Performance of anti-Salmonella lactic acid bacteria in the porcine intestine
Of five anti-Salmonella porcine cultures administered to pigs at 1010 cfu/day, two Lactobacillus murinus strains demonstrated superior survival during gastrointestinal transit. Both were detected at ~107 -108 cfu/g faeces which was higher (P\u3c0.05) than Pediococcus pentosaceus DPC6006 (~105 cfu/g). One Lb. murinus strain was also excreted at higher numbers (P\u3c0.05) than either Lb. salivarius DPC6005 or Lb. pentosus DPC6004 (both ~106 cfu/g). The Lb. murinus strains persisted in both the faeces and the caecum for at least 9 days post-administration. Animals fed a combination of all five strains at 1010 cfu/day excreted ~107 cfu/g of the administered strains, which was higher (P\u3c0.05) than only P. pentosaceus DPC6006. Randomly amplified polymorphic DNA (RAPD) PCR analysis revealed that both Lb. murinus strains predominated in the faeces of these animals during administration, while post-administration, both Lb. murinus strains and Lb. pentosus DPC6004 were recovered from the faeces and the caecum while P. pentosaceus DPC6006 was only detected in the caecum. After 21 days of culture administration, faecal Enterobacteriaceae counts were reduced in pigs fed Lb. salivarius DPC6005, P. pentosaceus DPC6006, Lb. pentosus DPC6004 and the culture mix, though not significantly. Overall, the porcine intestinal isolates offer potential as probiotics for enteropathogen reduction in pigs; possibly as a combination due to strain variation
Outgassing Behavior and Heat Treatment Optimization of JSC-1A Lunar Regolith Simulant
As NASA Strives towards a Long Duration Presence on the Moon, It Has Become Increasingly Important to Learn How to Better Utilize Resources from the Lunar Surface for Everything from Habitats, Vehicle Infrastructure, and Chemical Extraction. to that End, a Variety of Lunar Simulants Have Been Sourced from Terrestrially Available Volcanic Minerals and Glass as Apollo Regolith is Unavailable for Experimentation Needing Large Masses. However, While Mineralogy and Chemical Composition Can Approach that of Lunar Material in These Simulants, There Are Still Distinct Non-Lunar Phases Such as Hydrates, Carbonates, Sulfates, and Clays that Can Cause Simulants to Behave Distinctly Non-Lunar in a Variety of Processing Conditions that Maybe Applied In-Situ to Lunar Material. Notably, Severe Glassy Bubbling Has Been Documented in a Variety of Vacuum Sintering Experiments on JSC-1A Lunar Mare Simulant Heated Via Microwaves. the Origins of This Outgassing Have Not Been Well Understood But Are Normally Attributed to the Decomposition of Non-Lunar Contaminates Intrinsic to Virtually All Terrestrially Sourced Simulants. as Such, a Series of Controlled Environmental Tests Were Performed to Ascertain the Origins of the High Temperature Outgassing and to Develop Heat Treatments that Can Drive JSC-1A Closer to Lunar Composition and Behavior. It Was Found that in JSC-1A at Elevated Temperatures Distinct Gas Evolutions of Water, Carbon Dioxide, and Sulfur Dioxide Occur in Both Inert Gas and Vacuum. Additionally, the Presence of Hydrogen during Heat Treatments Was Shown to Dramatically Change Gas Evolutions, Leading to Distinctly More Lunar-Like Composition and Behavior from JSC-1A Simulant
Estimating the horizontal and vertical direction-of-arrival of water-borne seismic signals in the northern Philippine Sea

The definitive version was published in Journal of the Acoustical Society of America 134 (2013): 3282, doi:10.1121/1.4818843.Conventional and adaptive plane-wave beamforming with simultaneous recordings by large-aperture horizontal and vertical line arrays during the 2009 Philippine Sea Engineering Test (PhilSea09) reveal the rate of occurrence and the two-dimensional arrival structure of seismic phases that couple into the deep ocean. A ship-deployed, controlled acoustic source was used to evaluate performance of the horizontal array for a range of beamformer adaptiveness levels. Ninety T-phases from unique azimuths were recorded between Yeardays 107 to 119. T-phase azimuth and S-minus-P-phase time-of-arrival range estimates were validated using United States Geological Survey seismic monitoring network data. Analysis of phases from a seismic event that occurred on Yearday 112 near the east coast of Taiwan approximately 450 km from the arrays revealed a 22° clockwise evolution of T-phase azimuth over 90 s. Two hypotheses to explain such evolution—body wave excitation of multiple sources or in-water scattering—are presented based on T-phase origin sites at the intersection of azimuthal great circle paths and ridge/coastal bathymetry. Propagation timing between the source, scattering region, and array position suggests the mechanism behind the evolution involved scattering of the T-phase from the Ryukyu Ridge and a T-phase formation/scattering location estimation error of approximately 3.2 km.This research is supported

A preliminary report of an educational intervention in practice management
BACKGROUND: Practice management education continues to evolve, and little information exists regarding its curriculum design and effectiveness for resident education. We report the results of an exploratory study of a practice management curriculum for primary care residents. METHODS: After performing a needs assessment with a group of primary care residents at Wright State University, we designed a monthly seminar series covering twelve practice management topics. The curriculum consisted of interactive lectures and practice-based application, whenever possible. We descriptively evaluated two cognitive components (practice management knowledge and skills) and the residents' evaluation of the curriculum. RESULTS: The mean correct on the knowledge test for this group of residents was 74% (n = 12) and 91% (n = 12) before and after the curriculum, respectively. The mean scores for the practice management skill assessments were 2.62 before (n = 12), and 3.65 after (n = 12) the curriculum (modified Likert, 1 = strongly disagree, 5 = strongly agree). The residents rated the curriculum consistently high. CONCLUSIONS: This exploratory study suggests that this curriculum may be useful in developing knowledge and skills in practice management for primary care residents. This study suggests further research into evaluation of this curriculum may be informative for practice-based education
Mechanism of Action of Two Flavone Isomers Targeting Cancer Cells with Varying Cell Differentiation Status

Apoptosis can be triggered in two different ways, through the intrinsic or the extrinsic pathway. The intrinsic pathway is mediated by the mitochondria via the release of cytochrome C while the extrinsic pathway is prompted by death receptor signals and bypasses the mitochondria. These two pathways are closely related to cell proliferation and survival signaling cascades, which thereby constitute possible targets for cancer therapy. In previous studies we introduced two plant derived isomeric flavonoids, flavone A and flavone B which induce apoptosis in highly tumorigenic cancer cells of the breast, colon, pancreas, and the prostate. Flavone A displayed potent cytotoxic activity against more differentiated carcinomas of the colon (CaCo-2) and the pancreas (Panc28), whereas flavone B cytotoxic action is observed on poorly differentiated carcinomas of the colon (HCT 116) and pancreas (MIA PaCa). Apoptosis is induced by flavone A in better differentiated colon cancer CaCo-2 and pancreatic cancer Panc 28 cells via the intrinsic pathway by the inhibition of the activated forms of extracellular signal-regulated kinase (ERK) and pS6, and subsequent loss of phosphorylation of Bcl-2 associated death promoter (BAD) protein, while apoptosis is triggered by flavone B in poorly differentiated colon cancer HCT 116 and MIA PaCa pancreatic cancer cells through the extrinsic pathway with the concomitant upregulation of the phosphorylated forms of ERK and c-JUN at serine 73. These changes in protein levels ultimately lead to activation of apoptosis, without the involvement of AKT

Selection of porcine intestinal isolates as probiotics for pathogen reduction in pigs
Putative lactobacilli were isolated from the porcine small intestine and assessed in vitro for potential probiotic traits. Viability of all isolates tested was unaffected at porcine bile concentrations of 0.3 % (w/v), with some tolerating up to 3.5 % (w/v) bile. Randomly amplified polymorphic DNA (RAPD) and/or pulsed field gel electrophoresis (PFGE) were used to genetically fingerprint the porcine isolates. Some of these cultures demonstrated antagonistic activity against pathogens Escherichia coli O157:H45 and Salmonella typhimurium DT104 when assayed on agar plates and in co-culture. For example, no Salmonella were detected after 8 h of associative culture with one intestinal isolate,L. salivarius B-24. However, when co-cultured under constant pH conditions, no inhibition was observed, indicating that acid may be one of the mechanisms involved in the antimicrobial activity of this strain. Assessment of these strains is on-going with a view to the development of probiotic feed additives which could potentially reduce pathogen carriage in pigs
New hyperekplexia mutations provide insight into glycine receptor assembly, trafficking, and activation mechanisms
Background: Hyperekplexia mutations have provided much information about glycine receptor structure and function. Results: Weidentified and characterized nine new mutations. Dominant mutations resulted in spontaneous activation, whereas recessive mutations precluded surface expression. Conclusion: These data provide insight into glycine receptor activation mechanisms and surface expression determinants. Significance: The results enhance our understanding of hyperekplexia pathology and glycine receptor structure-function. © 2013 by The American Society for Biochemistry and Molecular Biology, Inc. TWEAK and Fn14 expression in the pathogenesis of joint inflammation and bone erosion in rheumatoid arthritis
Extent: 10p.INTRODUCTION: TNF-like weak inducer of apoptosis (TWEAK) has been proposed as a mediator of inflammation and bone erosion in rheumatoid arthritis (RA). This study aimed to investigate TWEAK and TWEAK receptor (Fn14) expression in synovial tissue from patients with active and inactive rheumatoid arthritis (RA), osteoarthritis (OA) and normal controls and assess soluble (s)TWEAK levels in the synovial fluids from patients with active RA and OA. Effects of sTWEAK on osteoclasts and osteoblasts were investigated in vitro. METHODS: TWEAK and Fn14 expression were detected in synovial tissues by immunohistochemistry (IHC). Selected tissues were dual labelled with antibodies specific for TWEAK and lineage-selective cell surface markers CD68, Tryptase G, CD22 and CD38. TWEAK mRNA expression was examined in human peripheral blood mononuclear cells (PBMC) sorted on the basis of their expression of CD22. sTWEAK was detected in synovial fluid from OA and RA patients by ELISA. The effect of sTWEAK on PBMC and RAW 264.7 osteoclastogenesis was examined. The effect of sTWEAK on cell surface receptor activator of NF Kappa B Ligand (RANKL) expression by human osteoblasts was determined by flow cytometry. RESULTS: TWEAK and Fn14 expression were significantly higher in synovial tissue from all patient groups compared to the synovial tissue from control subjects (P < 0.05). TWEAK was significantly higher in active compared with inactive RA tissues (P < 0.05). TWEAK expression co-localised with a subset of CD38+ plasma cells and with CD22+ B-lymphocytes in RA tissues. Abundant TWEAK mRNA expression was detected in normal human CD22+ B cells. Higher levels of sTWEAK were observed in synovial fluids isolated from active RA compared with OA patients. sTWEAK did not stimulate osteoclast formation directly from PBMC, however, sTWEAK induced the surface expression of RANKL by human immature, STRO-1+ osteoblasts. CONCLUSIONS: The expression of TWEAK by CD22+ B cells and CD38+ plasma cells in RA synovium represents a novel potential pathogenic pathway. High levels of sTWEAK in active RA synovial fluid and of TWEAK and Fn14 in active RA tissue, together with the effect of TWEAK to induce osteoblastic RANKL expression, is consistent with TWEAK/Fn14 signalling being important in the pathogenesis of inflammation and bone erosion in RA.Anak A.
