global climate models following the RCP 8.5, we forecast an average forest carbon stock loss of 17% (range: 7–24%) by the year 2080. For comparison, a spatially homogeneous deforestation of 0.5% per year on the same period would lead to a loss of 30% of the forest carbon stock. Synthesis. Our study shows that climate change is likely to induce a decrease in tropical forest carbon stocks. This loss could be due to a decrease in the average tree size and to shifts in tree species distribution, with the selection of small-statured species. In Madagascar, climate-induced carbon emissions might be, at least, of the same order of magnitude as emissions associated with anthropogenic deforestation

    Participatory planning of a community-based payments for ecosystem services initiative in Madagascar’s mangroves

    Although the dynamics of coastal resources are largely determined by the impacts of human users, spatially-explicit social data are rarely systematically integrated into coastal management planning in data-poor developing states. In order to plan a community-based mangrove payments for ecosystem services initiative in southwest Madagascar, we used two participatory approaches – public participation geographic information systems and concept modelling workshops – with 10 coastal communities to investigate the dynamics and spatial distribution of the mangrove resources they use. In each village we conducted participatory mapping of land and resource use with different livelihood groups using printed satellite images, and concept modelling workshops to develop concept models of the mangrove social-ecological system (including identification of threats and underlying drivers and proposal of targeted management strategies). Each community then proposed mangrove zoning consisting of strict conservation zones, sustainable use zones and restoration zones. Following validation and ground-truthing, the zones and management strategies proposed formed the basis of the zoning and management plan for the mangrove. Participatory approaches proved a simple and reliable way to gather spatial data and better understand the relationships between the mangrove and those who use it. Moreover, participation stimulated mangrove users to consider resource trends, the impacts of their activities, and required management actions, promoting a collective ‘buy-in’ for the project. Since participation extended beyond research to the development of management zones, rules and strategies, we believe that community ownership of the project has been strengthened and the chances of successfully conserving the mangrove improved

    Extended 2D myotube culture recapitulates postnatal fibre type plasticity

    Background: The traditional problems of performing skeletal muscle cell cultures derived from mammalian or avian species are limited myotube differentiation, and transient myotube persistence which greatly restricts the ability of myotubes to undergo phenotypic maturation. We report here on a major technical breakthrough in the establishment of a simple and effective method of extended porcine myotube cultures (beyond 50 days) in two-dimension (2D) that recapitulates key features of postnatal fibre types. Results: Primary porcine muscle satellite cells (myoblasts) were isolated from the longissimus dorsi of 4 to 6 weeks old pigs for 2D cultures to optimise myotube formation, improve surface adherence and characterise myotube maturation. Over 95 % of isolated cells were myoblasts as evidenced by the expression of Pax3 and Pax7. Our relatively simple approach, based on modifications of existing surface coating reagents (Maxgel), and of proliferation and differentiation (Ultroser G) media, typically achieved by 5 days of differentiation fusion index of around 80 % manifested in an abundance of discrete myosin heavy chain (MyHC) slow and fast myotubes. There was little deterioration in myotube viability over 50 days, and the efficiency of myotube formation was maintained over seven myoblast passages. Regular spontaneous contractions of myotubes were frequently observed throughout culture. Myotubes in extended cultures were able to undergo phenotypic adaptation in response to different culture media, including the adoption of a dominant postnatal phenotype of fast-glycolytic MyHC 2x and 2b expression by about day 20 of differentiation. Furthermore, fast-glycolytic myotubes coincided with enhanced expression of the putative porcine long intergenic non-coding RNA (linc-MYH), which has recently been shown to be a key coordinator of MyHC 2b expression in vivo. Conclusions: Our revised culture protocol allows the efficient differentiation and fusion of porcine myoblasts into myotubes and their prolonged adherence to the culture surface. Furthermore, we are able to recapitulate in 2D the maturation process of myotubes to resemble postnatal fibre types which represent a major technical advance in opening access to the in vitro study of coordinated postnatal muscle gene expression

    PIK3CA-associated developmental disorders exhibit distinct classes of mutations with variable expression and tissue distribution

    Mosaicism is increasingly recognized as a cause of developmental disorders with the advent of next-generation sequencing (NGS). Mosaic mutations of PIK3CA have been associated with the widest spectrum of phenotypes associated with overgrowth and vascular malformations. We performed targeted NGS using 2 independent deep-coverage methods that utilize molecular inversion probes and amplicon sequencing in a cohort of 241 samples from 181 individuals with brain and/or body overgrowth. We identified PIK3CA mutations in 60 individuals. Several other individuals (n = 12) were identified separately to have mutations in PIK3CA by clinical targetedpanel testing (n = 6), whole-exome sequencing (n = 5), or Sanger sequencing (n = 1). Based on the clinical and molecular features, this cohort segregated into three distinct groups: (a) severe focal overgrowth due to low-level but highly activating (hotspot) mutations, (b) predominantly brain overgrowth and less severe somatic overgrowth due to less-activating mutations, and (c) intermediate phenotypes (capillary malformations with overgrowth) with intermediately activating mutations. Sixteen of 29 PIK3CA mutations were novel. We also identified constitutional PIK3CA mutations in 10 patients. Our molecular data, combined with review of the literature, show that PIK3CA-related overgrowth disorders comprise a discontinuous spectrum of disorders that correlate with the severity and distribution of mutations

    The Tools for Integrated Management of Childhood Illness (TIMCI) study protocol: a multi-country mixed-method evaluation of pulse oximetry and clinical decision support algorithms.

    Effective and sustainable strategies are needed to address the burden of preventable deaths among children under-five in resource-constrained settings. The Tools for Integrated Management of Childhood Illness (TIMCI) project aims to support healthcare providers to identify and manage severe illness, whilst promoting resource stewardship, by introducing pulse oximetry and clinical decision support algorithms (CDSAs) to primary care facilities in India, Kenya, Senegal and Tanzania. Health impact is assessed through: a pragmatic parallel group, superiority cluster randomised controlled trial (RCT), with primary care facilities randomly allocated (1:1) in India to pulse oximetry or control, and (1:1:1) in Tanzania to pulse oximetry plus CDSA, pulse oximetry, or control; and through a quasi-experimental pre-post study in Kenya and Senegal. Devices are implemented with guidance and training, mentorship, and community engagement. Sociodemographic and clinical data are collected from caregivers and records of enrolled sick children aged 0-59 months at study facilities, with phone follow-up on Day 7 (and Day 28 in the RCT). The primary outcomes assessed for the RCT are severe complications (mortality and secondary hospitalisations) by Day 7 and primary hospitalisations (within 24 hours and with referral); and, for the pre-post study, referrals and antibiotic. Secondary outcomes on other aspects of health status, hypoxaemia, referral, follow-up and antimicrobial prescription are also evaluated. In all countries, embedded mixed-method studies further evaluate the effects of the intervention on care and care processes, implementation, cost and cost-effectiveness. Pilot and baseline studies started mid-2021, RCT and post-intervention mid-2022, with anticipated completion mid-2023 and first results late-2023. Study approval has been granted by all relevant institutional review boards, national and WHO ethical review committees. Findings will be shared with communities, healthcare providers, Ministries of Health and other local, national and international stakeholders to facilitate evidence-based decision-making on scale-up.Study registration: NCT04910750 and NCT05065320
