    De acuerdo con la Política Pesquera Común (PPC), todos los stocks pesqueros europeos deberían situarse en un estado que les permita producir en base al Rendimiento Máximo Sostenible (RMS) en el año 2020 como tarde. El proyecto Myfish, financiado por el Séptimo Programa Marco de la UE, tuvo como objetivo construir un marco operacional para la implementación del objetivo del RMS como herramienta para la futura gestión de stocks pesqueros europeos. Este informe presenta el Plan de Implementación Regional para pesquerías demersales de las Islas Baleares (Mediterráneo Occidental) desarrollado en el marco de Myfish en estrecha colaboración con los principales stakeholders locales. El estudio pretende ser de utilidad como primer paso hacia el diseño de planes de gestión pesquera en el área de estudio y un ejemplo práctico de la implementación de la PPC en el Mediterráneo

    Risk of cancer in family members of patients with lynch-like syndrome

    Lynch syndrome (LS) is a common cause of hereditary colorectal cancer (CRC). Some CRC patients develop mismatch repair deficiency without germline pathogenic mutation, known as Lynch-like syndrome (LLS). We compared the risk of CRC in first-degree relatives (FDRs) in LLS and LS patients. LLS was diagnosed when tumors showed immunohistochemical loss of MSH2, MSH6, and PMS2; or loss of MLH1 with BRAF wild type; and/or no MLH1 methylation and absence of pathogenic mutation in these genes. CRC and other LS-related neoplasms were followed in patients diagnosed with LS and LLS and among their FDRs. Standardized incidence ratios (SIRs) were calculated for CRC and other neoplasms associated with LS among FDRs of LS and LLS patients. In total, 205 LS (1205 FDRs) and 131 LLS families (698 FDRs) had complete pedigrees. FDRs of patients with LLS had a high incidence of CRC (SIR, 2.08; 95% confidence interval (CI), 1.56-2.71), which was significantly lower than that in FDRs of patients with LS (SIR, 4.25; 95% CI, 3.67-4.90; p < 0.001). The risk of developing other neoplasms associated with LS also increased among FDR of LLS patients (SIR, 2.04; 95% CI, 1.44-2.80) but was lower than that among FDR of patients with LS (SIR, 5.01, 95% CI, 4.26-5.84; p < 0.001). FDRs with LLS have an increased risk of developing CRC as well as LS-related neoplasms, although this risk is lower than that of families with LS. Thus, their management should take into account this increased risk

    Are ipsilateral breast tumour invasive recurrences in young (⩽40 years) women more aggressive than their primary tumours?

    The characteristics of ipsilateral breast tumour recurrences (IBTRs) relative to those of their primary tumours (PTs) remain scarcely studied. Of 70 young (⩽40 years) premenopausal women with IBTRs, we studied a series of 63 with paired histological data. Median follow-up since IBTR was 10 years. Rates of histological types, grades or hormonal receptors were not significantly different in PTs and in IBTRs. The concordance between IBTRs and their PTs was good for histological types. IBTRs with conserved histological types tended to occur more locally, but not significantly sooner than others. These IBTRs had good concordance for hormone receptors. In discordant cases there were as many losses as appearances of the receptors. The concordance was weak for grades, with equivalent numbers of IBTRs graded lower as higher than their PTs. The 10-year overall survival rate was 70%. Neither the conservation of histological type, location, nor of the two combined were associated with deaths. Early (<2 years) IBTRs, tended to be associated with poorer survival (HR=2.24 (0.92–5.41); P=0.08). IBTRs did not display features of higher aggressiveness than PTs. Neither clinical nor histological definition of a true recurrence could be established other than the conservation of the histological type

    Assessment and management of western Mediterranean small-scale fisheries

    Nearly 75% of European fishing vessels belongs to small-scale fisheries (SSF). However, SSF have received little attention compared to industrial fisheries. In the Mediterranean Sea, where the SSF traditionally had a high socio-economic relevance, there is a widespread interest in securing its sustainable exploitation and viability. Here we analyze the SSF from Mallorca (Balearic Islands) using fishery statistics from the last 25 years (1990–2014). Although fleet size declined markedly (−55%), landings remained constant which might be related to different, not mutually exclusive explanations such as a decrease in unreported catches, stagnant and closed market or technological creeping. Multivariate analysis revealed eight different fishing tactics corresponding to the main target species. Aggregated, these eight species accounted for 52% and 71% of SSF landings weight and value, respectively. The fishery of these species is markedly seasonal and the landings of most of them showed important fluctuations but no clear trends. In contrast with the claims of SSF having lower impact on the natural resources than other fisheries, surplus production models revealed a generalized overexploitation of these target stocks, especially for the most high-valued species. Simulations with a bioeconomic model showed that fishing effort reductions of 38% would improve the health of fish stocks while increasing the economic profits to as much as 15% from current profits. If all stocks were kept below their MSY level, the reduction in fishing effort would have to be of 53% from current values. If the economic yield from the fishery was to be maximized (MEY), fishing effort would need to be reduced by 28%. Owing to important quantities of unreported catches in the most high-valued species, output values of the stock status indicators and the bioeconomic modelling should be taken with care beyond tracking trends in the fisheryVersión del editor1,524

TG-1801, a bispecific antibody that targets CD47 selectively on CD19+ B-cells, is under clinical evaluation in relapsed/refractory (R/R) B-NHL patients either as a single-agent or in combination with ublituximab, a new generation CD20 antibody. A set of eight B-NHL cell lines and primary samples were cultured in vitro in the presence of bone marrow-derived stromal cells, M2-polarized primary macrophages, and primary circulating PBMCs as a source of effector cells. Cell response to TG-1801 alone or combined with the U2 regimen associating ublituximab to the PI3Kδ inhibitor umbralisib, was analyzed by proliferation assay, western blot, transcriptomic analysis (qPCR array and RNA sequencing followed by gene set enrichment analysis) and/or quantification of antibody-dependent cell death (ADCC) and antibody-dependent cell phagocytosis (ADCP). CRISPR-Cas9 gene edition was used to selectively abrogate GPR183 gene expression in B-NHL cells. In vivo, drug efficacy was determined in immunodeficient (NSG mice) or immune-competent (chicken embryo chorioallantoic membrane (CAM)) B-NHL xenograft models. Using a panel of B-NHL co-cultures, we show that TG-1801, by disrupting the CD47-SIRPα axis, potentiates anti-CD20-mediated ADCC and ADCP. This led to a remarkable and durable antitumor effect of the triplet therapy composed by TG-1801 and U2 regimen, in vitro, as well as in mice and CAM xenograft models of B-NHL. Transcriptomic analysis also uncovered the upregulation of the G protein-coupled and inflammatory receptor, GPR183, as a crucial event associated with the efficacy of the triplet combination. Genetic depletion and pharmacological inhibition of GPR183 impaired ADCP initiation, cytoskeleton remodeling and cell migration in 2D and 3D spheroid B-NHL co-cultures, and disrupted macrophage-mediated control of tumor growth in B-NHL CAM xenografts. Altogether, our results support a crucial role for GPR183 in the recognition and elimination of malignant B cells upon concomitant targeting of CD20, CD47 and PI3Kδ, and warrant further clinical evaluation of this triplet regimen in B-NHL

    Improving the performance of a Mediterranean demersal fishery toward economic objectives beyond MSY

    Mediterranean demersal fisheries are highly multispecific and many of their target stocks are overex-ploited. In addition, rocketing fuel costs and low market prices of traditionally high-value species arechallenging the viability of fisheries. Here, based on the numeric results of a simulation model, we con-clude that this situation can be remedied by reducing both fishing mortality and fishing costs. Accordingto our model results, fishing effort reductions of 48–71% would improve the health of fish stocks whileincreasing the economic profits of Mallorca islands bottom trawl fishery to as much as 1.9 MD (146%higher than current profits). If all fish stocks were exploited at their MSY (or below) level, the reductionin fishing effort would have to be of 71% from current values. If equilibrium profits from the fishery wereto be maximized (MEY), fishing effort would need to be reduced by 48%. These results must be takenwith caution due the many sources of uncertainty of our analysis. The modeling tools used to estimatethese values are conditional to the adequate treatment of two sources of uncertainty that are partic-ularly problematic in Mediterranean fisheries: insufficiently known recruitment variability and lack ofperiodic evaluations of the state of many species. Our results show that fishing effort reductions wouldproduce economic yield gains after a period of transition. Further studies on the benefits of changing thesize-selection pattern of fisheries, on better estimation of stock–recruitment relationships and on betterquantifications of the contribution of secondary species to these fisheries, are expected to improve thescientific recommendations for Mediterranean demersal fisheries toward sustainability principles.Versión del editor1,586
